Wind turbines are being designed in accordance with deterministic design rules. As wind turbines increase in size and power, these rules may not be adequate to ascertain a safe and well balanced design. Wind turbine industry can take advantage of the safety and reliability practices in e.g. aerospace, nuclear and offshore industry. These practices include System Reliability (such as FMECA, Event Sequence Analysis and Fault Tree Analysis) and Structural Reliability methods. The approach to introduce Probabilistic Safety Assessment (PSA) which incorporates these reliability methods into wind turbine engineering will be described. By means of two recent case studies, the applicability, benefits, and limitations of these methods will be illustrated.  相似文献

Small-cell variants of Sézary syndrome and mycosis fungoides (MF) have been described. However, in these studies the nuclear area of the small-cell variant of MF (SC-MF) as compared to histological classical MF (CL-MF) was not characterized objectively by quantitative electron microscopy. In a 14-year follow-up period, of a total of 76 patch/plaque stage MF patients seen in the Department of Dermatology of the University Hospital Utrecht, 14 (18%) had an infiltrate composed of atypical lymphocytes characterized by a distinctly smaller cell diameter and smaller, hyperchromatic, deeply indented nuclei as compared to the usual cell type of MF. The aim of the investigation was to confirm this observation objectively using quantitative electron microscopy (morphometry) and to define SC-MF as compared to CL-MF. The study was performed on the 14 patients with SC-MF, and 10 patients with clinical and histological CL-MF and 4 patients with chronic eczema. Electron micrographs of sections obtained from each biopsy were analysed by computer to produce the following data: a nuclear contour index (NCI), the mean nuclear area (MNA), the mean nuclear area of the cells above the 75th percentile (P75NA) and the percentage of cells larger than 30 microm2. The values of MNA differed significantly between patients with SC-MF and those with CL-MF (17.6 vs 23.2 microm2; P = 0.02), as did the values of P75NA (20.7 vs 27.9 microm2; P = 0.01). The NCI of the SC-MF and CL-MF patients were similar. These results are consistent with our observations that SC-MF does indeed exist.  相似文献

Myocardial tagging with magnetic resonance (MR) imaging offers unique possibilities for noninvasive left ventricular (LV) strain analysis. True three-dimensional strain analysis can be achieved with tags implemented in cardiac short axis and long axis images. Spin-echo (SE) techniques have been used for these studies. However, this approach is time-consuming: images at different phases of the cardiac cycle have to be obtained in successive measurements and hence the total number of measurements equals the number of time frames. Moreover, the images are often degraded by flow and motion artifacts. The purpose of this study was to optimize a faster and more robust MR tagging sequence for use on a clinical whole-body 1 T MR system with optimal persistence of the tags during the entire cardiac cycle. The tagging pulses were implemented in gradient-recalled-echo (GRE) sequences and compared to SE-based acquisitions. The effects of the use of flow-compensating gradients, the excitation angles, and the angles of the saturation pulses have been studied with MR signal simulations and in comparative measurements in volunteers. GRE acquisitions with flow-compensating gradients are robust techniques for myocardial tagging acquisitions. Use of optimized flip angles and saturation pulses can significantly improve delineation of the tag and can be used up to at least 700 ms after the R-wave. Therefore, LV tagging with GRE acquisitions using optimized MR parameters is a robust and promising technique.  相似文献

After retrograde filling of axons terminating in the glandular lobe of the corpus cardiacum (CC) of Locusta migratoria with cobalt chloride, a paired group of about 15 cobalt containing cells was demonstrated in the lateral area of the protocerebrum. The axons of these cells run via the NCC II into the glandular lobe of the CC. These small neurons have the characteristics of secretory cells; they contain secretory granules of about 1000 A in diameter. The axon terminals in the glandular lobe, making synaptic contacts with the glandular cells, contain secretory granules of the same size. It is therefore concluded that the cell groups in the protocerebrum control the activity of the glandular cells which produce an adipokinetic hormone. Arborizations of fibers of the lateral secretomotor cells are present in the dorsal neuropile of the protocerebrum, ventral of the mushroom bodies and along the tracts of the NCC I within the brain. It is proposed that these arborizations are sites of synaptic input. It is discussed that the axons of these cells might receive additional synaptic input in the storage lobe of the CC. The localization of cell bodies, the axons of which enter the storage part of the CC is described. The course of the axon tracts of the various cell groups in the protocerebrum and their connections with the NCC I and NCC II are demonstrated.  相似文献

