Antidotes against organophosphates often possess physicochemical properties that mitigate their passage across the blood–brain barrier. Cucurbit[7]urils may be successfully used as a drug delivery system for bisquaternary oximes and improve central nervous system targeting. The main aim of these studies was to elucidate the relationship between cucurbit[7]uril, oxime K027, atropine, and paraoxon to define potential risks or advantages of this delivery system in a complex in vivo system. For this reason, in silico (molecular docking combined with umbrella sampling simulation) and in vivo (UHPLC—pharmacokinetics, toxicokinetics; acetylcholinesterase reactivation and functional observatory battery) methods were used. Based on our results, cucurbit[7]urils affect multiple factors in organophosphates poisoning and its therapy by (i) scavenging paraoxon and preventing free fraction of this toxin from entering the brain, (ii) enhancing the availability of atropine in the central nervous system and by (iii) increasing oxime passage into the brain. In conclusion, using cucurbit[7]urils with oximes might positively impact the overall treatment effectiveness and the benefits can outweigh the potential risks.  相似文献

Figure-ground assignment is an important visual process; humans recognize, attend to, and act on figures, not backgrounds. There are many visual cues for figure-ground assignment. A new cue to figure-ground assignment, called lower region, is presented: Regions in the lower portion of a stimulus array appear more figurelike than regions in the upper portion of the display. This phenomenon was explored, and it was demonstrated that the lower-region preference is not influenced by contrast, eye movements, or voluntary spatial attention. It was found that the lower region is defined relative to the stimulus display, linking the lower-region preference to pictorial depth perception cues. An important issue in attention research concerns the representational format from which attention selects. S. P. Vecera and M. J. Farah (see record 1994-32352-001) presented results that they argued demonstrated attentional selection from a spatially invariant object representation. In their comment, A. F. Kramer, T. A. Weber, and S. E. Watson (see record 84-18014) questioned the interpretation of these results, and they presented evidence consistent with selection from a grouped location-based representation. In this reply, the author argues that although an absence of spatial, or distance, effects may be ambiguous as to whether attention is selecting from an object-based representation or from a location-based representation, there are computational considerations that favor object-based selection in certain tasks. The author concludes with a discussion of how object-based and location-based representations might interact with one another, thereby providing a possible explanation of Kramer et al.'s results. Does spatial attention follow a full preattentive analysis of the visual field, or can attention select from ungrouped regions of the visual field? We addressed this question by testing an apperceptive agnosic patient, J. W., in tasks involving both spatial selection and preattentive grouping. Results suggest that J. W. had intact spatial attention: He was faster to detect targets appearing at cued locations relative to targets appearing at uncued locations. However, his preattentive processes were severely disrupted. Gestalt grouping and symmetry perception, both thought to involve preattentive processes, were impaired in J. W. Also, he could not use gestalt grouping cues to guide spatial attention. These results suggest that spatial attention is not completely dependent on preattentive grouping processes. We argue that preattentive grouping processes and spatial attention may mutually constrain one another in guiding the attentional selection of visual stimuli but that these 2 processes are isolated from one another. Traditional bottom-up models of visual processing assume that figure-ground organization precedes object recognition. This assumption seems logically necessary: How can object recognition occur before a region is labeled as figure? However, some behavioral studies find that familiar regions are more likely to be labeled figure than less familiar regions, a problematic finding for bottom-up models. An interactive account is proposed in which figure-ground processes receive top-down input from object representations in a hierarchical system. A graded, interactive computational model is presented that accounts for behavioral results in which familiarity effects are found. Covert recognition of faces in prosopagnosia, in which patients cannot overtly recognize faces but nevertheless manifest recognition when tested in certain indirect ways, has been interpreted as the functioning of an intact visual face recognition system deprived of access to other brain systems necessary for consciousness. The authors propose an alternative hypothesis: that the visual face recognition system is damaged but not obliterated in these patients and that damaged neural networks will manifest their residual knowledge in just the kinds of tasks used to measure covert recognition. To test this, a simple model of face recognition is lesioned in the parts of the model corresponding to visual processing. The model demonstrates covert recognition in 3 qualitatively different tasks. Implications for the nature of prosopagnosia, and for other types of dissociations between conscious and unconscious perception, are discussed. Older adults are overrepresented in fatal crashes on a per-mile basis. Those with useful field of view (UFOV) reductions show a particularly elevated crash risk that might be mitigated with vehicle-based warnings. To evaluate cross-modal cues that could be used in these warnings, we applied a variation of Posner's orienting of attention paradigm. Twenty-nine older drivers with UFOV impairments and 32 older drivers without impairments participated. Cues were presented in either a single modality or a combination of modalities (visual, auditory, haptic). Drivers experienced three cue types (valid spatial information, invalid spatial information, neutral) and an uncued baseline. Following each cue, drivers discriminated the direction of a target (a Landolt square with a gap facing up or down) in the visual panorama. Drivers with and without UFOV impairments showed comparable response times (RTs) across the different cue modalities and cue types. Both groups benefited most from auditory and auditory/haptic cues. Redundant visual cues, when paired with auditory cues, undermined performance rather than enhanced it. Overall, drivers responded faster to targets with valid spatial information followed by neutral, invalid, and uncued targets. Cues provide the greatest benefit in alerting rather than orienting the driver. The cue expected to be most effective at orienting attention - the extra-vehicular cue - performs most poorly when the spatial information is either invalid or neutral. Even when the spatial information is valid the extra-vehicular cue underperforms the auditory cues. The results suggest that temporal information dominates spatial information in the ability of cues to speed responses to targets. This study represents a first step in assessing whether combining a cognitive science paradigm and a driving simulator environment can quickly assess how different warning signals alert and orient drivers.  相似文献

