SR proteins are required for constitutive pre-mRNA splicing and also regulate alternative splice site selection in a concentration-dependent manner. They have a modular structure that consists of one or two RNA-recognition motifs (RRMs) and a COOH-terminal arginine/serine-rich domain (RS domain). We have analyzed the role of the individual domains of these closely related proteins in cellular distribution, subnuclear localization, and regulation of alternative splicing in vivo. We observed striking differences in the localization signals present in several human SR proteins. In contrast to earlier studies of RS domains in the Drosophila suppressor-of-white-apricot (SWAP) and Transformer (Tra) alternative splicing factors, we found that the RS domain of SF2/ASF is neither necessary nor sufficient for targeting to the nuclear speckles. Although this RS domain is a nuclear localization signal, subnuclear targeting to the speckles requires at least two of the three constituent domains of SF2/ASF, which contain additive and redundant signals. In contrast, in two SR proteins that have a single RRM (SC35 and SRp20), the RS domain is both necessary and sufficient as a targeting signal to the speckles. We also show that RRM2 of SF2/ASF plays an important role in alternative splicing specificity: deletion of this domain results in a protein that, although active in alternative splicing, has altered specificity in 5' splice site selection. These results demonstrate the modularity of SR proteins and the importance of individual domains for their cellular localization and alternative splicing function in vivo.  相似文献

The creep and creep fracture behavior of two hot-pressed aluminas are presented, for both flexural and tensile testing. Steady-state power-law creep is observed with a stress exponent of about 2 for each material. Three distinct fracture regimes are found. At high stress in flexure, fracture occurs by slow crack growth with a high stress dependence of the failure time. At intermediate stresses, in both flexure and tension, creep fracture occurs by multiple microcracking after modest strains. Failure times exhibit a modest stress dependence (stress exponent of 2.5 in tension and 3 in flexure), with a constant failure strain equal to 0.09. The failure times are considerably longer in flexure than in tension, because of the constraint imposed on crack growth by the bending geometry. We conclude that flexure cannot be used for creep lifetime assessment, even in simple, single-phase materials such as Al2O3. At low stresses, in tension, failure also exhibits a modest stress dependence but with a much higher failure strain. The material shows the onset of super-plastic behavior.  相似文献

The study of behavioral and neurophysiological mechanisms involved in rat spatial cognition provides a basis for the development of computational models and robotic experimentation of goal-oriented learning tasks. These models and robotics architectures offer neurobiologists and neuroethologists alternative platforms to study, analyze and predict spatial cognition based behaviors. In this paper we present a comparative analysis of spatial cognition in rats and robots by contrasting similar goal-oriented tasks in a cyclical maze, where studies in rat spatial cognition are used to develop computational system-level models of hippocampus and striatum integrating kinesthetic and visual information to produce a cognitive map of the environment and drive robot experimentation. During training, Hebbian learning and reinforcement learning, in the form of Actor-Critic architecture, enable robots to learn the optimal route leading to a goal from a designated fixed location in the maze. During testing, robots exploit maximum expectations of reward stored within the previously acquired cognitive map to reach the goal from different starting positions. A detailed discussion of comparative experiments in rats and robots is presented contrasting learning latency while characterizing behavioral procedures during navigation such as errors associated with the selection of a non-optimal route, body rotations, normalized length of the traveled path, and hesitations. Additionally, we present results from evaluating neural activity in rats through detection of the immediate early gene Arc to verify the engagement of hippocampus and striatum in information processing while solving the cyclical maze task, such as robots use our corresponding models of those neural structures.  相似文献

The technical feasibility and performance of photocatalytic degradation of four water-soluble pesticides (diuron, imidacloprid, formetanate and methomyl) have been studied at pilot scale in two well-defined systems of special interest because natural-solar UV light can be used: heterogeneous photocatalysis with titanium dioxide and homogeneous photocatalysis by photo-Fenton. The pilot plant is made up of compound parabolic collectors (CPCs) specially designed for solar photocatalytic applications. Experimental conditions allowed disappearance of pesticide and degree of mineralisation achieved in the two photocatalytic systems to be compared. In order to assure that the photocatalytic results are consistent, hydrolysis and photolysis tests have been performed with the four pesticides. The initial concentration tested with imidacloprid, formetanate and methomyl was 50 and 30 mg/l with diuron, and the catalyst concentrations were 200 mg/l and 0.05 mM with TiO2 and iron, respectively. Total disappearance of the parent compounds and 90% mineralisation have been attained with all pesticides tested, methomyl being the most difficult to be degraded with both treatments. First-order rate constants, initial rate, time necessary for mineralising 90% of the initial TOC and hydrogen peroxide consumption were calculated in all cases, enabling comparison both of treatments and of the selected pesticide reactivity.  相似文献

