CD26 has been reported as a marker for colorectal cancer stem cells endowed with tumor-initiating properties and capable of colorectal cancer (CRC) metastasis. In this study, we investigated the functional effect of CD26 on CRC angiogenesis and metastasis, and the potential underlying mechanism. The functional effects of CD26 overexpression or repression were determined by a wound healing experiment, and cell migration and invasion assays in vitro and in mouse models. Differentially expressed genes regulated by CD26 were identified by genome-wide mRNA expression array and validated by quantitative PCR. CD26 functionally regulated CRC cell migration and invasion in vitro and angiogenesis and metastasis in vivo. Genome-wide mRNA expression array and qPCR showed that MMP1 was up-regulated in CD26+ subpopulation, and a subsequent experiment demonstrated the regulatory effect of CD26 on MMP1 in CRC cell lines with CD26 repression or overexpression. Furthermore, overexpression of CAV1 abrogated the CD26-regulated MMP1 induction in CRC cell lines. This study demonstrated the functional roles of CD26 in inducing CRC migration, invasion, angiogenesis and metastasis and identified the potential involvement of MMP1 and CAV1 in such process. CD26 is an attractive therapeutic target for combating tumor progression to improve the prognosis of CRC patients.  相似文献

This paper presents an implementation of a decoupled optimization technique for design of switching regulators using genetic algorithms (GAs). The optimization process entails the selection of component values in a switching regulator, in order to meet the static and dynamic requirements. Although the proposed method inherits characteristics of evolutionary computations that involve randomness, recombination, and survival of the fittest, it does not perform a whole-circuit optimization. Thus, intensive computations, that are usually found in stochastic optimization techniques can be avoided. Similar to many design approaches for power electronics circuits, a regulator is decoupled into two components, namely the power conversion stage (PCS) and the feedback network (FN). The PCS is optimized with the required static characteristics, whilst the FN is optimized with the required static and dynamic behaviors of the whole system. Systematic optimization procedures are described and the technique is illustrated with the design of a buck regulator with overcurrent protection. The predicted results are compared with the published results available in the literature and are verified with experimental measurements  相似文献

A comparative study on the stability and potential of alginate and pectin based beads for production of poultry probiotic cells using MRS medium in repeated batch fermentation was conducted. The bead cores, made of three types of materials, i.e., ca-alginate, ca-pectinate and ca-alginate/pectinate, were compared. The effect of single and double layer coatings using chitosan and core material, respectively, on the bead stability and cell production were also studied. The pectin based beads were found to be more stable than that of the alginate beads and their stability was further improved by coating with chitosan. The cell concentration in pectin based beads was comparable to that in the alginate beads. On the other hand, pectin based beads gave significantly lower cell concentration in the growth medium for the initial fermentation cycles when compared to the alginate beads. In conclusion, pectin was found to be potential encapsulation material for probiotic cell production owing to its stability and favourable microenvironment for cell growth.  相似文献

Railway level crossings are amongst the most complex of road safety control systems, due to the conflicts between road vehicles and rail infrastructure, trains and train operations. Driver behaviour at railway crossings is the major collision factor. The main objective of the present paper was to evaluate the existing conventional warning devices in relation to driver behaviour. The common conventional warning devices in Australia are a stop sign (passive), flashing lights and a half boom-barrier with flashing lights (active). The data were collected using two approaches, namely: field video recordings at selected sites and a driving simulator in a laboratory. This paper describes and compares the driver response results from both the field survey and the driving simulator. The conclusion drawn is that different types of warning systems resulted in varying driver responses at crossings. The results showed that on average driver responses to passive crossings were poor when compared to active ones. The field results were consistent with the simulator results for the existing conventional warning devices and hence they may be used to calibrate the simulator for further evaluation of alternative warning systems.  相似文献

