The objective of this study was to assess workers' exposure to bioaerosols in sludge drying units. Simultaneous collection of inhalable dust and airborne biological agents was carried out using rotating cup samplers (CIP 10, Arelco). Sixteen stationary and 30 personal measurements were performed in three drying plants, during summer and winter. Microbial content of air was characterized by high amounts of Gram-negative bacteria, endotoxin, and thermophilic actinomycetes. High differences between exposure levels were observed with regard to workplaces and tasks. Dryers and conveyors in operation were closed, and the monitoring task in the dryer room was associated with low levels of personal exposure, close to background levels. Conversely, cleaning and maintenance of the dryer and conveyors required the equipment to be opened, giving significantly higher task-based personal exposure levels. The fall of dried sludge into indoor containers resulted in high emission and spreading of dust. Mean personal exposure levels to fungi were moderate or low for all tasks. Based on the results of this study, appropriate means of operational control and personal protection are being developed and implemented on sites.  相似文献

In July 1996, the US Department of Agriculture (USDA), Food Safety and Inspection Service (FSIS), published the Pathogen Reduction; Hazard Analysis and Critical Control Point (HACCP) Systems final rule to improve food safety of meat and poultry products. The final rule established, among other requirements, pathogen reduction performance standards for Salmonella for food animal carcasses and raw ground products. The final rule is to be fully implemented in three stages in successively smaller federally inspected meat and poultry slaughter and processing establishments. Implementation began in January 1998 and was completed in January 2000. Samples of carcasses of four species of food animals (cattle, swine, chickens, turkeys), and raw ground product from each of these species, were collected by FSIS from establishments prior to implementation of the final rule and cultured for Salmonella. This paper reports Salmonella serotype results of samples collected from June 1997 through August 1998. These results represent a baseline for future comparisons.  相似文献

Plasma facing components (PFCs) in magnetic confinement controlled fusion machines are armoured with carbon fibre composite (CFC) bonded to a copper alloy heat sink. The manufacturing process induces high level of residual stresses due to the thermal expansion mismatch between CFC and copper and PFCs have to withstand strong stress ranges during operation. To study the initiation and propagation of damage in the CFC part, the ONERA damage model is used to describe the behaviour of the N11 material. The finite element simulations show that the damage is located near the interface and develops during the manufacturing of the PFCs as a consequence of the high amplitude of shear stresses. Under high heat flux, stresses decrease and the damage does not evolve. Further studies will take into account the damageable behaviour of the composite/copper interface, which will lead to geometrical optimisations and better knowledge of the link between damage and conductivity.  相似文献

Physical processes such as advection, dispersion, and air-water gas exchange play important roles in determining the movement and change in concentration of contaminants discharged into rivers. In the following, we report results from a large-scale SF6 tracer release experiment conducted in the tidal Hudson Riverto examine longitudinal dispersion and net advection. SF6 was injected into the Hudson River near Newburgh, NY, and surveyed for 13 days using a new, fully automated, high-resolution SF6 sampling and analysis system. Net down river advection of the water body originally tagged with SF6 was slow, averaging mean displacement rates of about 0.5 +/- 0.2 km d(-1). In contrast, spreading of the tracer was driven by tidal movement, causing rapid mixing of the water up and down river. By examining the change in the second moment of the tracer distribution with time, we determined the mean longitudinal dispersion coefficient to be 70.1 +/- 4.3 m2 s(-1). Temporal evolution of the SF6 inventory indicates an average gas transfer velocity over the period of the experiment of 6.5 +/- 0.5 cm h(-1) (1.56 +/- 0.12 m d(-1)). Vertical profiles show that mixing into the bottom layers of the river, in places reaching more than 53 m, seemed to be rapid.  相似文献

Loss of phosphoric acid is the most effective fragmentation reaction of pSer- and pThr-containing phosphopeptides of small size (up to 10-15 residues) in low-energy collision-induced dissociation. Therefore, tandem mass spectrometry with neutral loss scanning was evaluated for its utility to analyze protein phosphorylation using protein kinase A (PKA) catalytic subunit, which is phosphorylated at Thr197 and Ser338, as an example. Analysis of tryptic digests of phosphoproteins by tandem mass spectrometry with scanning for neutral loss of phosphoric acid resulted in spectra with poor signal-to-noise ratio, mainly because of the large size of the phosphopeptides formed (>2 kDa). This unfavorable size was caused by the distribution of tryptic cleavage sites in PKA and by interference of phosphorylation with tryptic cleavage. To generate a set of smaller peptide fragments, digestion was performed using the low-specificity protease elastase. Analysis of the total elastase digest with neutral loss scanning resulted in observation of a set of partially overlapping phosphopeptides with high abundance, providing a complete coverage of PKA phosphorylation sites. The peptide size generated by elastase (0.5-1.5 kDa) is ideally suited for this scan mode, which was found to provide the highest specificity for detection of singly charged phosphopeptides (neutral loss of 98). Identification of the PKA phosphorylation sites was performed by mass spectrometric sequencing of the elastase-derived phosphopeptides, which provided highly informative product ion spectra.  相似文献
