Aspect-oriented programming (AOP), now practically a consolidated academic discipline, has yet to build more solid industrial foundations, especially in the realms of the .NET platform. It's believed that this situation is caused by the lack of a robust and user-friendly AOP tool for .NET comparable with the Java-based AspectJ. This work investigates the basic infrastructure required for building such a tool: aspect-oriented weaving with the common language runtime (CLR) environment. In this regard, a classification schema is built, analysing the attributes a hypothetical aspect weaver for .NET might have. It assesses the different classes of weavers that can be built on top of the CLR today and investigates what extensions to the platform would be needed in order to enable more sophisticated weaving technologies. Some typical use cases for the resulting AOP tools, and classify what attributes a corresponding weaver would need to have in order to fulfil these requirements. Finally, two existing aspect weaver implementations in terms of these very same attributes are analysed.  相似文献

Obtaining quantitative information about molecular assemblies with high spatial and temporal resolution is a challenging task in fluorescence microscopy. Single‐molecule techniques build on the ability to count molecules one by one. Here, a method is presented that extends recent approaches to analyze the statistics of coincidently emitted photons to enable reliable counting of molecules in the range of 1–20. This method does not require photochemistry such as blinking or bleaching. DNA origami structures are labeled with up to 36 dye molecules as a new evaluation tool to characterize this counting by a photon statistics approach. Labeled DNA origami has a well‐defined labeling stoichiometry and ensures equal brightness for all dyes incorporated. Bias and precision of the estimating algorithm are determined, along with the minimal acquisition time required for robust estimation. Complexes containing up to 18 molecules can be investigated non‐invasively within 150 ms. The method might become a quantifying add‐on for confocal microscopes and could be especially powerful in combination with STED/RESOLFT‐type microscopy.  相似文献

The cAMP-dependent aquaporin-2 (AQP2) redistribution from intracellular vesicles into the plasma membrane of renal collecting duct principal cells induces water reabsorption and fine-tunes body water homeostasis. However, the mechanisms controlling the localization of AQP2 are not understood in detail. Using immortalized mouse medullary collecting duct (MCD4) and primary rat inner medullary collecting duct (IMCD) cells as model systems, we here discovered a key regulatory role of Aurora kinase A (AURKA) in the control of AQP2. The AURKA-selective inhibitor Aurora-A inhibitor I and novel derivatives as well as a structurally different inhibitor, Alisertib, prevented the cAMP-induced redistribution of AQP2. Aurora-A inhibitor I led to a depolymerization of actin stress fibers, which serve as tracks for the translocation of AQP2-bearing vesicles to the plasma membrane. The phosphorylation of cofilin-1 (CFL1) inactivates the actin-depolymerizing function of CFL1. Aurora-A inhibitor I decreased the CFL1 phosphorylation, accounting for the removal of the actin stress fibers and the inhibition of the redistribution of AQP2. Surprisingly, Alisertib caused an increase in actin stress fibers and did not affect CFL1 phosphorylation, indicating that AURKA exerts its control over AQP2 through different mechanisms. An involvement of AURKA and CFL1 in the control of the localization of AQP2 was hitherto unknown.  相似文献

