This study is based on a theoretical and photoelastic static contact stress analysis, relevant to the design of the plastic tibial plateau in an orthopaedic knee prosthesis. It involves the review of existing theoretical analyses concerned with the contact mechanics of layered elastic systems and, where possible, the validation of any relevant findings using two dimensional photoelasticity. Significant findings include the identification of a general minimum recommendable design thickness for the plastic tibial plateau, the endorsement of using the Hertzian contact theory to predict contact stress levels in the tibial plateau of certain knee implants and the slight improvement in contact conditions in the plastic when the plateau is perfectly bonded to its supporting metal tray.  相似文献

Previous analyses of the Wechsler Memory Scale-Revised (WMS-R; Wechsler, 1987) have reported one-, two-, and three-factor solutions and raised questions about the validity of the visual memory subtests. These various findings may stem in part from different methods of analysis, and from the study of different participant samples. To address these issues, we analysed data from the WMS-R and a spatial maze test administered to 154 participants with a history of alcohol dependence. Results from confirmatory factor analysis supported the interpretation of three factors underlying the WMS-R subtests and the spatial maze score in this sample, namely, attention-concentration, immediate memory, and delayed recall. This result held despite the inclusion of the maze score which is a well-validated measure of visuo-spatial memory.  相似文献

Lipid droplets (LDs) have traditionally been thought of as solely lipid storage compartments for cells; however, in the last decade, they have emerged as critical organelles in health and disease. LDs are highly dynamic within cells, and their movement is critical in organelle–organelle interactions. Their dynamics are known to change during cellular stress or nutrient deprivation; however, their movement during pathogen infections, especially at very early timepoints, is under-researched. This study aimed to track LD dynamics in vitro, in an astrocytic model of infection. Cells were either stimulated with a dsRNA viral mimic, poly I:C, or infected with the RNA virus, Zika virus. Individual LDs within infected cells were analysed to determine displacement and speed, and average LD characteristics for multiple individual cells calculated. Both LD displacement and mean speed were significantly enhanced in stimulated cells over a time course of infection with an increase seen as early as 2 h post-infection. With the emerging role for LDs during innate host responses, understanding their dynamics is critical to elucidate how these organelles influence the outcome of viral infection.  相似文献

The forward scattering of light in a conventional inverted optical microscope by nanoparticles ranging in diameter from 10 to 50nm has been used to automatically and quantitatively identify and track their location in three-dimensions with a temporal resolution of 200ms. The standard deviation of the location of nominally stationary 50-nm-diameter nanoparticles was found to be about 50nm along the light path and about 5nm in the plane perpendicular to the light path. The method is based on oscillating the microscope objective along the light path using a piezo actuator and acquiring images with the condenser aperture closed to a minimum to enhance the effects of diffraction. Data processing in the time and spatial domains allowed the location of particles to be obtained automatically so that the technique has potential applications both in the processing of nanoparticles and in their use in a variety of fields including nanobiotechnology, pharmaceuticals and food processing where a simple optical microscope maybe preferred for a variety of reasons.  相似文献

This paper describes the Si-doping of GaAs that was grown using the AsCl3:H2:GaAs, Ga Chemical vapor deposition process. The doping sources were AsCl3:SiCl4 liquid solutions which proved to be highly reproducible for Si doping within the range, 1×1O16 to 2×1019 cm?3. Incorporation of Si into the GaAs apparently occurs under near equilibrium conditions. This point is considered in detail and the consequences experimentally utilized to grow n, n+ bilayers using a single AsCl3:SiCl4 doping solution. Si impurity profiles based upon differential capacitance and SIMS data are presented. These can be very abrupt for n, n+ structures with order of magnitude changes occurring within 500 Å. For the 1×1016 to 8×l018 cm?3 doped samples the mobilities at 78 and 298°K are comparable to the higher values reported for GaAs thin films grown by CVD. Power FET devices made from this material have demonstrated an output density of 0.86 watts/mm at 10 GHz.  相似文献

Although rare, inherited retinal degenerations (IRDs) are the most common reason for blind registration in the working age population. They are highly genetically heterogeneous (>300 known genetic loci), and confirmation of a molecular diagnosis is a prerequisite for many therapeutic clinical trials and approved treatments. First-tier genetic testing of IRDs with panel-based next-generation sequencing (pNGS) has a diagnostic yield of ≈70–80%, leaving the remaining more challenging cases to be resolved by second-tier testing methods. This study describes the phenotypic reassessment of patients with a negative result from first-tier pNGS and the rationale, outcomes, and cost of second-tier genetic testing approaches. Removing non-IRD cases from consideration and utilizing case-appropriate second-tier genetic testing techniques, we genetically resolved 56% of previously unresolved pedigrees, bringing the overall resolve rate to 92% (388/423). At present, pNGS remains the most cost-effective first-tier approach for the molecular assessment of diverse IRD populations Second-tier genetic testing should be guided by clinical (i.e., reassessment, multimodal imaging, electrophysiology), and genetic (i.e., single alleles in autosomal recessive disease) indications to achieve a genetic diagnosis in the most cost-effective manner.  相似文献

Poly(vinylalcohol) (PVA)-stabilized silver nanoparticles were prepared by a seeding method. Nanoparticles of varying morphology were obtained by controlled variation of the reaction conditions, and this method allowed the tailoring of the position of the surface plasmon resonance. The samples show two bands in the visible absorption spectrum: one in the 410-440-nm region and a second peak between 500 and 600 nm. This tunable surface plasmon resonance serves to increase the third-order optical nonlinearity (chi 3) of the nanoparticles (measured at 532 nm) by a factor of 16.  相似文献

The chemistry and microstructure of iron-base alloys resistant to galling wear were determined by using optical microscopy and scanning electron microscopy (SEM). Castings and weld overlays, deposited by the gas tungsten arc and plasma arc welding (GTAW and PAW, respectively) processes, were evaluated. The microstructure typically consisted of a primary austenitic matrix, eutectic carbides (M7C3 type), and noneutectic carbides. Processing techniques that resulted in high cooling rates yielded microstructures with finer features, less complete partitioning of alloying elements to the carbides, and improved resistance to galling wear. Carbon and manganese appeared to improve resistance to galling wear. Nickel was detrimental to galling wear resistance. This paper presents micro-interferometry as a measurement technique to extract temperature profiles and/or mass transfer gradients rapidly and locally in micro-devices. Interferometry quantifies the phase change between two or more coherent light beams induced by temperature and/or mass concentration. Previous work has shown that temporal noise is a limiting factor in microscale applications. This paper examines phase stepping and heterodyne phase retrieval techniques with both CCD and CMOS cameras. CMOS cameras are examined owing to the high speed at which images can be acquired which is particularly relevant to heterodyne methods. It is found that heterodyne retrieval is five times better than phase stepping being limited to 0.01 rad or λ/628. This is twice the theoretical limit of λ/1,000. The technique is demonstrated for mixing in a T-junction with a 500 μm square channel and compared favourably to a theoretical prediction from the literature. Further issues regarding application to temperature measurements are discussed.  相似文献
