Mechanical properties of miscible blends of high molecular weight poly(N-vinyl pyrrolidone) (PVP) with a short-chain, liquid poly(ethylene glycol) (PEG) of molecular weight 400 g/mol have been examined as a function of PVP-PEG composition and degree of hydration. The small-strain behavior in the linear elastic region has been evaluated with the dynamic mechanical analysis and compared with the viscoelastic behavior of PVP-PEG blends under large strains in the course of uniaxial drawing to fracture and under cyclic extension. A strong decoupling between the small-strain and the large strain properties of the blends has been observed, indicative of a pronounced deviation from rubber elasticity in the behavior of the blends. This deviation, also seen on tensile tests under fast drawing, is attributed to the peculiar phase behavior of the blends and the molecular mechanism of PVP-PEG interaction. Nevertheless, for the PVP blend with 36% PEG, under comparatively low extension rates, the reversible contribution to the total work of deformation up to ε=300% has been found to be maximum at around 70%, while the blends containing 31 and 41% PEG behave rather as an elastic-plastic solid and a viscoelastic liquid, respectively.  相似文献

Nine-analyte detection using an array-based biosensor   总被引:4,自引:0,他引:4  
A fluorescence-based multianalyte immunosensor has been developed for simultaneous analysis of multiple samples. While the standard 6 x 6 format of the array sensor has been used to analyze six samples for six different analytes, this same format has the potential to allow a single sample to be tested for 36 different agents. The method described herein demonstrates proof of principle that the number of analytes detectable using a single array can be increased simply by using complementary mixtures of capture and tracer antibodies. Mixtures were optimized to allow detection of closely related analytes without significant cross-reactivity. Following this facile modification of patterning and assay procedures, the following nine targets could be detected in a single 3 x 3 array: Staphylococcal enterotoxin B, ricin, cholera toxin, Bacillus anthracis Sterne, Bacillus globigii, Francisella tularensis LVS, Yersiniapestis F1 antigen, MS2 coliphage, and Salmonella typhimurium. This work maximizes the efficiency and utility of the described array technology, increasing only reagent usage and cost; production and fabrication costs are not affected.  相似文献

Spontaneous cleavage of the less abundant form of tobacco ringspot virus satellite RNA is readily reversible. Capitalizing on earlier observations by Feldstein and Bruening that small 'mini-monomer' RNAs derived from this molecule and containing little more than covalently attached ribozyme and substrate cleavage products are able to efficiently circularize, we have constructed a series of self-circularizing RNAs of precisely known size. Mixtures of linear and circular RNAs synthesized in vitro and containing 225-1132 nt could be completely resolved using a novel two-dimensional denaturing polyacrylamide gel electrophoresis system. Similar analyses of a complex mixture of coconut cadang-cadang viroid RNAs revealed the presence of relatively large amounts of a previously undescribed 'fast-slow' heterodimeric RNA species in infected palms. Only a single DNA template is required to prepare each pair of circular and linear RNA markers.  相似文献

Mechanisms of molecular interaction in the blends of a polybase, a copolymer of N,N‐dimethylaminoethylmethacrylate with methylmethacrylate and butylmethacrylate (PDMAEMA–MMA/BMA), with a polyacid, a copolymer of methacrylic acid with ethylacrylate (PMAA‐co‐EA), and plasticizer, triethylcitrate (TEC), have been investigated with FTIR Spectroscopy and potentiometry. To evaluate the strengths of hydrogen and ionic bonds in the polyelectrolyte complexes, quantum‐chemical calculations were performed. According to this analysis, the energy of ionic and hydrogen bonding diminishes in the order: multi‐component complexes involving protonated aminogroup of DMAEMA (ammonium cation) in the presence of chlorine counterion with ionized or unchanged carboxyl groups and water molecules (690–520 kJ/mol) > ternary H‐bonded acid‐base complexes associated with molecule of water (520–420 kJ/mol) > binary ionic complex of carboxylate anion and ammonium cation (404 kJ/mol) > H‐bonded complex of carboxylate and ammonium ions (257 kJ/mol) > binary H‐bonded complex of uncharged carboxyl group with ammonium cation (114 kJ/mol) > ternary H‐bonded complex of uncharged carboxyl group, aminogroup and water molecule (43 kJ/mol) > binary H‐bonded complex between nonionized carboxyl and amino groups (26 kJ/mol). Proton‐donating capability of functional groups in the studied polyelectrolyte blends diminishes in the order: HN+(CH3)2 ? > HOOC? > HO? . The proton‐donating capacity can be significantly improved in the presence of Cl? ions, the effect of which may be appreciably inhibited if Na+ cations are available in the blend or solution. Proton‐accepting capability weakens in the order: uncharged aminogroup > carboxylate anion > uncharged carboxyl group > hydroxyl group. This study evaluated the validity of the Desired Effects of Drinking Scale (DEoDS) with a late adolescent sample. This brief measure (37 items) was designed to assess reasons for drinking across 9 domains. Across a culturally diverse sample of late adolescents (ages 18-20 years), this measure evidenced high internal consistency, particularly when all items were summed in 1 full-scale score. Contrary to the factors found with adults, 8 factors emerged with this sample. No differences were found by culture, providing initial evidence for the cross-cultural validity of this measure. In addition, gender differences emerged for only 1 factor. Overall, the results provide empirical support for the use of this measure to assess late adolescents' and emerging adults' reasons for drinking. The effect of respiratory constraint on heart rate control was assessed in a biofeedback situation with feedback consisting of changes in both illumination level and intensity of pre-recorded baby's cry. Subjects were reinforced for alternately increasing and decreasing heart rate on each of seven days during which respiration was unconstrained (Phase 1). This phase was followed by eight days when respiration was constrained during training sessions with a control respirator (Phase 2). Seven additional days of training followed in the unconstrained situation (Phase 3). Results indicate that the control of heart rate in biofeedback situations is very closely related to respiratory and other somatic activity. The implication of these findings for the field of visceral control is discussed.  相似文献
