Treatment of primary cultured rat hepatocytes with hepatocyte growth factor (HGF) gives rise to inositol phosphate formation, cytosolic calcium oscillation, activation of mitogen-activated protein (MAP) kinase and phospholipase D (PLD), and arachidonic acid release, leading to DNA synthesis. Pretreatment of cultured hepatocytes with pertussis toxin (PT), which is known to adenosine diphosphate-ribosylate Gi and Go guanine nucleotide -binding proteins and to inhibit their functions, partially inhibited HGF-induced [3H]thymidine incorporation in a concentration-dependent manner. These results suggest that HGF-mediated DNA synthesis of hepatocytes is partly regulated via PT-sensitive guanine nucleotide-binding protein. Therefore, the effects of PT treatment on HGF-induced signal-transduction pathways were investigated. HGF-induced MAP kinase activation and arachidonic acid release were decreased by PT treatment, whereas PLD activation was diminished by PT to the level of unstimulated control. PT also interfered with HGF-induced inositol phosphate formation and cytosolic calcium oscillation. These results suggest that both PT-sensitive and PT-insensitive pathways are involved in HGF-induced signaling.  相似文献

The association of cytoskeletons with the Golgi apparatus (GA) in cultured 3Y1 cells was investigated by stereo electron microscopy of thick sections and computer-graphic reconstruction of serial thin sections. The 3-dimensional analysis has demonstrated that: 1) both microtubules (MT) and vimentin intermediate filaments (IF) were abundantly present in close vicinity to GA, and some of them were closely associated with GA; and 2) such GA-associated cytoskeletons were attached to GA either at their termini or at their lateral side. The present observations suggest that both MT and vimentin IF are responsible for maintaining the structural integrity of GA.  相似文献

Erythrocyte-granulocyte rosettes (EGR) were found in the capillary blood of two cases of autoimmune haemolytic anemia, the first caused by an IgM non-I antibody and the second by an IgG. Both antibodies activated complement, and C3b was demonstrated on the erythrocytes. The rosettes appeared as arrangements of 6-10 red cells adhering to a central granulocyte; phagocytosis was most generally absent. The finding of EGR in the capillary blood may be useful for the recognition of complement activation up to the C3b stage in some cases of autoimmune haemolytic anemia.  相似文献

The surface melting and crystallization behavior of two biodegradable polyesters, poly(3-hydroxybutyrate-co-3-hydroxyhexanoate) (P(HB-co-HHx)) (HHx=12 mol%) and poly(3-hydroxybutyrate) (PHB), were investigated by using attenuated total reflection (ATR) and transmission infrared (IR) spectroscopy coupled with the generalized two-dimensional (2D) correlation analysis. IR bands in the CO stretching vibration region were analyzed to explore the changes in crystallinity at the surface and those in the bulk of the polymer film samples during the melting and crystallization processes. Due to the intrinsic sub-micrometer sampling depth of the ATR technique, spectral information attributed to the chemical moieties situated at the surface region of the film samples can be observed. The present study revealed that the surface melting of P(HB-co-HHx) takes place through an intermediate state. The distribution of crystalline phase detected by the ATR technique and that detected by the transmission technique suggested that the polymer crystals tend to grow at the surface in a manner different from that in the bulk. It is very likely that the population of polymer crystals at the surface is higher than that in the bulk for both P(HB-co-HHx) and PHB. The time-dependent IR spectral variations for P(HB-co-HHx) and those for PHB indicated that P(HB-co-HHx) crystallizes much slower than PHB. This observation suggested that the HHx units incorporated in P(HB-co-HHx) markedly reduce not only the degree of crystallinity but also the crystallization rate of PHB homopolymer.  相似文献

C2-Symmetric 9,9′-spirobifluorene-containing polyesters (PEs) were synthesized by polycondensation of 2,2′-dihydroxy-9,9′-spirobifluorene (1) with bis(acyl chloride)s (2) at 230 °C in diphenylether. The molecular weights of PEs 3a-3f were sufficiently high (Mw 13,400-41,600). PEs displayed high thermal stability. The glass transition temperatures (Tg) estimated by differential scanning calorimetry analysis appeared in a range 177-352 °C depending on the spacer structure, while the 5% decomposition temperatures (Td5) measured by thermogravimetric analysis were over 416 °C both under nitrogen atmosphere and in air. PEs showed good solubility in typical organic solvents such as CHCl3 and THF easily to afford the tough, transparent, and flexible cast films. The transmittance of the polymer films reached over 90% in the wavelength range from ca. 410-900 nm. In addition, PEs exhibited higher refractive index rather than that of commercially available 9,9-diarylfluorene-containing PE, in addition to very low degree of birefringence presumably due to the C2-symmetric structure.  相似文献

The techniques of pre- and post-embedding immunocolloidal gold electron microscopy were tested for their usefulness for analyzing the morphogenesis of human immunodeficiency virus (HIV) strain LAV employing U-937 cells persistently infected with the virus (U-937/LAV). By both techniques the following results were obtained. (1) Budding process was restricted to a localized area at the membrane adjacent to the Golgi apparatus. (2) The distribution of viral envelope antigens was restricted to the electron-dense crescent structures on the cell surface. (3) Virions released from the cells could be classified mainly into 2 categories: virions with doughnut-shaped cores or with conical cores at the center of the particles. And p24 proteins localized on the thick electron-dense cores of both types. These findings support the idea that pre- and post-embedding immunogold electron microscopy is very useful for studying the morphogenesis of viruses.  相似文献