Understanding perception of dairy cows to common human contact such as stroking is important for improving the human-animal relationship, animal welfare, and production. We hypothesized that repeated stroking of body regions licked most during social grooming, especially the ventral neck, would reduce cows’ avoidance of and increase their approach to humans. Sixty tethered dairy cows were randomly allocated to 4 treatment groups that received 5 min of daily human contact 5 d/wk during 3 consecutive weeks: 3 groups were stroked on different body regions. The first group was stroked on the ventral part of the neck (neck); the second group on the withers (both licked often in social grooming); the third group on the lateral side of the chest (chest, licked rarely); and the last group (control) was exposed to simple human presence. The reactions to the person who had provided the treatment were measured using 2 tests in the home tie-stall assessing avoidance from an approaching person who tried to touch the head (approaching person test) and avoidance/approach reactions to a stationary person (stationary person test). Approach behavior was recorded in a novel environment using a standard arena test. In the home tie-stall, cows stroked on the neck showed less avoidance (median avoidance score: 3.33) in the approaching person test compared with cows stroked on the chest and the controls (both: 4.00). That is, at least 75% of the animals stroked on the neck tolerated the touching of their heads (75th percentile ≤ 3.75), whereas at least 50% of the cows in the other treatment groups did not accept it. The stationary person test did not reveal any differences between the treatment groups. In the arena test, the 3 stroked groups showed more approach behavior (median latencies to contact: from 145 to 240 s) compared with simple human presence (300 s), but stroking treatments did not differ from each other. Stroking, particularly the neck, reduced avoidance of and increased approach reactions to humans in both the home tie-stall and the arena. Increasing acceptance of being touched after being stroked on the neck suggests that this procedure should be adopted to improve routine handling of dairy cattle.  相似文献

The targeted analysis of customer data becomes increasingly important for data-driven business models. At the same time, the customers’ concerns regarding data privacy have to be addressed properly. Existing research mostly describes data privacy as a necessary evil for compliance and risk management and does not propose specific data privacy measures which address the customers’ concerns. We therefore aim to shed light on the upside of data privacy. In this paper, we derive specific measures to deal with customers’ data privacy concerns based on academic literature, legislative texts, corporate privacy statements, and expert interviews. Next, we leverage the Kano model and data from two internet-based surveys to analyze the measures’ evaluation by customers. From a customer perspective, the implementation of the majority of measures is obligatory as those measures are considered as basic needs of must-be quality. However, delighting measures of attractive quality do exist and have the potential to create a competitive advantage. In this, we find some variation across different industries suggesting that corporations aiming to improve customer satisfaction by superior privacy protection should elicit the demands of their specific target customers.  相似文献

Automated analysis of submicron particles by computer-controlled scanning electron microscopy is generally possible. The minimum diameter of the detectable particles is dependent on the mean atomic number of the particles and the operating parameters of the scanning microscope. The main limitation with regard to particle size is set by the quality of the particle detection system, which generally is the backscatter electron detector. The accuracy of the results of the x-ray analyses is very often strongly affected by specimen damage, omnipresent especially for environmental particles even at low electron energies and probe currents. With the exception for light elements, the detection limit is approximately 1 wt%. Device-related limitations to automated analysis may be specimen drift and an unreliable autofocus function.  相似文献

ILA cells were established from tumors induced by the pancreatic carcinogen N-nitrosobis(2-oxopropyl)amine (BOP) in hamster islets. The proliferation, morphology, karyotype, immunoreactivity with certain antibodies and growth factor secretion of these tumor cells were compared with the same parameters in tumor cells induced by BOP in hamster ductal cells (TAKA-1-BOP) established in a previous study. Minor differences were found in the morphology and ultrastructure of the 2 cell lines. Contrary to TAKA-1-BOP cells, ILA cells did not express cytokeratins 8.13, 13 or 18 but did express DU-PAN-2 and TAG-72, 2 known human pancreatic cancer-associated antigens. No endocrine cell markers were expressed. A significant difference also was found in the chromosomal pattern in that there were more abnormalities and marker chromosomes in ILA cells than in TAKA-1-BOP cells and the Y or X chromosomes were missing in ILA cells. ILA cells produced TGF-alpha, IGF-I, bombesin and gastrin and expressed specific binding sites for hEGF. TGF-alpha secretion from ILA cells was much greater than that from TAKA-1-BOP cells. Our results indicate that pancreatic cancer cells grown in vitro are not a single clone. We conclude that there are some genetic and biological differences between tumors arising from pancreatic duct and islets and that pancreatic ductal adenocarcinomas originating from islets have a profound malignant potential.  相似文献
