Shear stress, the dragging force generated by fluid flow, differentially activates extracellular signal-regulated kinase (ERK) and c-Jun NH2-terminal kinase (JNK) in bovine aortic endothelial cells (BAEC) (Jo, H., Sipos, K., Go, Y. M., Law, R., Rong, J., and McDonald, J. M. (1997) J. Biol. Chem. 272, 1395-1401). Here, we examine whether cholesterol-enriched compartments in the plasma membrane are responsible for such differential regulation. Pretreatment of BAEC with a cholesterol-binding antibiotic, filipin, did not inhibit shear-dependent activation of JNK. In contrast, filipin and other membrane-permeable cholesterol-binding agents (digitonin and nystatin), but not the lipid-binding agent xylazine, inhibited shear-dependent activation of ERK. The effect of cholesterol-binding drugs did not appear to be due to membrane permeabilization, since treatment of BAEC with a detergent, Triton X-100 which also permeabilizes membranes, did not inhibit shear-dependent activation of ERK. Furthermore, shear-dependent activation of ERK, but not JNK, was inhibited by cyclodextrin, a membrane-impermeable cholesterol-binding agent, which removes cell-surface cholesterol. Moreover, the effects of cyclodextrin were prevented by adding cholesterol during the incubation. These results indicate that cholesterol or cholesterol-sensitive compartments in the plasma membrane play a selective and essential role in activation of ERK, but not JNK, by shear stress. Although exposure to shear stress (1 h) increased the number of caveolae by 3-fold, treatment with filipin had no effect in either control or shear-exposed cells suggesting that caveolae density per se is not a crucial determinant in shear-dependent ERK activation. In summary, the current study suggests that cholesterol-sensitive microdomains in the plasma membrane, such as caveolae-like domains, play a critical role in differential activation of ERK and JNK by shear stress.  相似文献

Cynipid gall-wasp communities correlate with oak chemistry   总被引:1,自引:0,他引:1  
Host-plant association data, gathered from field surveys conducted throughout Florida and from the literature, were used to identify the specificity of cynipid gall inducers to one or more of six Quercus species that occur at Archbold Biological Station, Lake Placid, Florida, USA, including the red oaks Q. laevis, Q. myrtifolia, and Q. inopina, and the white oaks Q. chapmanii, Q. geminata, and Q. minima. Quercus myrtifolia had the highest cynipid richness and diversity (37 cynipid species, Shannon H = 3.61, Simpson's D = 0.97), followed by Q. chapmanii, Q. laevis, Q. inopina, Q. geminata, and finally Q. minima (10 species, H = 2.30, D = 0.90). All cynipid species showed strong fidelity to a particular host plant or a restricted set of host plants. An ordination of gall-wasp host associations indicated that the cynipid communities of each oak species were distinct and specific to a given oak species. Leaf samples taken from each oak species were analyzed for condensed and hydrolyzable tannins, total phenolics, lignin, cellulose and hemicellulose, nitrogen, and carbon. All of these chemical traits, with the exception of carbon, differed by oak species, and the differences were strongly correlated with the axes of the cynipid--species ordination. These results suggest that gall-wasp occurrence is influenced by oak chemistry and imply that experimental studies of cynipid gall inducers that examine host-plant chemistry and female oviposition choice and larval performance will yield useful insights.  相似文献

A discussion is presented describing the possible influence of the internal surface energy of polycrystalline and pyrolytic graphites on three phenomena: (a) the thermodynamic “standard” state of carbon, (b) the graphitization process, and (c) the structural stability of graphites at high temperatures. In order to do this, the surface enthalpies and free energies of the basal surface and edge faces have been calculated from a number of sources, and their temperature dependence up to 3000K discussed. Suggestions have been made for methods to measure both basal surface and “edge” surface energies more directly.  相似文献

Treatment guidelines for psychotherapy describe a set of best practices based in part on scientific evidence. There have been several sets of such guidelines published recently by governmental, professional, and health services organizations. The adequacy of such guidelines, which perforce reduce the variability of professional services, depends on the adequacy of the scientific evidence with respect to both efficacy and clinical utility. The American Psychological Association formed a Task Force to develop a Template for Developing Guidelines in order to evaluate the scientific evidence for these guidelines. This article describes the Template and then applies it to a set of recently promulgated guidelines. The Template was able to demonstrate the strengths and weaknesses of the various documents. A thick mat of fine fibres and crystallites has been found on the surfaces of graphite and carbon anodes after low current arc operation in nitrogen. Similar operation in air leaves no trace of the fibres. The dimensions and other properties have been studied by scanning and transmission electron microscopy. Radiance decays observed at the anode face of the “standard carbon arc” after rapid current cut-off have been studied over a range of wavelengths. The temperature drop at the surface is too rapid for bulk graphite cooling, as has been pointed out by Lozier and Null. Purely radiative and conductive-radiative cooling mechanisms in surface layers have been considered, and it is found that conduction through the fibre layer and a porous layer beneath the fibres controls the cooling rate for the first 100 μs.  相似文献
