We previously reported that the abl promoter (Pa) undergoes de novo DNA methylation in the course of chronic myelocytic leukemia (CML). The clinical implications of this finding are the subject of the present study in which samples of CML patients, including a group treated with interferon alpha (IFNalpha) were surveyed. The methylation status of the abl promoter was monitored by polymerase chain reaction (PCR) amplification of the Pa region after digestion with several site-methylation sensitive restriction enzymes. Some 74% of the DNA samples from blood and marrow drawn in the chronic phase were nonmethylated, similar to control samples from non-CML patients. The remaining 26% were partially methylated in the abl Pa region. The latter samples were derived from patients who were indistinguishable from the others on the basis of clinical presentation. Methylated samples were mostly derived from patients known to have a disease of longer duration (26 months v 7.5 months, P = .01). Samples of 30 IFNalpha-treated patients were sequentially analyzed in the course of treatment. Fifteen patients with no evidence of Pa methylation before treatment remained methylation-free. The remainder, who displayed Pa methylation before treatment, reverted to the methylation-free status. The outcome is attributed to IFNalpha therapy, as the Pa methylation status was not reversed in any of the patients treated with hydroxyurea. Methylation of the abl promoter indicates a disease of long-standing, most likely associated with a higher probability of imminent blastic transformation. It appears to predict the outcome of IFNalpha therapy far better than the cytogenetic response.  相似文献

The static magnetic field of a magnetic resonance imaging scanner can be distorted by the presence of materials, perturbing the spatial encoding process in magnetic resonance imaging and often resulting in image artifacts. The relationship between the image artifact size and magnetic susceptibility of the material specimen is of interest to engineers for the design of devices that are to be compatible with the imaging volume of the scanner. In this study, a finite-element method was used to simulate the distorted magnetic field of samples with different susceptibilities. With the knowledge of the external- and self- magnetic field interactions, a Lorentz correction was applied to compute the magnetic field deviation. The simulated results were then validated by the corresponding experimental magnetic resonance images.  相似文献

The development and promotion of biofortified foods plants are a sustainable strategy for supplying essential micronutrients for human health and nutrition. We set out to identify quantitative trait loci (QTL) associated with carotenoid content in cowpea sprouts. The contents of carotenoids, including lutein, zeaxanthin, and β-carotene in sprouts of 125 accessions were quantified via high-performance liquid chromatography. Significant variation existed in the profiles of the different carotenoids. Lutein was the most abundant (58 ± 12.8 mg/100 g), followed by zeaxanthin (14.7 ± 3.1 mg/100 g) and β-carotene (13.2 ± 2.9 mg/100 g). A strong positive correlation was observed among the carotenoid compounds (r ≥ 0.87), indicating they can be improved concurrently. The accessions were distributed into three groups, following their carotenoid profiles, with accession C044 having the highest sprout carotenoid content in a single cluster. A total of 3120 genome-wide SNPs were tested for association analysis, which revealed that carotenoid biosynthesis in cowpea sprouts is a polygenic trait controlled by genes with additive and dominance effects. Seven loci were significantly associated with the variation in carotenoid content. The evidence of variation in carotenoid content and genomic regions controlling the trait creates an avenue for breeding cowpea varieties with enhanced sprouts carotenoid content.  相似文献

The potential for utilizing nuclear magnetic resonance measurements for evaluating the quality of fresh prunes was investigated. In a 2 T magnetic flux density, the chemical shift between proton hydrogens of soluble solids and those of water is less than 100 Hz. The band width of the water resonance spectrum of fruits is relatively large and therefore it is difficult to resolve between the spectral peaks of water and soluble solids. Measurements with a 100 mm Birdcage coil failed to resolve between the resonance peaks of soluble solids and water. Good resolution was achieved with a 20 mm diameter Surface coil. Signals (free induction decay) of 30 sugar prunes were acquired, and the spectra were calculated. Tuning and matching the coil before each measurement were not necessary, and the results were better when the magnetic field was not shimmed. The ratio of the soluble solids peak height to the water peak height correlated well (r = 0.907) with the soluble solids content of the prunes, as estimated by a refractometer.  相似文献

Photonic crystal microcavities, formed by local defects within an otherwise perfectly periodic structure, can be used as narrowband optical resonators and filters. The coupled-cavity waveguide (CCW) is a linear array of equally spaced identical microcavities. Tunneling of light between microcavities forms a guiding effect, with a central frequency and bandwidth controlled by the local defects' parameters and spacing, respectively. We employ cavity perturbation theory to investigate the sensitivity of microcavities and CCWs to random structure inaccuracies. For the microcavity, we predict a frequency shift that is due to random changes in the lattice structure and show an approximate linear dependence between the standard deviation of the structure inaccuracy and that of the resonant frequency. The effect of structural inaccuracy on the CCW devices, however, is different; it has practically no effect on the CCW performance if it is below a certain threshold but may destroy the CCW if this threshold is exceeded.  相似文献

Real-time processing of MRIs is reported as a method of 3D tracking of mechanical devices within the field of view using passive microcoil fiducials. The specific implementation described makes use of two scan planes for full 3D tracking of a 5-DOF manipulator arm used for prostate biopsy under image guidance. Real-time tracking was observed with a maximum update rate of 0.42 frames per second for a maximum probe velocity of 10 mm/s. The localization of fiducials had a mean error of 0.36 (plusmn0.17) mm (p < 0.02), leading to a mean error in the needle tip position of 2.6 (plusmn0.3) mm (p < 0.05).  相似文献

Recent research on mobile Internet protocol television and digital right management (DRM) interconnections has focused on multimedia technologies designed to enhance content scalability and adaptive content distribution. However, due to the architectural and scalable limitations, recent systems are not flexible and securable with respect to their adaptive content distribution and protective policy management. Therefore, we propose a content protective multi‐agent platform that provides secure multimedia services, correlation management, pattern‐based management, and multisource multi‐use (MsMu)‐based services. Our architecture, supported by DRM, lets us create a rich set of MsMu‐based content protection and seamless multimedia services through the extension of one source multi‐use (OsMu)‐based content services. We have verified our platform, which provides scalable and securable services with a 17% lower service response time by using a testbed.  相似文献

We apply multiresolution techniques to study the effective properties of boundary value problems. Conditions under which boundary values are preserved in the effective (‘homogenized') formulation are developed and discussed. Relations between the eigenfunctions and eigenvalues of the generic formulation and those of the effective formulation are explored. Applications to the construction of effective Green function in a complex lamination are discussed. The analytic results are demonstrated via numerical computations.  相似文献

“Energy Tower” is a technology for producing renewable and clean electricity by means of cooling hot and dry air, which is continuously supplied to arid lands. We assess the potential of an Energy Tower by incorporating topographic and meteorological parameters into a computational model, providing evaluations for the net power production and the electricity production cost. We formulate a highly simplified model for the Energy Tower’s flow, setup and process a spatial dataset of topographic and Meteorological upper air parameters. The model was applied to the Australian continent. A model simulation of one annual cycle enabled the ranking and selection of promising sites. The highest potential for energy towers is in the Port Hedland region, where favorable meteorological and topographic conditions would result in high average net power (≈370 ± 160 MW), potentially providing the electricity needs of 0.5 million people, for an economically competitive costs (3.5 ¢kWh).  相似文献
