The effect of divalent cations on the inactivation of Escherichia coli by high hydrostatic pressure was investigated. The presence of 0.5 mmol l-1 of CaCl2, MgCl2, MnCl2 and FeCl2 reduced pressure inactivation of E. coli MG1655, while 0.5 mmol l-1 of ZnCl2, NiCl2, CuCl2 and CoCl2 increased inactivation. Baroprotection by Ca2+ was found to be dose-dependent up to at least 80 mmol l-1 and was studied in more detail in terms of inactivation kinetics. Logarithmic survivor plots against time deviated from first order kinetics, suggesting that MG1655 cultures were heterogeneous with regard to pressure resistance. All cultures were shown to contain a small proportion of cells that were only slowly inactivated. Addition of Ca2+ increased the proportion of these tolerant cells in the cultures up to 1000-fold at 80 mmol l-1, but did not affect their inactivation rate. The addition of EDTA resulted in the opposite effect, lowering the proportion of pressure-tolerant cells in the cultures. Three pressure-resistant mutants of E. coli MG1655 were found to be more resistant to EDTA under pressure compared with MG1655, and were unaffected by Ca2+ under pressure. In addition, these mutants had a 30-40% lower Ca2+ content than MG1655. Based on these results, it is postulated that pressure killing of E. coli MG1655 is mediated primarily by the destabilization of Ca(2+)-binding components, and that the mutations underlying pressure resistance have resulted in pressure-stable targets with reduced Ca(2+)-binding affinity.  相似文献

Certain non-crystalline germanium films (> 10 μm in thickness) prepared by rf-sputtering crystallize “explosively” at room temperature when initiated by pricking the surface with a sharp point (or certain other methods). The propagation velocity of the crystallization at room temperature was found to be as fast a 1200 mm sec?1 depending somewhat on the conditions of film preparation, thickness, etc. The density of several such crystallizable films was determined as 5.05 g cm?3±1%. The crystallite size in the crystallized films, measured by X-ray broadening, was typically larger than 500 Å. A model for the crystallization process invokes a cascade energy transfer process, basically thermal in nature.  相似文献

To improve methanol-oxidation performances of membrane-electrode assemblies composed of a hydrocarbon-based ionomers, the resistances involved in the reaction were decreased. Electrochemical impedance spectroscopy revealed that the proton-conductive resistance (Ri) in the anode was decreased from 0.54 to 0.40 Ω cm2 by increasing a loading ratio of platinum-ruthenium to carbon support of anode catalyst from 54 to 73 wt.%. In addition, Ri was decreased to be 0.25 Ω cm2 by increasing ion-exchange capacity (IEC) of the ionomer from 1.4 to 2.9 mequiv. g−1. Consequently, the polarization resistance of the anode was significantly decreased, in turn, increasing current density of methanol oxidation at the potential of 0.45 V from 0.110 to 0.244 A cm−2.  相似文献

Distal myopathy with rimmed vacuoles is an autosomal recessive muscular disorder, characterized clinically by weakness of the distal muscles in the lower limbs in early adulthood. Recently, the gene locus for familial vacuolar myopathy with autosomal recessive inheritance (hereditary inclusion body myopathy) was mapped to chromosome 9 by genome-wide linkage analysis of nine Persian-Jewish families. Since both disease conditions share similar clinical, genetic, and histopathological features, we analyzed seven families with distal myopathy with rimmed vacuoles using ten microsatellite markers within the region of the hereditary inclusion body myopathy locus. Significantly high cumulative pairwise lod scores were obtained with three markers: D9S248 (Z(max) = 5.90 at theta = 0), D9S43 (Z(max) = 5.25 at theta = 0), and D9S50 (Z(max) = 4.23 at theta = 0). Detection of obligate recombination events as well as multipoint linkage analysis revealed that the most likely location of the distal myopathy with rimmed vacuoles gene is in a 23.3-cM interval defined by D9S319 and D9S276 on chromosome 9. The results raise the possibility that distal myopathy with rimmed vacuoles and hereditary inclusion body myopathy in Persian Jews are allelic diseases.  相似文献

BACKGROUND: To assess the role of contrast enemas for the evaluation of leaks in symptomatic and asymptomatic patients after the first stage of restorative proctocolectomy. METHODS: We reviewed the findings of 59 contrast enemas in 40 patients who underwent total proctocolectomy with creation of an ileoanal pouch and a proximal diverting ileostomy. Thirty-seven patients initially underwent routine contrast studies of the ileoanal pouch, and three underwent contrast studies because of suspected fistulas or obstruction. Medical records were also reviewed to determine the clinical presentation and course of these patients. RESULTS: Of 37 patients who underwent routine postoperative contrast enemas, 33 (89%) had normal studies, three (8%) had clinically silent leaks (including two blind-ending tracks from the ileoanal anastomosis and one from the pouch), and one (3%) had pouchitis. In all three patients with unsuspected leaks, ileostomy closure was delayed. In two other patients with abdominal pain and fever, contrast enemas revealed leaks from the ileoanal pouch and distal ileum, respectively. The remaining patient had a contrast enema because of abdominal pain and distention; this patient had a distal ileal obstruction due to adhesions. CONCLUSIONS: Routine postoperative contrast studies revealed clinically silent leaks from the ileal J pouch or ileoanal anastomosis in three of 37 patients (8%) after the first stage of restorative proctocolectomy. Our findings suggest that routine contrast enema can detect clinically silent leaks after this surgery.  相似文献

Graft copolymerization of methyl methacrylate onto novoloid fiber was carried out by the electron beam mutual irradiation method. The characteristics of the grafted fiber were almost the same as those of the original novoloid fiber. The grafted fiber was carbonized and activated to yield an efficiently activated carbon fiber. The yield and the specific surface area of the activated carbon fiber, derived from the grafted novoloid fiber, were much greater than those derived from the original novoloid fiber. This is because grafted poly(methyl methacrylate) decomposes into monomer and gaseous products, forming pores on the surface and inside of the novoloid fiber on heating. © 1993 John Wiley & Sons, Inc.  相似文献

A plasma-sprayed zinc silicate phosphor was heat-treated at ∼800°C informing gas; the major phase present was the metastable β phase, as revealed by bright yellow fluorescence under uv excitation. On heat treatment at ∼1000°C in air, a green, fluorescence developed as a result of the formation of the stable a phase.
