By using perfusions and bolus administration, coupled with postembedding immunocytochemical procedures, we have identified the structures involved in the transport of derivatized orosomucoid (alpha1-acidic glycoprotein) across the continuous microvascular endothelium of the murine myocardium. Our findings indicate that: (i) monomeric orosomucoid binds to the luminal surface of the endothelium; (ii) it is restricted to caveolae during its transport across the endothelium; (iii) it is detected in the perivascular spaces at early time points (by 1 min) and in larger quantities at later time points (>5 min) from the beginning of its perfusion or its intravascular administration; (iv) no orosomucoid molecules are found in the intercellular junctions or at the abluminal exits of interendothelial spaces; and (v) the vesicular transport of orosomucoid is strongly inhibited by N-ethylmaleimide (>80%). Because, by size and shape, the orosomucoid qualifies as a preferential probe for the postulated small pore system, our results are discussed in relation to the pore theory of capillary permeability.  相似文献

Regular eye screening is essential for the early detection and treatment of the diabetic retinopathy. This paper presents a novel automatic screening system for diabetic retinopathy that focuses on the detection of the earliest visible signs of retinopathy, which are microaneurysms. Microaneurysms are small dots on the retina, formed by ballooning out of a weak part of the capillary wall. The detection of the microaneurysms at an early stage is vital, and it is the first step in preventing the diabetic retinopathy. The paper first explores the existing systems and applications related to diabetic retinopathy screening, with a focus on the microaneurysm detection methods. The proposed decision support system consists of an automatic acquisition, screening and classification of diabetic retinopathy colour fundus images, which could assist in the detection and management of the diabetic retinopathy. Several feature extraction methods and the circular Hough transform have been employed in the proposed microaneurysm detection system, alongside the fuzzy histogram equalisation method. The latter method has been applied in the preprocessing stage of the diabetic retinopathy eye fundus images and provided improved results for detecting the microaneurysms.  相似文献

The accuracy of Moderate-resolution Imaging Spectroradiometer (MODIS) level 3 1 km land surface temperature (LST) products was assessed through long-term validation carried out in a mountainous site in Sierra Nevada, southeast Spain. A total of 1458 day and night thermal images, acquired by Terra and Aqua satellites during 2008, were processed and compared to ground-truth data recorded at the meteorological station of Robledal de Cañar with a frequency of one measurement every 10 min. The purpose of this investigation was to understand whether MODIS LST data can be used as input for climate models to be constructed for mountainous environments. Several trends in the MODIS LST data were observed, including the underestimation of daytime values and the overestimation of night-time values. Although all the data sets (Terra and Aqua, diurnal and nocturnal) showed high correlation coefficients with ground measurements, only night values maintained a relatively high accuracy of approximately 2°C of annual average error. Factors that may cause errors in the MODIS LST data, like acquisition angle, cloud, and snow cover, were analysed without conclusive results. High accuracy levels, i.e. close to 1°C, similar to other validation studies carried out over simpler and much more homogenous land-cover types such as cultivated fields, have been achieved for night images acquired during the summer months, thus making these datasets reliable for their use in climatic models over mountainous regions.  相似文献

Fuzzy-based Refinement of the Fault Diagnosis Task in Industrial Devices   总被引:2,自引:0,他引:2  
This paper first describes a fuzzy classifier to be used for fault diagnosis. Then, the paper presents a refinement of the diagnosis task performed with this fuzzy classifier. For each fault, a number of 20 levels of fault strength have been considered. In previous work, more than one single category per fault has been used to improve the classifier performance, i.e. distributing the strength levels into small, medium and, respectively large strength subsets. However, this distribution scheme is too rigid. This paper introduces a flexible distribution scheme that takes into account the (di)similarities between different strength levels. The refinement proposed here offers better insight on the behavior of each fault and it increases separation between overlapping faults, which improves the final outcome of the diagnosis process.  相似文献

Perforated patch recording with nystatin, amphotericin B and gramicidin can be more difficult in the hands of some investigators than others. In addition, it is difficult to introduce low molecular weight substances such as dyes into the cytoplasm in such experiments. Radio frequency energy is utilised for heating in a wide range of applications, particularly in the food industry. A major challenge of RF processing is non-uniform heating in loads of variable and angular geometry, leading to reduced quality and product damage. In this study, the specific effects of geometry on the heating profiles of a range of geometrically variable loads in an industrial scale RF system are analysed, and the understanding used to derive a general tool to predict heating uniformity. Potato was selected as a test material for experimental work; dielectric properties were measured using a 44-mm coaxial probe. Analysis of simulated and experimental surface temperature profiles and simulated power uniformity indices indicates the presence of vertices and edges on angular particles, and their proximity to faces perpendicular to the RF electrodes increases localised heating; faces parallel to the electrodes heated less than those faces perpendicular to them. Comparison of the same geometrical shape in different orientations indicates that overall power absorption uniformity can be better even when localised heating of edges is greater. It is suggested, for the first time, that the rotation of angular shapes within a parallel plate electric field can improve heating uniformity, and that this can be achieved through the design of bespoke electrode systems. A Euler characteristic-based shape factor is proposed, again for the first time, that can predict heating uniformity for solid, dielectrically homogeneous shapes. This gives industry a tool to quickly determine the feasibility for uniform RF heating of different three-dimensional shapes based on geometry alone. This provides a screening method for food technologists developing new products, allowing rapid assessment of potential heating uniformity, and reducing the need for early-stage specialist computational modelling.  相似文献

The ratio of free to thylakoid-bound chloroplast ribosomes in Chlamydomonas reinhardtii undergoes periodic changes during the synchronous light-dark cycle. In the light, when there is an increase in the chlorophyll content and synthesis of thylakoid membrane proteins, about 20-30% of the chloroplast ribosomes are bound to the thylakoid membranes. On the other hand, only a few or no bound ribosomes are present in the dark when there is no increase in the chlorophyll content. The ribosome-membrane interaction depends not only on the developmental stage of the cell but also on light. Thus, bound ribosomes were converted to the free variety after cultures at 4 h in the light had been transferred to the dark for 10 min. Conversely, a larger number of chloroplast ribosomes became attached to the membranes after cultures at 4 h in the dark had been illuminated for 10 min. Under normal conditions, when there was slow cooling of the cultures during cell harvesting, chloroplast polysomal runoff occurred in vivo leading to low levels of thylakoid-bound ribosomes. This polysomal runoff could be arrested by either rapid cooling of the cells or the addition of chloramphenicol or erythromycin. Each of these treatments prevented polypeptide chain elongation on chloroplast ribosomes and thus allowed the polyosomes to remain bound to the thylakoids. Addition of lincomycin, an inhibitor of chain initiation on 70S ribosomes, inhibited the assembly of polysome-thylakoid membrane complex in the light. These results support a model in which initiation of mRNA translation begins in the chloroplast stroma, and the polysome subsequently becomes attached to the thylakoid membrane. Upon natural chain termination, the chloroplast ribosomes are released from the membrane into the stroma.  相似文献

In this paper, we discuss a quantum approach for the all-pair multiclass classification problem. In an all-pair approach, there is one binary classification problem for each pair of classes, and so there are k(k???1)/2 classifiers for a k-class classification problem. As compared to the classical multiclass support vector machine that can be implemented with polynomial run time complexity, our approach exhibits exponential speedup due to quantum computing. The quantum all-pair algorithm can also be used with other classification algorithms, and a speedup gain can be achieved as compared to their classical counterparts.  相似文献
