In the present work, we propose a robust calibration of some bi‐parametric multiaxial fatigue criteria applied in conjunction with the theory of critical distances (TCD). This is based on least‐square fitting fatigue data generated using plain and sharp‐notched specimens tested at two different load ratios and allows for the estimation of the critical distance according to the point and line method formulation of TCD. It is shown that this combination permits to incorporate the mean stress effect into the fatigue strength calculation, which is not accounted for in the classical formulation of TCD based on the range of the maximum principal stress. It is also shown that for those materials exhibiting a low fatigue‐strength‐to‐yield‐stress ratio σfl,R = ?1/σYS, such as 7075‐T6 (σfl,R = ?1/σYS = 0.30), satisfactorily accurate predictions are obtained assuming a linear‐elastic stress distribution, even at the tip of sharp notches and cracks. Conversely, for any materials characterized by higher values of this ratio, as quenched and tempered 42CrMo4 (σfl,R = ?1/σYS = 0.54), it is recommended to consider the stabilized elastic‐plastic stress/strain distribution, also for plain and blunt‐notched samples and even in the high cycle fatigue regime still with the application of the TCD. 相似文献

The study evaluates the tumor distribution of the rapid clearance blood pool agent (RCBPA) gadomelitol, in a breast tumor model. Different techniques were used : (1) tissue gadolinium concentrations measured by inductively coupled plasma atomic emission spectroscopy (ICP-AES), (2) whole body quantitative autoradiography using radiolabeled [153Gd] gadomelitol and (3) dynamic contrast-enhanced MRI with compartmental analysis. An accumulation of gadomelitol in tumors compared to muscle was observed 30 min and 3 h post injection (p.i.). Thirty minutes p.i., the gadomelitol tumor distribution evaluated by autoradiography showed a marked difference between the rim and the center, whereas both areas showed comparable concentrations after 3 h. Using dynamic contrast-enhanced MRI, three phases could be observed during the 1 hour observation period: (1) rapid tumor uptake within the first few minutes post-injection (2) a progressive increase in tumor signal enhancement over 10 min and (3) a steady-state phase. Average +/− SD (n=5) transendothelial permeability KPS and the fractional blood volume fBV were 12.2±1.6 μl/min−1/g and 5.4±0.2% respectively. Due to its slow extravasation and high tumor residence time, gadomelitol may potentially be useful to improve characterization between benign versus malignant tumors using dynamic MRI. 相似文献

To investigate the molecular mechanism of gastric carcinogenesis, we analyzed genetic instability and p53 gene mutations in 40 primary gastric carcinomas. Tumor samples were from untreated patients with no family history suggestive of genetic predisposition to cancer. We screened six microsatellite loci by the polymerase chain reaction (PCR) method, and exons 5-8 of the p53 gene by the PCR-based denaturing gradient gel electrophoresis and sequencing techniques. Microsatellite instability was detected in 32.5% (13/40), and gene mutations in 40% (16/40), of the tumors analyzed. No statistically significant associations were found between genetic alterations and clinico-pathological variables (with the exception of diffusion of lymph node metastases, which was inversely associated with the presence of microsatellite alterations; P < 0.01). Interestingly, a negative association was found between genetic instability and p53 gene mutations: 11 out of 13 tumors showing instability proved to carry a nonmutated p53 gene versus 2/13 carrying a mutated gene (P = 0.03). These observations suggest that genetic instability and p53 gene mutations play a crucial role in the gastric carcinogenic process, but likely act through distinct pathways during cancer development. However, genetic instability is not in and of itself neoplastic. Therefore, we investigated whether insertion/deletion mutations of the polyadenine tract within the transforming growth factor-beta type II receptor gene (TGF-beta RII) were frequently present in gastric tumors with an RER+ (replication error) phenotype. We found RII mutations in 8/40 (20%) samples: mutations were present in 7/13 (54%) RER+ tumors versus 1/27 (4%) RER- cases (P < 0.001). 相似文献

Abstract: The paper describes a test rig designed to check and assess the accuracy of the incremental hole drilling (IHD) method. An external load produces a controlled linearly through thickness variable uniaxial stress field (reference bending stress), known with good accuracy, that can be applied and removed at each hole increment. After the separation between the bending relaxed strain from the residual stress relaxed strain, it is possible to reproduce the bending stress distribution in order to have complete confidence of the residual stress measurement. The bending verification of the IHD method was already proposed by other investigators before, but residual stress measurements were then performed on independent configurations. The proposed testing procedure gives a ‘real time’ verification of the residual stress measurement. Any experimental malfunctioning due to the operator inexperience, or any error during the stress calculation from the relaxed strain would produce an evident difference between the expected reference bending stress and the IHD bending stress output. Moreover, the reference bending stress helped for understanding that the not perfect hole cylindrical shape causes an underestimate of the predicted stress near the surface. A zero depth offset correction was proposed. This correction was tested on the reference bending stress, and then applied to the residual stress prediction. Three shot peening residual stresses IHD measurements were successfully validated by means of the bending stress; moreover, they were in good agreement with independent X‐ray diffraction measures also proposed in the paper. 相似文献

