An efficient bioreactor, termed a 'synchronized fresh cell bioreactor', was developed and consisted of a pH-dependent substrate feed system coupled with cross flow filtration and turbidity control. The effect of high dilution rate and high cell density coupled with high cell viability on the production of l-lactic acid in continuous culture by Lactococcus lactis IO-1 in enzyme-hydrolysed sago starch medium was investigated. For all changes in dilution rate, cells responded in a synchronized way to the addition of glucose by increasing the rate of biomass formation. Consequently, a glucose-free feed solution was required to maintain the cell concentration at a particular pre-set value. This set-up facilitated the maintenance of the cells in a permanent log phase. At a cell concentration of 15 gl(-1) and a feed glucose concentration of 53 gl(-1), volumetric LA productivities of 8.2, 19.3 and 33.1 gl(-1)h(-1) were obtained at dilution rates of 0.21, 0.50 and 1.1 h(-1), respectively. The respective residual glucose concentrations in the spent medium were 1.90, 0.24 and 3.80 gl(-1). By increasing the cell density, the volumetric productivity increased proportionally. At high cell density, higher dilution rates resulted in lower lactate concentrations in the culture medium resulting in higher productivity. This reactor facilitated efficient operation with high cell viability by maintaining the cells in continuous growth phase for long-term fermentation. Therefore, the growth rate (mu) was calculated according to the Monod equation. Using this system, high specific productivities can be obtained which guarantees high commercial productivity at economical cost with only a small investment for setting up the sago industry.  相似文献

Transarterial vascular embolization and chemoembolization has become common medical procedures, where partially hydrolyzed poly(vinyl alcohol) (PVA) beads remains as one of the most used embolic agent materials. Although synthetic, PVA cannot be synthesized by direct polymerization and must be obtained by chemical modification of another polymer, usually poly(vinyl acetate) (PVAc). The aim of the present work is to synthesize spherical core‐shell PVAc/PVA particles and study the morphological and molecular modifications during shell formation. The polymer particles where produced in two stages, where first the PVAc core was obtained by suspension polymerization of vinyl acetate (VAc) and then the PVA shell synthesized through hydrolysis. Spherical PVAc particles were successfully produced and isolated using an optimized suspension polymerization process. During the shell formation, it was shown that none of the conditions used affected the overall morphology of the particles although changes in the final size distribution could be observed. However, it was possible to identify the process variables and reaction condition that affect the molecular weight averages and polydispersities of the final copolymer. POLYM. ENG. A mathematical model to predict the size distribution and chemical composition of a cloud of sulfide particles during high-temperature oxidation in a flash reactor is presented. The model incorporates the expansion and further fragmentation of the reacting particles along their trajectories throughout the reaction chamber. A relevant feature of the present formulation is its flexibility to treat a variety of flash reacting systems, such as the flash smelting and flash converting processes. This is accomplished by computing the chemical composition of individual particles and the size distribution and overall composition of the particle cloud in separate modules, which are coupled through a database of particle properties previously stored on disk. The flash converting of solid copper mattes is considered as an example. The model predictions showed good agreement with the experimental data collected in a large laboratory reactor in terms of particle size distribution and sulfur remaining in the population of particles. The cumulative contribution and distribution coefficients are introduced to quantify the relationship between specific particle sizes in the feed and those in the reacted products upon oxidation, the latter of which has practical implications on the amount and chemical composition of dust particles produced during the industrial operation.  相似文献

