ABSTRACT: The thermal stability of Bacillus licheniformis α-amylase at low moisture content was studied, based on isothermal experiments performed in a temperature range 113 to 125 °C. The thermal inactivation was monitored by measuring the decrease in thermal denaturation enthalpy and/or the decrease in enzymic activity on pnitrophenyl-α-D-maltoheptaoside, or on starch as a substrate. Based on enthalpy readings, an enzymic system with a z-value of 10.4 °C was observed when using a relative humidity of 81% at 4 °C. A theoretical study showed that this system could be used as a Time Temperature Integrator (TTI) to monitor the safety of sterilization processes of numerous food products. 相似文献

Biphasic and nth-order models were tested as to their usefulness to fit experimental inactivation data of Bacillus licheniformis α-amylase, immobilized on glass beads, and were discussed with respect to their suitability to characterize the considered enzymic system as a time—temperature integrator (TTI) to evaluate heat processes. Both isothermal and non-isothermal inactivation experiments were carried out. Model (kinetic) parameters (rate constant k, activation energy EA and reaction order n) were estimated using a non-linear regression procedure. The results obtained, especially the activation energy of about 293 kJ mole–1 , indicated a potential use of this system as a TTI for heating processes in the temperature range of 96–108°C. 相似文献

Currently, the sterility of heat‐processed food and pharmaceuticals is assessed in terms of an F0 value, based on the equivalence of the heat treatment to an isothermal process at a reference temperature. This F0 value, however, has a meaning if, and only if, the inactivation kinetics of the targeted spores (or cells) follow a first‐order relationship and the temperature dependence of the D value, the reciprocal of the rate constant, is log‐linear. There is growing evidence that these conditions are not satisfied by many spores, including those of Clostridium botulinum and vegetative cells. Consequently, a replacement for the F0 value is proposed in the form of a momentary equivalent time at the reference temperature based on the actual survival pattern of the spores, which need not be log‐linear. This equivalent time can be calculated together with the theoretical survival ratio in real time, thus enabling an operator to monitor the lethality of ongoing industrial heat processes. The concept is demonstrated with published survival data of C. botulinum, for which the Weibullian and log‐logistic models served as primary and secondary models, respectively. The safety factor according to the proposed method is in the number of added minutes of processing, beyond the theoretical time needed to reduce the survival ratio of the targeted spores or cells to a level that would produce practical (or commercial) sterility. Global bacterial infections associated with conventional polyvinyl chloride (PVC) medical devices place a heavy burden on healthcare systems and thus it will be desirable if medical devices are made from antimicrobial PVC. There are numerous studies focusing on polymer surface modifications to either leach antimicrobial agents or kill pathogenic microbes upon direct contact. In this work, mannitol fumarate ester-based aluminum metal alkoxide (MFE-Al) additive was developed to confer simultaneously improved antibacterial property and enhanced high temperature sterilization resistance of the resultant PVC. Data obtained confirm that the MFE-Al stabilized PVC sheets significantly inhibit 98% bacterial growth. They also show biocompatibility with cultured H9C2 cardiomyocytes and hemocompatibility in vitro. Dry heat sterilization is generally not suitable for PVC medical wares due to their poor thermal compatibility. Surprisingly, our antimicrobial-biocompatible PVC can maintain stability at 180°C for 90 min. Such a high thermal stability indicates the MFE-Al stabilized PVC can endure 90 cycles of dry-heat sterilization without significant damage. A new non-heating technique was developed for the sterilization of food stuff. Applying a roller compactor, ultra-high pressure sterilization has experimentally demonstrated its ability to sterilize dry powders, such as corn flour and Chinese herbs, with little quality deterioration. Also, the degree of food sterilization was found correlated well with the linear press forces between the rollers, roller gaps, and number of compaction passes. In comparison to the conventional high pressure sterilization technique, the new dry continuous processing method has the advantage of lower investment cost and is more versatile for sterilizing various food powders. 相似文献