The fatigue strength at a high number of cycles with initial elastic–plastic behavior was experimentally investigated on quenched and tempered 42CrMo4 steel. Fatigue tests on unnotched specimens were performed both under load and strain controls, by imposing various levels of amplitude and with several high load ratios. Different ratcheting and relaxation trends, with significant effects on fatigue, are observed and discussed, and then reported in the Haigh diagram, highlighting a clear correlation with the Smith–Watson–Topper model. High load ratio tests were also conducted on notched specimens with C (blunt) and V (sharp) geometries. A Chaboche model with three parameter couples was proposed by fitting plain specimen cyclic and relaxation tests, and then finite element analyses were performed to simulate the notched specimen test results. A significant stress relaxation at the notch root became clearly evident by reporting the numerical results in the Haigh diagram, thus explaining the low mean stress sensitivity of the notched specimens. 相似文献

Advanced high-strength steels (AHSS) have a better combination between strength and ductility than conventional HSS, and higher crash resistances are obtained in concomitance with weight reduction of car structural components. These steels have been developed in the last few decades, and their use is rapidly increasing. Notwithstanding, some of their important features have to be still understood and studied in order to completely characterize their service behavior. In particular, the high mechanical resistance of AHSS makes hydrogen-related problems a great concern for this steel grade. This article investigates the hydrogen embrittlement (HE) of four AHSS steels. The behavior of one transformation induced plasticity (TRIP), two martensitic with different strength levels, and one hot-stamping steels has been studied using slow strain rate tensile (SSRT) tests on electrochemically hydrogenated notched samples. The embrittlement susceptibility of these AHSS steels has been correlated mainly to their strength level and to their microstructural features. Finally, the hydrogen critical concentrations for HE, established by SSRT tests, have been compared to hydrogen contents absorbed during the painting process of a body in white (BIW) structure, experimentally determined during a real cycle in an industrial plant. 相似文献

Cheese production and consumption are increasing in many countries worldwide. As a result, interest has increased in strategies for genetic selection of individuals for technological traits of milk related to cheese yield (CY) in dairy cattle breeding. However, little is known about the genetic background of a cow's ability to produce cheese. Recently, a relatively large panel (1,264 cows) of different measures of individual cow CY and milk nutrient and energy recoveries in the cheese (REC) became available. Genetic analyses showed considerable variation for CY and for aptitude to retain high proportions of fat, protein, and water in the coagulum. For the dairy industry, these characteristics are of major economic importance. Nevertheless, use of this knowledge in dairy breeding is hampered by high costs, intense labor requirement, and lack of appropriate technology. However, in the era of genomics, new possibilities are available for animal breeding and genetic improvement. For example, identification of genomic regions involved in cow CY might provide potential for marker-assisted selection. The objective of this study was to perform genome-wide association studies on different CY and REC measures. Milk and DNA samples from 1,152 Italian Brown Swiss cows were used. Three CY traits expressing the weight (wt) of fresh curd (%CYCURD), curd solids (%CYSOLIDS), and curd moisture (%CYWATER) as a percentage of weight of milk processed, and 4 REC (RECFAT, RECPROTEIN, RECSOLIDS, and RECENERGY, calculated as the % ratio between the nutrient in curd and the corresponding nutrient in processed milk) were analyzed. Animals were genotyped with the Illumina BovineSNP50 Bead Chip v.2. Single marker regressions were fitted using the GenABEL R package (genome-wide association using mixed model and regression–genomic control). In total, 103 significant associations (88 single nucleotide polymorphisms) were identified in 10 chromosomes (2, 6, 9, 11, 12, 14, 18, 19, 27, 28). For RECFAT and RECPROTEIN, high significance peaks were identified in Bos taurus autosome (BTA) 6 and BTA11, respectively. Marker ARS-BFGL-NGS-104610 (~104.3 Mbp) was highly associated with RECPROTEIN and Hapmap52348-rs29024684 (~87.4 Mbp), closely located to the casein genes on BTA6, with RECFAT. Genomic regions identified may enhance marker-assisted selection in bovine cheese breeding beyond the use of protein (casein) and fat contents, whereas new knowledge will help to unravel the genomic background of a cow's ability for cheese production. 相似文献

Two techniques for assembling an aluminium drill pipe to a steel tool joint were investigated: ‘hot’ assembling and ‘cold’ assembling. During hot assembling a difference of temperature between the two parts is exploited to impose contact interferences and then to obtain a friction bond that generates torsional strength. Hot assembling is ordinarily used. The cold assembling technique is introduced here as an alternative. During cold assembling connection make up, the contact interferences are obtained by imposing an external torque. This technique requires a proper lubricating compound between the mating surfaces, in order to reduce the torque required to make up the connection. After lubricating compound curing, the coefficient of static friction rises and a high torsional strength of the connection is obtained. Full scale experimental tests were performed both on hot assembled connections and on cold assembled connections. Cold assembling showed higher torsional strength than hot assembling; moreover cold assembling does not expose the aluminium alloy to high temperature, that can be a risk for the aluminium alloy microstructure. However, the use of the compound can be a reason of cold assembling torsional strength scatter. 相似文献
