An experimental study was carried out on the ultrafiltration (UF) of protein-containing solutions under different conditions, as compared with a solution of a linear synthetic polymer. Three different fluidynamic regimes were investigated, namely, unstirred batch system, stirred batch system, and recirculating system. The results obtained substantially agree with the predictions of the gel polarization model. A significant effect of the electrolytes on the UF flux has been observed, which can be attributed to solute–solute interaction. Peritoneal dialysis (PD) represents the dialysis modality of choice for pediatric patients with end-stage kidney disease. Indeed, compared with hemodialysis (HD), it offers many advantages, including more flexibility, reduction of the risk of hospital-acquired infections, preservation of residual kidney function, and a better quality of life. However, despite these positive aspects, PD may be associated with several long-term complications that may impair both patient’s general health and PD adequacy. In this view, chronic inflammation, caused by different factors, has a detrimental impact on the structure and function of the peritoneal membrane, leading to sclerosis and consequent PD failure both in adults and children. Although several studies investigated the complex pathogenic pathways underlying peritoneal membrane alterations, these processes remain still to explore. Understanding these mechanisms may provide novel approaches to improve the clinical outcome of pediatric PD patients through the identification of subjects at high risk of complications and the implementation of personalized interventions. In this review, we discuss the main experimental and clinical experiences exploring the potentiality of the proteomic analysis of peritoneal fluids and extracellular vesicles as a source of novel biomarkers in pediatric peritoneal dialysis. 相似文献

mitochondrial neuro-gastrointestinal encephalomyopathy (MNGIE) is a rare genetic disorder characterized by thymidine phosphorylase (TP) enzyme defect. The absence of TP activity induces the imbalance of mitochondrial nucleotide pool, leading to impaired mitochondrial DNA (mtDNA) replication and depletion. Since mtDNA is required to ensure oxidative phosphorylation, metabolically active tissues may not achieve sufficient energy production. The only effective life-saving approach in MNGIE has been the permanent replacement of TP via allogeneic hematopoietic stem cell or liver transplantation. However, the follow-up of transplanted patients showed that gut tissue changes do not revert and fatal complications, such as massive gastrointestinal bleeding, can occur. The purpose of this study was to clarify whether the reintroduction of TP after transplant can recover mtDNA copy number in a normal range. Using laser capture microdissection and droplet-digital-PCR, we assessed the mtDNA copy number in each layer of full-thickness ileal samples of a naive MNGIE cohort vs. controls and in a patient pre- and post-TP replacement. The treatment led to a significant recovery of gut tissue mtDNA amount, thus showing its efficacy. Our results indicate that a timely TP replacement is needed to maximize therapeutic success before irreversible degenerative tissue changes occur in MNGIE. 相似文献

Neuropathy development is a major dose-limiting side effect of anticancer treatments that significantly reduces patient’s quality of life. The inadequate pharmacological approaches for neuropathic pain management warrant the identification of novel therapeutic targets. Mitochondrial dysfunctions that lead to reactive oxygen species (ROS) increase, cytosolic Ca2+ imbalance, and lactate acidosis are implicated in neuropathic pain pathogenesis. It has been observed that in these deregulations, a pivotal role is played by the mitochondrial carbonic anhydrases (CA) VA and VB isoforms. Hence, preclinical studies should be conducted to assess the efficacy of two novel selenides bearing benzenesulfonamide moieties, named 5b and 5d, and able to inhibit CA VA and VB against paclitaxel-induced neurotoxicity in mice. Acute treatment with 5b and 5d (30–100 mg/kg, per os – p.o.) determined a dose-dependent and long-lasting anti-hyperalgesic effect in the Cold plate test. Further, repeated daily treatment for 15 days with 100 mg/kg of both compounds (starting the first day of paclitaxel injection) significantly prevented neuropathic pain development without the onset of tolerance to the anti-hyperalgesic effect. In both experiments, acetazolamide (AAZ, 100 mg/kg, p.o.) used as the reference drug was partially active. Moreover, ex vivo analysis demonstrated the efficacy of 5b and 5d repeated treatments in reducing the maladaptive plasticity that occurs to glia cells in the lumbar portion of the spinal cord and in improving mitochondrial functions in the brain and spinal cord that were strongly impaired by paclitaxel-repeated treatment. In this regard, 5b and 5d ameliorated the metabolic activity, as observed by the increase in citrate synthase activity, and preserved an optimal mitochondrial membrane potential (ΔΨ) value, which appeared depolarized in brains from paclitaxel-treated animals. In conclusion, 5b and 5d have therapeutic and protective effects against paclitaxel-induced neuropathy without tolerance development. Moreover, 5b and 5d reduced glial cell activation and mitochondrial dysfunction in the central nervous system, being a promising candidate for the management of neuropathic pain and neurotoxicity evoked by chemotherapeutic drugs. 相似文献

