In several autoimmune disorders, including rheumatoid arthritis (RA), autoantibodies are thought to be the driving force of pathogenicity. Glycosylation of the Fc-part of human Igs is known to modulate biological activity. Hitherto, glycosylation of human IgG-Fc has been analyzed predominantly at the level of total serum IgG, revealing reduced galactosylation in RA. Given the pathogenic relevance of autoantibodies in RA, we wished, in the present study, to address the question whether distinct Fc-glycosylation features are observable at the level of antigen-specific IgG subpopulations. For this purpose, we have developed a method for the microscale purification and Fc-glycosylation analysis of anti-citrullinated peptide antibodies (ACPA). ACPA represent a group of autoantibodies that occur with unique specificity in RA patients. Their presence is associated with increased inflammatory disease activity and rapid joint destruction. Results indicate that ACPA of the IgG1 subclass vary considerably from total serum IgG1 with respect to Fc-galactosylation, with galactosylation being higher on ACPA than on serum IgG1 for some patients, while other patients show higher galactosylation on serum IgG1 than on ACPA. Using this method, studies can be performed on the biological and clinical relevance of ACPA glycosylation within RA patient cohorts.  相似文献

The specific absorption rates (SAR) determined computationally in the specific anthropomorphic mannequin (SAM) and anatomically correct models of the human head when exposed to a mobile phone model are compared as part of a study organized by IEEE Standards Coordinating Committee 34, Sub-Committee 2, and Working Group 2, and carried out by an international task force comprising 14 government, academic, and industrial research institutions. The detailed study protocol defined the computational head and mobile phone models. The participants used different finite-difference time-domain software and independently positioned the mobile phone and head models in accordance with the protocol. The results show that when the pinna SAR is calculated separately from the head SAR, SAM produced a higher SAR in the head than the anatomically correct head models. Also the larger (adult) head produced a statistically significant higher peak SAR for both the 1- and 10-g averages than did the smaller (child) head for all conditions of frequency and position.  相似文献

We analyzed the past and future trends in river export of dissolved and particulate nitrogen (N), phosphorus (P) and carbon (C) to the coastal waters of China, for sixteen rivers, as calculated by the Global NEWS models (Nutrient Export from WaterSheds). Between 1970 and 2000, the dissolved N and P export increased significantly, while export of other nutrients changed less. We analyzed the future trends (2000-2050) for the Millennium Ecosystem Assessment (MEA) scenarios. In general, the largest increases of dissolved nutrients export are projected for the Global Orchestration scenario, assuming a globalized world and a reactive approach toward environmental management. Future trends in river export of nutrients vary largely among basins, nutrient forms and scenarios. We calculate both increasing and decreasing trends between 2000 and 2050. We also identify the sources contributing to the nutrient export. For selected river basins we present results for alternative scenarios, which are based on the Global Orchestration scenario, but assume more environmental management. This illustrates how the NEWS models can be useful in regional analyses for decision making.  相似文献

This study focuses on the uncertainties in the fate of nitrogen (N) in the Netherlands. Nitrogen inputs into the Netherlands in products, by rivers, and by atmospheric deposition, and microbial and industrial fixation of atmospheric N2 amount to about 4450 Gg N y–1. About 60% of this N is transported out of the Netherlands in products. The fate of the remaining 40%, however, is less clear. We discuss uncertainties in losses to the atmosphere (as ammonia or through denitrification), by leaching and runoff, and in N accumulation in biomass and soils. These processes may account for the fate of about 40% of the N in the Netherlands, and for the fate of about 60% of the N in Dutch agricultural soils. Reducing uncertainties in the estimates of these fluxes is necessary for reducing the impact of excess N in the environment. In particular, monitoring the environmental effects of ammonia emissions and nitrate leaching to groundwater and aquatic systems requires an increased understanding of the fate of N. Uncertainties arise because (1) some N fluxes cannot be measured directly and are usually quantified indirectly as the balance in N budgets, (2) direct measurements of N fluxes have inevitable inaccuracies, (3) lack of experimental data and other information (e.g. statistics) needed for upscaling, (4) large spatial and temporal variability of fluxes, and (5) poor understanding of the processes involved. These uncertainties can be reduced by additional experimental studies and by further development of process-based models and N budget studies. We prioritize these future research needs according to a range of different criteria.  相似文献

In this paper, present-day emissions of greenhouse gases and acidifying compounds from agriculture are analysed at the farm level. Quantitative estimates are given for these emissions from three nested systems in the Netherlands: the agricultural sector, greenhouse horticulture, and tomato cultivation under glass. Total emissions are subdivided into emissions from biogenic sources and abiogenic sources. We conclude that, although most of the emissions from the agricultural sector have biogenic sources, those from abiogenic sources should not be neglected. Abiogenic emissions are mainly from greenhouse horticulture. The cost-effectiveness of options to reduce carbon dioxide (CO2) and nitrogen oxides (NOx) emissions from on-farm combustion of natural gas in tomato cultivation under glass is analysed. An inventory is given of technical reduction options that are presently available in practice. Based on information about the costs and the reduction potential of each option, cost-efficiency curves are derived for both types of emissions. Relative to a situation where none of the described options were applied (early nineties), CO2 and NOx emissions from tomato cultivation can be reduced at most by about 70% and 75%, respectively, by combinations of technical options. In this study, the safety of cyclists at unsignalized priority intersections within built-up areas is investigated. The study focuses on the link between the characteristics of priority intersection design and bicycle–motor vehicle (BMV) crashes. Across 540 intersections that are involved in the study, the police recorded 339 failure-to-yield crashes with cyclists in four years. These BMV crashes are classified into two types based on the movements of the involved motorists and cyclists:

    type I: through bicycle related collisions where the cyclist has right of way (i.e. bicycle on the priority road);
  • • 
    type II: through motor vehicle related collisions where the motorist has right of way (i.e. motorist on the priority road).
The probability of each crash type was related to its relative flows and to independent variables using negative binomial regression. The results show that more type I crashes occur at intersections with two-way bicycle tracks, well marked, and reddish coloured bicycle crossings. Type I crashes are negatively related to the presence of raised bicycle crossings (e.g. on a speed hump) and other speed reducing measures. The accident probability is also decreased at intersections where the cycle track approaches are deflected between 2 and 5 m away from the main carriageway. No significant relationships are found between type II crashes and road factors such as the presence of a raised median.  相似文献

We studied the influence of the hydrophobic hydrocarbon chain length of amphiphilic ruthenium dyes on the device performance in solid-state dye-sensitized solar cells. We found that the dyes with longer hydrocarbon chains gave higher efficiency values when used as a sensitizer in solid-state dye-sensitized solar cells. With increasing chain length, we observed higher currents and open-circuit voltages up to a limiting chain length. We attribute this improvement to the expected larger distance between TiO2 and the hole conductor, which seems to suppress recombination effectively.  相似文献
