Neurodegenerative disorders are primarily characterized by neuron loss. The most common neurodegenerative disorders include Alzheimer’s and Parkinson’s disease. Although there are several medicines currently approved for managing neurodegenerative disorders, a large majority of them only help with associated symptoms. This lack of pathogenesis-targeting therapies is primarily due to the restrictive effects of the blood–brain barrier (BBB), which keeps close to 99% of all “foreign substances” out of the brain. Since their discovery, nanoparticles have been successfully used for targeted delivery into many organs, including the brain. This review briefly describes the pathophysiology of Alzheimer’s, Parkinson’s disease, and amyotrophic lateral sclerosis, and their current management approaches. We then highlight the major challenges of brain-drug delivery, followed by the role of nanotherapeutics for the diagnosis and treatment of various neurological disorders.  相似文献

An aptamer is a short sequence of synthetic oligonucleotides which bind to their cognate target, specifically while maintaining similar or higher sensitivity compared to an antibody. The in-vitro selection of an aptamer, applying a conjoining approach of chemistry and molecular biology, is referred as Systematic Evolution of Ligands by Exponential enrichment (SELEX). These initial products of SELEX are further modified chemically in an attempt to make them stable in biofluid, avoiding nuclease digestion and renal clearance. While the modification is incorporated, enough care should be taken to maintain its sensitivity and specificity. These modifications and several improvisations have widened the window frame of aptamer applications that are currently not only restricted to in-vitro systems, but have also been used in molecular imaging for disease pathology and treatment. In the food industry, it has been used as sensor for detection of different diseases and fungal infections. In this review, we have discussed a brief history of its journey, along with applications where its role as a therapeutic plus diagnostic (theranostic) tool has been demonstrated. We have also highlighted the potential aptamer-mediated strategies for molecular targeting of COVID-19. Finally, the review focused on its future prospective in immunotherapy, as well as in identification of novel biomarkers in stem cells and also in single cell proteomics (scProteomics) to study intra or inter-tumor heterogeneity at the protein level. Small size, chemical synthesis, low batch variation, cost effectiveness, long shelf life and low immunogenicity provide advantages to the aptamer over the antibody. These physical and chemical properties of aptamers render them as a strong biomedical tool for theranostic purposes over the existing ones. The significance of aptamers in human health was the key finding of this review.  相似文献

Design and development of a cloud monitoring camera (CMC) as a secondary camera for imaging/remote-sensing satellites is proposed here. It is well known that presence of clouds alters the earth-atmospheree energy budget by scattering and absorbing the shortwave radiation and absorbing and re-emitting the infrared radiation. Detection of clouds at precise positions, prior to the initiation of the imaging session of the primary (super resolution) camera can aid in saving power as well as memory storage of the satellite. Key feature of the proposed system is the implementation of an onboard cloud detection system in the visible band. The proposed system is miniature in size and cost-effective when compared to the existing integrated visible and infrared band detection system. It primarily consists of lens assembly, detector head assembly, field programmable gate array (FPGA)-based camera electronics system with supporting proximity electronics. Cloud detection algorithm for the visible band images is derived and simulated with the help of Laboratory Virtual Instrument Engineering Workbench software. The verification and hardware realization of the algorithm is achieved by implementing it on a FPGA-based system. Overall simulated and measured efficiency of more than 94% was achieved. The volume of the system is 85 mm × 85 mm × 25 mm and weighs approximately 2 kg, making it suitable for satellite mounting.  相似文献

Transmission of power from 42 ± 0.2 GHz gyrotron (TE03 mode) to tokamak or dummy load requires a set of transmission line components. It includes a set of mode converters that converts circularly unpolarised TE03 mode to polarised HE11 mode. The mode conversion sequence is methodised in two steps; first from TE03 to TE01 mode and then from TE01 to TM11 mode. The proposed mode converters performance depends on the parameters such as waveguide radius, beat wavelength, number of ripples, its perturbation amplitude (η) and bending angle. These parameters are numerically optimised and verified in CST microwave studio. TE03 to TE01 mode conversion is realised by the generation of intermediate mode TE02. The mode conversion length for TE03 to TE02 mode converter is6λ0, (where λb is the beat wavelength of corresponding mode conversion) which converts 99.15% of an incident circular TE03 mode power into TE02 mode power. Next, mode converter converts TE02 mode into TE01 mode with 99.06% efficiency along the optimal length of6λb. For TE01 to TM11 mode conversion, two conversion methodologies have been proposed: first using axis arc bend (34.94°) technique to convert TE01 into its analogous degenerate mode with 99.01% conversion efficiency and second using parabolic curve with 90° bend, which gives a conversion efficiency of 97.5%.  相似文献