Philadelphia-negative chronic myeloproliferative neoplasms (MPNs) represent a group of hematological disorders that are traditionally considered as indistinct slow progressing conditions; still, a subset of cases shows a rapid evolution towards myelofibrotic bone marrow failure. Specific abnormalities in the megakaryocyte lineage seem to play a central role in this evolution, especially in the bone marrow fibrosis but also in the induction of myeloproliferation. In this review, we analyze the current knowledge of prognostic factors of MPNs related to their evolution to myelofibrotic bone marrow failure. Moreover, we focused the role of the megakaryocytic lineage in the various stages of MPNs, with updated examples of MPNs in vitro and in vivo models and new therapeutic implications. 相似文献

Delay/Disruption-Tolerant Network- ing (DTN) originated from research on Interplanetary Internet and still today space appli- cations are the most important application field and research stimulus. This paper investigates DTN communications between the Earth and the far side of the Moon, by means of a lunar orbiter acting as relay. After an introductory part, the paper presents a comprehensive ana- lysis of the DTN performance that can be ac- hieved on the identified communication scen- ario. The focus is on the evaluation of the state- of-the-art ability of Interplanetary Overlay Network (ION), the NASA DTN implementation of Bundle Protocol (BP) and Contact Graph Routing (CGR), to meet the many challenges of the space communication scenario investigated (and more generally of a future interplay- netary Internet): intermittent links, network partitioning, scarce bandwidth, long delays, dyn- amic routing, handling of high priority and emergency traffic, interoperability issues. A study of security threats and Bundle Security Protocol (BSP) countermeasures complete the work. The many results provided, confirm the essential role of DTN in future space communications. 相似文献

Several DSP algorithms need to remove high-frequency or impulsive noise while preserving edges, e.g., in speech and image processing applications: median filtering has been proved to be more effective for achieving this goal than other filtering techniques. Efficient architectural implementation for real-time applications involves a careful VLSI design, which takes into account modularity, regularity, adaptability, scalability, throughput, circuit complexity and fault tolerance. Four new architectural approaches are presented and evaluated in this paper to deal with different application and implementation constraints. They are: the serial-input polarizing median filter, the floating median filter, the pipelined polarizing median filter and the pipelined sorting median filter. The 1st and the 2nd architectures are based on majority voting, while the 3rd and the 4th ones are based on sorting techniques. The first evidence of out‐of‐plane resonances in hybrid metallo‐dielectric quasi‐crystal (QC) nanostructures composed of metal‐backed aperiodically patterned low‐contrast dielectric layers is reported. Via experimental measurements and full‐wave numerical simulations, these resonant phenomena are characterized with specific reference to the Ammann‐Beenker (quasi‐ periodic, octagonal) tiling lattice geometry and the underlying physics is investigated. In particular, it is shown that, by comparison with standard periodic structures, a moderately richer spectrum of resonant modes may be excited, due to the easier achievement of phase‐matching conditions endowed by its denser Bragg spectrum. Such modes are characterized by a distinctive plasmonic or photonic behavior, discriminated by their field distribution and dependence on the metal film thickness. Moreover, the response is accurately predicted via computationally affordable periodic‐approximant‐based numerical modeling. The enhanced capability of QCs to control number, spectral position, and mode distribution of hybrid resonances may be exploited in a variety of possible applications. To assess this aspect, label‐free biosensing is studied via characterization of the surface sensitivity of the proposed structures with respect to local refractive index changes. Moreover, it is also shown that the resonance‐engineering capabilities of QC nanostructures may be effectively exploited in order to enhance the absorption efficiency of thin‐film solar cells. 相似文献
