Cathepsin D (CathD) is overexpressed and secreted by several solid tumors and stimulates their growth, the mechanism of which is still not understood. In this context, the pepstatin bioconjugate JMV4463 [Ac‐arg‐O2Oc‐(Val)3‐Sta‐Ala‐Sta‐(AMPA)4‐NH2; O2Oc=8‐amino‐3,6‐dioxaoctanoyl, Sta=statine, AMPA=ortho‐aminomethylphenylacetyl], containing a new kind of cell‐penetrating vector, was previously shown to exhibit potent antiproliferative effects in vitro and to delay the onset of tumors in vivo. In this study, we performed a structure–activity relationship analysis to evaluate the significance of the inhibitor and vector moieties of JMV4463. By modifying both statine residues of pepstatin we found that the antiproliferative activity is correlated with CathD inhibition, supporting a major role of the catalytic activity of intracellular CathD in cancer cell proliferation. Replacing the vector composed of four AMPA units with other vectors was found to abolish cytotoxicity, although all of the conjugates enabled pepstatin transport into cells. In addition, the AMPA4 vector must be localized at the C terminus of the bioconjugate. The unexpected importance of the vector structure and position for cytotoxic action suggests that AMPA4 enables pepstatin to inhibit the proteolysis of critical CathD substrates involved in cell proliferation via a unique mechanism of action.  相似文献

Hydropower energy generation depends on the available water resources. Therefore, planning and operation of the water resource systems are paramount tasks for energy management. Since reservoirs are one of the important components of water resources systems, extracting optimal operating policies for proper management of energy generated from these systems is an imperative step. Optimizing reservoir system operation (ORSO) is a non-linear, large-scale, and non-convex problem with a large number of constraints and decision variables. To solve ORSO problem effectively, a robust diversity-based, sine-cosine algorithm (RDB-SCA) is developed in the present study by introducing several strategies to balance the global exploration and local exploitation ability and to achieve accurate and reliable solutions. An efficient linear operation rule is coupled with the RDB-SCA to maximize the energy generation. The proposed method is then applied to a real-world, multi-reservoir system to extract optimal operational policies and, consequently, maximize the energy production. It is shown that the RDB-SCA is able to generate 24, 14, and 6% more energy than the original SCA, respectively for 2-, 3-, and 4-reservoir systems. The present findings are useful to suggest guidelines for efficient operation of hydropower multi-reservoir systems. This paper is supported by https://imanahmadianfar.com/codes.

Past project data sources provide key information for construction cost estimators. Previous research studies show that relying only on one’s own experience during estimation results in estimators’ bias. Having and referring to historical databases, containing objective information on what happened in past projects, are essential for reducing estimators’ biases. The first step toward development of useful project history databases is to understand what information estimators require from past projects. The research described in this paper targets estimators’ information needs identified through interviews, brainstorming sessions, task analyses, and card games conducted with estimators with different experience levels and specialized in heavy/civil and commercial construction projects, and exploration of historical and standard databases available in companies to determine what is being currently represented. Findings show that estimators need contextual information, depicting the conditions under which specific production rates were achieved, so that they can identify which production rate would be more realistic to use during the production rate estimation of an activity in a new bid. Comparison of the contextual information needs identified in this research with information items available in historical data sources (such as company cost reports, RSMeans, previous studies) highlighted some gaps and important opportunities for improvements in those sources. The identified contextual information items are significant for practitioners in developing ways to augment their existing project history databases to make them more beneficial for estimators.  相似文献

The shape of the lactation curve for 475 Turkish Holsteins was estimated by fitting a gamma function to daily milk yields from monthly recording of 754 lactations. Lactation curve traits that were analyzed included a scaling factor associated with yield at the beginning of lactation, the inclining and declining slopes before and after peak yield, DIM at peak yield, and peak and lactation yields. Persistency of lactation yield was measured from 1) the gamma function, 2) the coefficient of variation for monthly test-day yields, and 3) the ratio of lactation yield to peak yield. The log-transformed gamma function explained 71% of variation in daily yield. Effects of farm operation, calving year, calving season, parity, and service period were significant for the various lactation curve traits. Peak and lactation yields were higher for cows that calved in fall and winter, and persistency was higher for cows that calved in summer and fall. Peak and lactation yields were lower, but persistency was higher during first lactation. Repeatability estimates were moderate for peak (0.26) and lactation (0.34) yields and lower (0.06 to 0.20) for other lactation curve traits.  相似文献