This study estimated the existing stored carbon (C) and rate of sequestration by vegetation that can potentially serve as a sink for the carbon dioxide emitted from eight geothermal plants in Leyte Geothermal Reservation, Philippines. For the 20,438 ha watershed in the vicinity of the power project, the total C storage is 3.84 Mt C (14.10 Mt CO2) while C sequestration based on biomass change was 47.35 kt C (173.77 kt CO2). Relative to power plant emission, the C stored in the reserve is equivalent to more than 22 years of CO2 emission. Annual C sequestration is 27% of CO2 emission per year. For the next 25 years, two scenarios were projected. Under Scenario I (“Business as Usual”), the forest reserve will be able to store and sequester more than 32 years of CO2 emission from the power plants. Under Scenario II (“Accelerated Reforestation”), the reserve will be able to store and sequester about 34 years of CO2 emission.In addition, the rate of C sequestration based on biomass change in vegetation was recorded to assess the optimum land use that can absorb the carbon dioxide emitted by the power project. These are as follows: tree plantations (10.09 tC/ha/yr)>coconut (4.78 tC/ha/yr)>brushland (4.29 tC/ha/yr)>natural forest (0.92 tC/ha/yr).In terms of cost, the power project operator is spending P1.22 per t CO2 (P4.4 or US$0.12 per tC) for every year of C storage and sequestration. For 25 years, the total cost is P30.40 per tCO2 (P111.5 or US$2.94 per tC) which is comparable to the cost of C offset in other tropical countries.  相似文献

With the spark-erosion method we have produced spheroidal particles of an uranium-molybdenum alloy using pure water as dielectric. The particles were characterized by optical metallography, scanning electron microscopy, energy dispersive spectrometry and X-ray diffraction. Mostly spherical particles of UO2 with a distinctive size distribution with peaks centered at 70 and 10 μm were obtained. The particles have central inclusions of U and Mo compounds.  相似文献

Class 1 myosins (Myo1s) were the first unconventional myosins identified and humans have eight known Myo1 isoforms. The Myo1 family is involved in the regulation of gene expression, cytoskeletal rearrangements, delivery of proteins to the cell surface, cell migration and spreading. Thus, the important role of Myo1s in different biological processes is evident. In this study, we have investigated the effects of pentachloropseudilin (PClP), a reversible and allosteric potent inhibitor of Myo1s, on angiogenesis. We demonstrated that treatment of cells with PClP promoted a decrease in the number of vessels. The observed inhibition of angiogenesis is likely to be related to the inhibition of cell proliferation, migration and adhesion, as well as to alteration of the actin cytoskeleton pattern, as shown on a PClP-treated HUVEC cell line. Moreover, we also demonstrated that PClP treatment partially prevented the delivery of integrins to the plasma membrane. Finally, we showed that PClP caused DNA strand breaks, which are probably repaired during the cell cycle arrest in the G1 phase. Taken together, our results suggest that Myo1s participate directly in the angiogenesis process.  相似文献

Agent-oriented software engineering and software product lines are two promising software engineering techniques. Recent research work has been exploring their integration, namely multi-agent systems product lines (MAS-PLs), to promote reuse and variability management in the context of complex software systems. However, current product derivation approaches do not provide specific mechanisms to deal with MAS-PLs. This is essential because they typically encompass several concerns (e.g., trust, coordination, transaction, state persistence) that are constructed on the basis of heterogeneous technologies (e.g., object-oriented frameworks and platforms). In this paper, we propose the use of multi-level models to support the configuration knowledge specification and automatic product derivation of MAS-PLs. Our approach provides an agent-specific architecture model that uses abstractions and instantiation rules that are relevant to this application domain. In order to evaluate the feasibility and effectiveness of the proposed approach, we have implemented it as an extension of an existing product derivation tool, called GenArch. The approach has also been evaluated through the automatic instantiation of two MAS-PLs, demonstrating its potential and benefits to product derivation and configuration knowledge specification.  相似文献

The mechanical properties of metals with bcc structure, such as niobium and its alloys, have changed significantly with the introduction of heavy interstitial elements. These interstitial elements (nitrogen, for example), present in the alloy, occupy octahedral sites and constitute an elastic dipole of tetragonal symmetry and might produce anelastic relaxation. This article presents the effect of nitrogen on the anelastic properties of Nb-1.0 wt% Zr alloys, measured by means of mechanical spectroscopy using a torsion pendulum. The results showed complex anelastic relaxation structures, which were resolved into their constituent peaks, representing each relaxation process. These processes are due to stress-induced ordering of the interstitial elements around the niobium and zirconium of the alloy.  相似文献