Ferritic chromium-molybdenum steels with chromium contents of 1 wt% up to 12 wt% have been exposed for 8370 h to flowing sodium at 550°C. The oxygen content of the sodium was 6–7 ppm by weight. Weight measurements, carbon analyses and metallographic examinations were carried out. The low chromium steels show weight loss and decarburisation. The high chromium steels show weight gain and carburisation. The crossover point is at about 5 wt% Cr. The composition at the utmost surface (<10 μm) of the various steels tends to about 8 wt% chromium, about 2 wt% nickel and 0.02–0.09 wt% carbon. Sodium chromite crystals were present on the steels with a chromium content of 5 wt% or more. At the exposed surface of the 214 wt% chromium steel sodium chromite crystals were found locally.  相似文献

The cellular composition of the dark and light zones of germinal centres in human tonsils was quantitatively determined by electron microscopy. In addition to the well known germinal-centre B-cells, we defined the cleaved blast as a new distinct cell type in the germinal centre. The dark and the light zones clearly differed in their content of lymphoid and non-lymphoid germinal-centre cells. The dark zone was characterized by higher frequencies of cleaved blasts and small centroblasts, a higher blast-centrocyte ratio and a higher incidence of mitotic figures. In contrast, the light zone had higher frequencies of centrocytes, centroplasmacytoid cells, lymphocytes and follicular dendritic cells (FDC) and an inverted blast-centrocyte ratio. Seven distinct appearances of FDC (FDC.1-FDC.7) could be recognized on the basis of their ultrastructure. The distribution pattern of these subtypes differed between the dark and light zone. The undifferentiated subtypes FDC.2 and FDC.3 predominated in the dark zone. In contrast, the highly differentiated subtypes FDC.4 and FDC.5 were present at a much higher density in the light zone. These findings suggest that the dark and light zones are different compartments with regard to proliferation and differentiation of germinal-centre B-cells and that both compartments have a specific microenvironment influenced by FDC.  相似文献

To evaluate the reproducibility of measurements of left ventricular (LV) dimensions, function, and myocardial mass, segmentedk-space gradient-recalled-echo (GRE) magnetic resonance (MR) imaging was performed on two occasions on 12 healthy volunteers. To compare the MR data, all volunteers underwent a two-dimensional echocardiography with determination of LV dimensions and function. The left ventricle was imaged during breath-hold by consecutive, contiguous short-axis views at end-diastole and end-systole. An average of eight short-axis views was needed to encompass the whole left ventricle. This fast MR sequence limited the total acquisition time to 12 min. LV volumes and masses were calculated after manual delineation of epicardial and endocardial surfaces by two observers in a blinded fashion. Interstudy variability varied between 4.1% and 10.3% for LV end-diastolic volume and end-systolic volume, respectively. Differences in interobserver variability were smaller and varied between 3.6% and 7.3% for LV ejection fraction and end-diastolic volume, respectively. Intraobserver variabilities ranged between 2.0% and 7.0% for LV ejection fraction and end-systolic volume, respectively. These variability percentages agree very well with other studies in literature using other MR sequences. No significant differences in LV dimensions or function were found between MR imaging and echocardiography. In conclusion, this MR sequence allows fast and reproducible LV quantification.  相似文献

Rheumatoid arthritis (RA) is a chronic joint disease characterized by leukocyte invasion and synoviocyte activation followed by cartilage and bone destruction. Its etiology and pathogenesis are poorly understood. We describe a spontaneous mouse model of this syndrome, generated fortuitously by crossing a T cell receptor (TCR) transgenic line with the NOD strain. All offspring develop a joint disease highly reminiscent of RA in man. The trigger for the murine disorder is chance recognition of a NOD-derived major histocompatibility complex (MHC) class II molecule by the transgenic TCR; progression to arthritis involves CD4+ T, B, and probably myeloid cells. Thus, a joint-specific disease need not arise from response to a joint-specific antigen but can be precipitated by a breakdown in general mechanisms of self-tolerance resulting in systemic self-reactivity. We suggest that human RA develops by an analogous mechanism.  相似文献