The ease of deployment of Wireless Sensor Networks (WSNs) makes them very popular and useful for data collection applications. Nodes often use multihop communication to transmit data to a collector node. The next hop selection in order to reach the final destination is done following a routing policy based on a routing metric. The routing metric value is exchanged via control messages. Control messages transmission frequency can reduce the network bandwidth and affect data transmission. Some approaches like trickle algorithm have been proposed to optimize the network control messages transmission. In this paper, we propose a collaborative load balancing algorithm (CoLBA) with a prediction approach to reduce network overhead. CoLBA is a queuing delay based routing protocol that avoids packet queue overflow and uses a prediction approach to optimize control messages transmission. Simulation results on Cooja simulator show that CoLBA outperforms other existing protocols in terms of delivery ratio and queue overflow while maintaining a similar end-to-end delay.

Vanadium nitride(VN) was deposited by DC-sputtering on a vertically aligned carbon nanotube(CNTs)template for the purpose of nano-structuration. This led to the fabrication of hierarchically composite electrodes consisting of porous and nanostructured VN grown on vertically aligned CNTs in a nano-treelike configuration for micro-supercapacitor application. The electrodes show excellent performance with an areal capacitance as high as 37.5 m F cm~(-2) at a scan rate of 2 mV s~(-1) in a 0.5 MK_2SO_4 mild electrolyte solution. Furthermore, the capacitance decay was only 15% after 20,000 consecutive cycles. Moreover,the capacitance was found to increase with VN deposit thickness. The X-ray photoelectron spectroscopy analyses of the electrodes before and after cycling suggest that the oxide layers that form at the VN surface is the responsible for the redox energy storage in this material. Such electrodes can compete with other transition metal nitride based electrodes for micro-supercapacitors.  相似文献

The use of naturally present heterogeneous catalysts has recently been an essential issue in the Fenton and photo-Fenton processes. In this study, the uses of basalt as a catalyst for the Fenton and photo-Fenton reactions for methylene Blue (MB) and Basic Red 18 (BR18) degradations were investigated. Basalt was selected because of the presence of the iron (III) oxide in the structure. Basalt was characterized by X-Ray Fluorescence (XRF) and X-Ray Diffraction (XRD) analysis to obtain the chemical composition and the crystalline phase. The surface charge and the surface area were obtained by zeta potential and Brunauer Emmett-Teller (BET) analysis. Fourier Transform Infrared Spectroscopy (FTIR) and scanning electron microscope (SEM) were utilized to explore the functional group and the surface morphology. Fenton and photo-Fenton processes were applied to explore the best degradation method. Adsorption was also tested and the adsorption process had minimum removal efficiency (12% for MB and 17% for BR18). The removal efficiencies for MB and BR18 by the Fenton process were 87% and 28%, respectively. The photo-Fenton process had maximum removal efficiency with 100% for MB and 70% for BR18. The optimum conditions were 70 mg/L dye concentration, 5 mM H2O2, 1.0 g/L basalt loading and pH 2. Basalt has shown reuse capability as a catalyst for three consecutive cycles.  相似文献

The impregnation of o-phenylene dioxydiacetic acid (OPDA) into a polymeric matrix, Amberlite XAD-2000, is reported and was characterized by infrared spectroscopy. The amount of attached OPDA to the polymer resin was found to be 1.77mmolg(-1) resin. The resin was used for the sorption of U(VI) and Th(IV) from aqueous solution. This sorbent was capable of preconcentrating U(VI) and Th(IV) from weakly acidic or neutral solution. The retained metals were eluted sequentially using 0.25molL(-1) HCl for U(VI) and 1molL(-1) HCl for Th(IV) and determined spectrophotometrically using arsenazo-(III). The capacity of the resin for U(VI) and Th(IV) was found to be 0.121 and 0.113mmolg(-1), respectively. The impregnated resin exhibits a high chemical stability, reusability and fast equilibration. The method was used for the determination of U(VI) and Th(IV) in synthetic samples and rock samples.  相似文献
