Conventional enzyme-linked immunosorbent assay (ELISA) is a gold standard for screening antibodies and testing for protein or antigen presence. A significant limitation of this assay resides in the fact that only one analyte can be assessed per microplate well. Here, we describe and investigate a new technology consisting of an automated ELISA system in which up to 10 analytes can be measured within one single well, thus improving productivity, accuracy, and repeatability by reducing the amount of human labor required. Another strength of the platform is that a user can load any necessary sets/subsets of beads to perform required assays, with improved flexibility compared to manufactured-loaded arrays for multiplex analysis. We also demonstrate that this system can be used to determine the pathogenicity (i.e., presence of Shiga toxins) and serotype (i.e., Escherichia coli O157) of E. coli isolates.  相似文献

We present a simple optical micro-system used to measure the transmission spectra of oocytes in order to qualify their maturation stage. Two applications of the device are possible: (i) the evaluation of the maturation stages of oocytes, and (ii) the development of a fertilization indicator. For the first application, GV, MI and MII oocytes were also analysed. Transmission spectra allow the 3 maturation stages to be identified but cannot be used to estimate the maturity of an unknown oocyte. Oocytes are subject to continuous development. This is why spectral separation of the 3 maturation stages cannot be made although they can be visually identified. However, the visual observation remains biologist-dependent. We therefore investigated the temporal maturation evolution of the oocytes in terms of transmission spectra and probability analysis. Results show that oocytes to be fertilized should not only be chosen in the MII stage, but also at the right time during the MII stage. This particular aspect requires further investigation. However, spectral measurements could be used as a technique for monitoring the maturation evolution of the oocytes. Fertilized oocytes exhibiting fertilization abnormalities were also tested. The device proved to be an efficient fertilization indicator.  相似文献

Lanthanum therapy is an efficient therapy of hyperphosphoremia by chelating phosphore in the digestive tract. Lanthanum is a silvery white metallic element that belongs to group 3 of the periodic table. This drug is lightly absorbed and has low water solubility. It should be borne in mind that abdominal X‐rays of patients taking lanthanum carbonate may have a radio‐opaque appearance typical of imaging agents. This characteristic is suggested to confirm adherence of the patient by doing an abdominal X‐ray. We describe in our case a particular good compliant patient with slow digestive transit, which ends by pseudo‐occlusion.  相似文献

Zinc phosphate glass is considered to be the main constituent of tribofilms generated under boundary lubrication with zinc dialkyldithiophosphate (ZDDP), a well-known antiwear additive. The reaction occurring during friction between zinc phosphate glasses and steel native iron oxide layer is investigated by both an experimental approach and by Molecular Dynamics simulations (MD). The importance of this “tribochemical” reaction in the general ZDDP antiwear process is discussed.  相似文献

In this study, we are interested in the anti-wear properties of zinc dialkyl phosphate additive (ZP) in comparison with ‘classical’ zinc dialkyldithiophosphate (ZDDP). Friction tests were performed on a reciprocating tribometer using both ball-on-flat and cylinder-on-flat configurations under a Hertzian contact pressure of 0.9 GPa. Experiments were carried out as a function of temperature (25 and 100 °C), sliding speed (25, 50 and 100 mm/s) and additives concentrations. Ball wear scar diameters as well as friction coefficient were measured. In order to better understand the anti-wear mechanisms of these additives, friction tests were followed by surface analyses such as AES (Auger Electron Spectroscopy) and XPS (X-Ray Photoelectron Spectroscopy). Transmission Electron Microscopy (TEM) observations of the ZDDP and ZP tribofilms were also carried out to visualise the generated layers. The anti-wear capability of ZP molecule is discussed.  相似文献

The fabrication of 2D systems for electronic devices is not straightforward, with top‐down low‐yield methods often employed leading to irregular nanostructures and lower quality devices. Here, a simple and reproducible method to trigger self‐assembly of arrays of high aspect‐ratio chiral copper heterostructures templated by the structural anisotropy in black phosphorus (BP) nanosheets is presented. Using quantitative atomic resolution aberration‐corrected scanning transmission electron microscopy imaging, in situ heating transmission electron microscopy and electron energy‐loss spectroscopy arrays of heterostructures forming at speeds exceeding 100 nm s?1 and displaying long‐range order over micrometers are observed. The controlled instigation of the self‐assembly of the Cu heterostructures embedded in BP is achieved using conventional electron beam lithography combined with site specific placement of Cu nanoparticles. Density functional theory calculations are used to investigate the atomic structure and suggest a metallic nature of the Cu heterostructures grown in BP. The findings of this new hybrid material with unique dimensionality, chirality, and metallic nature and its triggered self‐assembly open new and exciting opportunities for next generation, self‐assembling devices.  相似文献

