The interaction between alpha-actinin and actin was further characterized using natural and synthetic peptides of actin together with anti-actin antibodies of known specificity. We demonstrated that two alpha-actinin binding sequences on actin are located within residues 112-125 and 360-372. Each peptide was shown to directly bind alpha-actinin and was able to dissociate the alpha-actinin-actin complex using solid phase binding assays and cosedimentation experiments. Taking into account the three-dimensional structure of actin (Kabsch, W., Mannherz, H. G., Suck, D., Pai, E. F., and Holmes, K. C. (1990) Nature 347, 37-44), we postulate that these two segments, proximal in the actin structure, are part of the same site. In addition, we compared these two segments with those recently found for filamin (Méjean, C., Lebart, M. C., Boyer, M., Roustan, C., and Benyamin, Y. (1992) Eur. J. Biochem. 209, 555-562), Egan, S., Stewart, M., Stossel, T. P., Kwiatkowski, D. J., and Hartwig, J. H. (1990) J. Cell Biol. 111, 1089-1105), and concluded that the two actin-binding proteins interact with closely spaced or overlapping but not identical sequences of actin subdomain 1.  相似文献

In this paper, a novel algorithm for image encryption based on hash function is proposed. In our algorithm, a 512-bit long external secret key is used as the input value of the salsa20 hash function. First of all, the hash function is modified to generate a key stream which is more suitable for image encryption. Then the final encryption key stream is produced by correlating the key stream and plaintext resulting in both key sensitivity and plaintext sensitivity. This scheme can achieve high sensitivity, high complexity, and high security through only two rounds of diffusion process. In the first round of diffusion process, an original image is partitioned horizontally to an array which consists of 1,024 sections of size 8 × 8. In the second round, the same operation is applied vertically to the transpose of the obtained array. The main idea of the algorithm is to use the average of image data for encryption. To encrypt each section, the average of other sections is employed. The algorithm uses different averages when encrypting different input images (even with the same sequence based on hash function). This, in turn, will significantly increase the resistance of the cryptosystem against known/chosen-plaintext and differential attacks. It is demonstrated that the 2D correlation coefficients (CC), peak signal-to-noise ratio (PSNR), encryption quality (EQ), entropy, mean absolute error (MAE) and decryption quality can satisfy security and performance requirements (CC <0.002177, PSNR <8.4642, EQ >204.8, entropy >7.9974 and MAE >79.35). The number of pixel change rate (NPCR) analysis has revealed that when only one pixel of the plain-image is modified, almost all of the cipher pixels will change (NPCR >99.6125 %) and the unified average changing intensity is high (UACI >33.458 %). Moreover, our proposed algorithm is very sensitive with respect to small changes (e.g., modification of only one bit) in the external secret key (NPCR >99.65 %, UACI >33.55 %). It is shown that this algorithm yields better security performance in comparison to the results obtained from other algorithms.  相似文献

We demonstrate the use of heat to count microscopic particles. A thermal particle detector (TPD) was fabricated by combining a 500-nm-thick silicon nitride membrane containing a thin-film resistive temperature detector with a silicone elastomer microchannel. Particles with diameters of 90 and 200 μm created relative temperature changes of 0.11 and ?0.44 K, respectively, as they flowed by the sensor. A first-order lumped thermal model was developed to predict the temperature changes. Multiple particles were counted in series to demonstrate the utility of the TPD as a particle counter.  相似文献

The article has investigated the function of water belonging to the set class of quality by established indicators of the composition and properties. It has been shown that the employment of the mathematical apparatus of the tolerance measuring control allows using Taguchi and Harrington functions to assess the degree of the sought-for belonging both within the boundaries of the allowable interval and outside it, thereby increasing the informational content of control and monitoring. A special attention is given to problems of assessing the function of belonging in classification of waters having indefiniteness zones between classes. The proposed methods are used in treatment of real statistical data characterizing water quality. Algorithms of treatment are methodological recommendations facilitating the employment of the developed approach for solving practical problems assessing the composition and properties of water with the aim of water use.  相似文献

Direct extraction of molybdenum from sulfate solution with synergistic extractants (mixture of D2EHPA and TBP) was studied in the rotation column. The influence of extractant concentration and initial pH of aqueous phase was studied in the bench scale experiments. The outcomes demonstrated that the synergistic solvent extraction enhances the constancy of the extracted complexes for transfer into the organic phase. In the continuous experiments, the effect of different operating parameters such as speed of agitation, inlet solvent flow rate and inlet aqueous flow rate on the holdup, mean drop size, drop size distribution, slip and characteristic velocities and extraction percentage were examined. Modified correlations were proposed for prediction of hydrodynamic parameters with consideration of reaction extraction condition in the rotation column. Furthermore, these correlations were compared with the experimental data. According to the results, the direct extraction of Mo (VI) from aqueous solution and sulfuric media with extraction efficiency of 90.4% was obtained at higher rotor speed (240 r·min-1 rpm) in this column.  相似文献