A new type of aerosol collector employing a liquid at laboratory temperature for continuous sampling of atmospheric particles is described. The collector operates on the principle of a Venturi scrubber. Sampled air flows at high linear velocity through two Venturi nozzles "atomizing" the liquid to form two jets of a polydisperse aerosol of fine droplets situated against each other. Counterflow jets of droplets collide, and within this process, the aerosol particles are captured into dispersed liquid. Under optimum conditions (air flow rate of 5 L/min and water flow rate of 2 mL/min), aerosol particles down to 0.3 microm in diameter are quantitatively collected in the collector into deionized water while the collection efficiency of smaller particles decreases. There is very little loss of fine aerosol within the aerosol counterflow two-jets unit (ACTJU). Coupling of the aerosol collector with an annular diffusion denuder located upstream of the collector ensures an artifact-free sampling of atmospheric aerosols. Operation of the ACTJU in combination with on-line detection devices allows in situ automated analysis of water-soluble aerosol species (e.g., NO2-, NO3-)with high time resolution (as high as 1 s). Under the optimum conditions, the limit of detection for particulate nitrite and nitrate is 28 and 77 ng/m(3), respectively. The instrument is sufficiently rugged for its application at routine monitoring of aerosol composition in the real time.  相似文献

The study is primarily focused on the possibility to utilize organic asymmetric flow field‐flow fractionation (A4F) coupled to a multi‐angle light scattering (MALS) detector for the characterization of copolymers of methyl methacrylate with various acrylates prepared by emulsion polymerization. The effects of acrylate monomer type and content on the molar mass distribution and degree of branching of acrylic copolymers have been studied by A4F‐MALS using tetrahydrofuran as a carrier solvent. It has been found that the growing amount of acrylate results in the increase of molar mass, polydispersity, and branching degree as a result of chain transfer to polymer. Highly branched compact macromolecules with ultra‐high molar mass were identified in all copolymers containing a high level of acrylate. In contrast to size traditionally used exclusion chromatography, organic A4F‐MALS has been proved as a very efficient technique for the characterization of high molar mass acrylic emulsion copolymers. © 2014 Wiley Periodicals, Inc.