Improperly disposed of dredged sediments contaminated with metals may induce long-term leaching and an increase of metal concentrations in ground waters and vegetal cover plants. The objective of the study was to quantify the sediment-to-plant transfer of Cu, Pb, Hg and Zn with a particular focus on the pathway of Hg and to determine whether the establishment of vegetal cover modifies the metal availability. A pot experiment with rape (Brassica napus), cabbage (Brassica oleraccea) and red fescue (Festuca rubra) was set up using a sediment first spiked with the radioisotope 203Hg. Zinc concentrations (197-543 mg kg(-1) DM) in leaves were higher than Cu concentration (197-543 mg kg(-1) DM), Pb concentration (2.3-2.6 mg kg(-1) DM) and Hg concentration (0.9-1.7 mg kg(-1) DM). Leaves-to-sediment ratios decreased as follows: Zn > Cu > Hg > Pb. According to Ti measurements, metal contamination by dry deposition was less than 1%. Mercury concentration in plant leaves was higher than European and French thresholds. Foliar absorption of volatile Hg was a major pathway for Hg contamination with a root absorption of Hg higher in rape than in cabbage and red fescue. Growth of each species increased Cu solubility. Zinc solubility was increased only in the presence of rape. The highest increase of Cu solubility was observed for red fescue whereas this species largely decreased Zn solubility. Dissolved organic carbon (DOC) measurements suggested that Cu solubilisation could result from organic matter or release of natural plant exudates. Dissolved inorganic carbon (DIC) measures suggested that the high Zn solubility in the presence of rape could originate from a generation of acidity in rape rhizosphere and a subsequent dissolution of calcium carbonates. Consequently, emission of volatile Hg from contaminated dredged sediments and also the potential increase of metal solubility by a vegetal cover of grass when used in phytostabilisation must be taken into account by decision makers.  相似文献

The aim of this study is to assess the tribological behavior of pure crystalline zinc orthophosphate under boundary lubrication in order to model zinc phosphate-based anti-wear additives. Boundary films were generated from α-Zn3(PO4)2 powder dispersed in poly-alpha-olefin oil, at ambient temperature, by means of a steel sphere-on-flat contact in reciprocating motion. Electrical contact resistance and friction coefficient evolutions enable an understanding of the tribological behavior of crystalline zinc orthophosphate at the sliding interface. A conductive atomic force microscope (C-AFM) equipped with a current sensing setup, Raman spectroscopy, and nano-indentation were used to characterize the resulting film. When involved in a tribological contact, zinc orthophosphate powder forms a continuous patchy adherent film, changing its structure to amorphous orthophosphate, on both sliding steel surfaces. Morphological and mechanical properties of the film are discussed with respect to the ZDTP tribofilm models.  相似文献

melatonin (MT) is a hormone produced by the pineal gland at night, involved in the regulation of circadian rhythms. For clinical purposes, exogenous MT administration should mimic the typical nocturnal endogenous MT levels, but its pharmacokinetics is not favourable due to short half-life of elimination. Aim of this study is to examine pharmacokinetics of MT incorporated in solid lipid nanoparticles (SLN), administered by oral and transdermal route. SLN peculiarity consists in the possibility of acting as a reservoir, permitting a constant and prolonged release of the drugs included. In 7 healthy subjects SLN incorporating MT 3 mg (MT-SLN-O) were orally administered at 8.30 a.m. MT 3 mg in standard formulation (MT-S) was then administered to the same subjects after one week at 8.30 a.m. as controls. In 10 healthy subjects SLN incorporating MT were administered transdermally (MT-SLN-TD) by the application of a patch at 8.30 a.m. for 24 hours. Compared to MT-S, Tmax after MT-SLN-O administration resulted delayed of about 20 minutes, while mean AUC and mean half life of elimination was significantly higher (respectively 169944.7 +/- 64954.4 pg/ml x hour vs. 85148.4 +/- 50642.6 pg/ml x hour, p = 0.018 and 93.1 +/- 37.1 min vs. 48.2 +/- 8.9 min, p = 0.009). MT absorption and elimination after MT-SLN-TD demonstrated to be slow (mean half life of absorption: 5.3 +/- 1.3 hours; mean half life of elimination: 24.6 +/- 12.0 hours), so MT plasma levels above 50 pg/ml were maintained for at least 24 hours. This study demonstrates a significant absorption of MT incorporated in SLN, with detectable plasma level achieved for several hours in particular after transdermal administration. As dosages and concentrations of drugs included in SLN can be varied, different plasma level profile could be obtained, so disclosing new possibilities for sustained delivery systems.  相似文献

In this paper, the lubrication mechanism of fatty acids is revisited with a new approach combining experimental and computational chemistry studies. The lubricating properties of single and mixtures of stearic, oleic and linoleic acids in a synthetic Poly-Alpha-Olefin base oil (PAO4) on iron oxide surface are investigated under mixed boundary regime with temperatures from 50 °C up to 150 °C. Low friction coefficient (about 0.055) with no visible wear is reported in the presence of single stearic acid at high temperature. This lubricating behavior is inhibited in the presence of unsaturated fatty acids highlighting an anti-synergic effect of a saturated/unsaturated mixture, especially at 150 °C. To understand the anti-synergic effect and the adsorption mechanism of these molecules, molecular dynamic (MD) and quantum chemistry simulations are performed to evaluate their diffusion coefficient in PAO4 and their adsorption mechanism on iron oxide at different temperatures. MD simulation results show a faster diffusion toward the surface for unsaturated fatty acids than for saturated fatty acid at all the studied temperatures. This means that unsaturated molecules arrive and mainly adsorb before stearic acid on the surface leading to a tribological behavior of the mixture characteristic of the unsaturated molecule. Computational chemistry suggests that all fatty acids (saturated and unsaturated) adsorption mechanism is due to the chemisorption of the carboxylic group on iron oxide surface with no desorption up to 150 °C.  相似文献