In the past 15 years our research group has been creating new optical devices to characterize in real time the extrusion process. These detectors are made of a slit‐die fitted at the extruder exit from where the molten polymer flows, with a pair of transparent windows that allows a light beam to pass orthogonally through the molten flow. Following the reduction of the transmitted light intensity one is able to quantify the turbidity, which is a function of the type, concentration and particle size and shape of the second phase present in the flow. By evaluating the scattering pattern of a laser beam (LALLS) it is possible to get information upon the morphology of the molten polymeric system in real time during the extrusion. With the interposition of a pair of crossed polarizers in the optical beam, rheo‐polarimetry, it is possible to evaluate quantitatively the flow birefringence, which is a function of the degree of the polymer matrix orientation. In order to improve the thermal efficiency of building thermal energy storage (TES) systems, the feasibility of using encapsulated phase change materials (EPCMs) as heat storage media is analysed in this work. Specifically, the finite element method is used to perform thermal behaviour analyses of several EPCMs. These analyses include technical and economic assessments in order to identify the best combination of PCM and shell material, using as main parameters: thermal energy storage, heat transfer rate, materials cost, among others. The results show that EPCMs composed by Na2SO4·6H2O as PCM and covered by stainless steel highlight as TES materials.  相似文献

Radiation damage induced by neutron irradiation was studied in undoped MgO crystals and in MgO doped with either iron, hydrogen or lithium impurities. The oxygen-vacancy concentration produced by irradiation increases with neutron fluence. The net production rates resulting from irradiations with 14.8 MeV neutrons are about twice those produced by fission neutrons.

In nominally pure crystals, the oxygen-vacancy concentration incurred by the fission-neutron irradiation is higher in crystals with a larger number of inherent impurities (such as iron) due to trapping of interstitials by impurities. Suppression of these defects is observed in MgO:H crystals and attributed to migration of oxygen vacancies to microcavities filled with H2 gas. In MgO:Li crystals irradiated with neutron fluences below 1018 n/cm2, most of the oxygen vacancies are camouflaged as hydride ions. Nanoindentation experiments show that hardness increases with neutron fluence and is independent of the presence of lithium in the crystal. Comparison between a neutron-irradiated and a thermochemically reduced crystal containing similar concentrations of oxygen vacancies shows that 70% of the neutron-irradiation hardening is produced by interstitials, 30% by oxygen vacancies and a negligible amount by higher-order point defects.  相似文献

In this paper we present deterministic parallel algorithms for the coarse-grained multicomputer (CGM) and bulk synchronous parallel (BSP) models for solving the following well-known graph problems: (1) list ranking, (2) Euler tour construction in a tree, (3) computing the connected components and spanning forest, (4) lowest common ancestor preprocessing, (5) tree contraction and expression tree evaluation, (6) computing an ear decomposition or open ear decomposition, and (7) 2-edge connectivity and biconnectivity (testing and component computation). The algorithms require O(log p) communication rounds with linear sequential work per round (p = no. processors, N = total input size). Each processor creates, during the entire algorithm, messages of total size O(log (p) (N/p)) . The algorithms assume that the local memory per processor (i.e., N/p ) is larger than p ε , for some fixed ε > 0 . Our results imply BSP algorithms with O(log p) supersteps, O(g log (p) (N/p)) communication time, and O(log (p) (N/p)) local computation time. It is important to observe that the number of communication rounds/ supersteps obtained in this paper is independent of the problem size, and grows only logarithmically with respect to p . With growing problem size, only the sizes of the messages grow but the total number of messages remains unchanged. Due to the considerable protocol overhead associated with each message transmission, this is an important property. The result for Problem (1) is a considerable improvement over those previously reported. The corrosion evolution over time of a carbon steel rotating disk immersed in aerated NaCl solutions was analyzed using a superposition model. Using this approach, partial polarization curves for iron oxidation and oxygen reduction were synthesized from experimental current-potential data at different corrosion time in order to determine the kinetics parameters, corrosion potential and current density of the underlying anodic and cathodic subprocesses. The distinctive features of the polarization curves are well described in terms of the simplifying assumptions of the model. In particular, the time evolution of the corrosion current density was linked to the morphology of the corroding surface under different NaCl concentrations.  相似文献