Palm fatty acid distillate (PFAD) is a rich source of vitamin E. As compared to other vegetable oil, PFAD has higher tocotrienol (70–80%) over tocopherol content, which makes it a valuable source for vitamin E extraction. Current vitamin E extraction methods are not sustainable due to the intensive usage of chemical and high operational cost. Hence, the present study investigated for the first time using dry fractionation process as a green and economical pretreatment method for separating solid fraction (stearin) and liquid fraction (olein) in order to concentrate vitamin E from PFAD in olein fraction. We examined the dry fractionation conditions: crystallization ending temperature (36–44 °C), cooling rate (0.3 and 1.5°C min−1), stirring speed (20–125 rpm), and holding time (0–60 min) on the composition of unsaturated and saturated fatty acids as well as vitamin E content in liquid fraction (olein) and solid fraction (stearin) using gas chromatography and high performance liquid chromatography, respectively. In most of these conditions, vitamin E was ultimately higher in olein fraction as compared to stearin fraction, which is correlated with the high degree of unsaturation. Under a cooling rate of 0.3°C min−1, 90 rpm stirring speed, and ending crystallization of 38 °C, the highest vitamin E rich olein fraction was attained with 1479 ± 10.51 ppm in 50 g olein fraction as compared to 1366 ± 7.94 ppm in 500 g of unfractionated PFAD.  相似文献

Recently, there have been considerable interests to immobilize photocatalyst in alginate beads for removing pollutants from water sources. However, the feasibility of using alginate beads in industry largely depends on its long‐term stability during operation. This study investigated the physicochemical stability of alginate/titanium dioxide beads (Alg/TiO2) when exposed to UV irradiation in aqueous environment. The degradation of Alg/TiO2 beads was evident because the diameter and mass of the beads was reduced by 12% and 40%, respectively, after 120 h of irradiation. A substantial amount of TiO2 was leached into the external medium. Consequently, the removal efficiency of model cationic dye was found to reduce after every process cycle. Morphological analysis showed the formation of cavities on the surface of the Alg/TiO2 beads. Interestingly, the blank alginate beads degraded more rapidly than the Alg/TiO2 beads, confirming the UV‐shielding effect of TiO2. Nevertheless, this study reveals the need to improve the UV stability of alginate‐based beads before they can be considered for practical application. © 2017 Wiley Periodicals, Inc. J. Appl. Polym. Oil‐in‐water (O/W) emulsion‐gel systems containing high oil payloads are of increasing interest for food applications because of the reduction in encapsulation cost, consumption frequency or volume of food products. This study shows a facile approach to prepare stable alginate‐based O/W emulsions at high oil loading using a mixture of nonionic surfactants (Tween 80 and Span 20) as a template to form gelled‐emulsions. The synergistic effects of alginate and surfactants on the O/W emulsion properties were evaluated in terms of oil droplet size and emulsion stability. At 2% (w/v) of alginate and 1% (w/v) of surfactants, the size distribution of oil droplets was narrow and monomodal, even at an oil loading of 70% (v/v). The emulsions formed were stable against phase separation. The oil droplet size could be further reduced to below 1 μm using a high‐shear homogenizer. The emulsions formed could be easily molded and gelled into solids of different shapes via ionic gelation. The findings of this study create possible avenues for applications in food industries.  相似文献

Diamond thin films grown on high resistivity, 100 oriented silicon substrates by the hot filament chemical vapor deposition (HFCVD) method have been characterized by four-point probe and current-voltage (through film) techniques. The resistivities of the as-grown, chemically etched and annealed samples lie in the range of 102 Ω cm to 108 Ω cm. The Raman measurements on these samples indicate sp3 bonding with a sharp peak at 1332 cm−1. The surface morphology as determined by scanning electron microscope shows polycrystalline films with (100) or (111) faceted structures with average grain size of ≈2.5 μm. The through film current-voltage characteristics obtained via indium contacts on these diamond films showed either rectifying or ohmic behavior. The difference in Schottky and ohmic behavior is explained on the basis of the high or low sheet resistivities measured by four-point probe technique. 5% methane to hydrogen concentration during film growth resulted in poor surface morphology, absence of sp3 bonds, and low resistivity.  相似文献

The application of electron beam irradiation in polymers blend with natural rubber such as thermoplastic polymers, specialty polymers, and thermoset polymers are discussed in this paper for the aspects of physical, mechanical, and thermal properties. The interaction of additives and irradiation for natural rubber blends were also been analyzed. The addition of cross-linking agent such as trimetylolpropane trimethacrylate with appropriate amount could enhance the mechanical properties of the polymer–natural rubber blend and lower the optimum dose of electron beam irradiation. In short, electron beam is another potential aspect worth for study in natural rubber research area.  相似文献