Actin, together with associated proteins, such as myosin, cross-linking or capping proteins, has been observed in all eukaryotic cells. Presence of actin or actin-like proteins has also been reported in prokaryotic organisms belonging to the cyanobacteria. Our aim was first to extend the characterization of an actin-like protein to another prokaryotic cell, i.e. Spirulina, then to compare the antigenic reactivity of this new protein with that of Synechocystis and skeletal actins. We observed that some of the conserved antigenic epitopes corresponded to actin regions known to interact with cross-linking proteins. We also report for the first time that alpha-actinin and filamin purified from chicken gizzard both interact with a prokaryotic actin-like protein. Finally, we searched for the occurrence of a cross-linking protein in these cyanobacteria and identified a 105-kDa protein as an alpha-actinin-like protein using specific antibodies.  相似文献

The human sensory test is often used for obtaining the sensory quantities of odors, however, the fluctuation of results due to the expert's condition can cause discrepancies among panelists. Authors have studied the artificial odor discrimination system using a quartz resonator sensor and a back-propagation neural network as the recognition system, however, the unknown category of odor is always recognized as the known category of odor. In this paper, a kind of fuzzy algorithm for learning vector quantization (LVQ) is developed and used as a pattern classifier. In this type of fuzzy LVQ, the neuron activation is derived through fuzziness of the input data, so that the neural system could deal with the statistics of the measurement error directly. During learning, the similarity between the training vector and the reference vectors are calculated, and the winning reference vector is updated by shifting the central position of the fuzzy reference vector toward or away from the input vector, and by modifying its fuzziness. Two types of fuzziness modifications are used, i.e., a constant modification factor and a variable modification factor. This type of fuzzy-neuro (FN) LVQ is different in nature from fuzzy algorithm (FA) LVQ, and in this paper, the performance of FNLVQ network is compared with that of FALVQ in an artificial odor recognition system. Experimental results show that both FALVQ and FNLVQ could provide high recognition probability in determining various known categories of odors, however, the FNLVQ neural system has the ability to recognize the unknown category of odor that could not be recognized by the FALVQ neural system.  相似文献

α-Actinin release and its degradation from myofibrils Z-line were studied in post mortem white dorsal muscle from bass and sea trout stored at 4°C and 10°C. Using α-actinin specific antibodies, we show that this protein is rapidly released within the first 24 h for the two species, and reaches a plateau within 4 days. Proteolysis take place very rapidly in bass muscle yielding 80 and 40 kDa fragments from α-actinin as major bands of proteolysis. Sea trout muscle is more resistant, and muscle stored at 4°C is not significantly α-actinin degraded even 10 days after death. In the case of sea trout muscle stored at 10°C, an increasing quantity of 80 and 40 kDa fragment can be observed after the third day. These results show that release and proteolysis of α-actinin are time- and temperature-dependent processes that take place at the early stages of fish storage. Furthermore, we observed that proteolysis of α-actinin seems to be dependent on fish species. In both species studied, the early release of α-actinin comes before the degradation of released molecules, and appears as a biphasic process throughout the disorganisation of post mortem muscle in fish cold-stored above 0°C.  相似文献

The cabbage aphid Brevicoryne brassicae is a specialist herbivore that sequesters glucosinolates from its host plant as a defense against its predators. It is unknown to what extent parasitoids are affected by this sequestration. We investigated herbivore-mediated effects of glucosinolates on the parasitoid wasp Diaeretiella rapae and the predator Episyrphus balteatus. We reared B. brassicae on three ecotypes of Arabidopsis thaliana that differ in glucosinolate content and on one genetically transformed line with modified concentrations of aliphatic glucosinolates. We tested aphid performance and the performance and behavior of both natural enemies. We correlated this with phloem and aphid glucosinolate concentrations and emission of volatiles. Brevicoryne brassicae performance correlated positively with concentrations of both aliphatic and indole glucosinolates in the phloem. Aphids selectively sequestered glucosinolates. Glucosinolate concentration in B. brassicae correlated negatively with performance of the predator, but positively with performance of the parasitoid, possibly because the aphids with the highest glucosinolate concentrations had a higher body weight. Both natural enemies showed a positive performance-preference correlation. The predator preferred the ecotype with the lowest emission of volatile glucosinolate breakdown products in each test combination, whereas the parasitoid wasp preferred the A. thaliana ecotype with the highest emission of these volatiles. The study shows that there are differential herbivore-mediated effects of glucosinolates on a predator and a parasitoid of a specialist aphid that selectively sequesters glucosinolates from its host plant.  相似文献
