PRO teolysis TA rgeting C himeras (PROTACs) promote the degradation, rather than inhibition, of a drug target as a mechanism for therapeutic treatment. Bifunctional PROTAC molecules allow simultaneous binding of both the target protein and an E3-Ubiquitin ligase, bringing the two proteins into close spatial proximity to allow ubiquitinylation and degradation of the target protein via the cell's endogenous protein degradation pathway. We utilized native mass spectrometry (MS) to study the ternary complexes promoted by the previously reported PROTAC GNE-987 between Brd4 bromodomains 1 and 2, and Von Hippel Lindeau E3-Ubiquitin Ligase. Native MS at high resolution allowed us to measure ternary complex formation as a function of PROTAC concentration to provide a measure of complex affinity and stability, whilst simultaneously measuring other intermediate protein species. Native MS provides a high-throughput, low sample consumption, direct screening method to measure ternary complexes for PROTAC development. 相似文献

Leucine-rich repeat kinase 2 (LRRK2) is a large protein involved in the pathogenesis of Parkinson's disease (PD). It has been demonstrated that PD is mainly conferred by LRRK2 mutations that bring about increased kinase activity. As a consequence, selective inhibition of LRRK2 may help to recover the normal functions of LRRK2, thereby serving as a promising alternative therapeutic target for PD treatment. The mapping of LRRK2 by positron emission tomography (PET) studies allows a thorough understanding of PD and other LRRK2-related disorders; it also helps to validate and translate novel LRRK2 inhibitors. However, no LRRK2 PET probes have yet been reported in the primary literature. Herein we present a facile synthesis and preliminary evaluation of [11C]GNE-1023 as a novel potent PET probe for LRRK2 imaging in PD. [11C]GNE-1023 was synthesized in good radiochemical yield (10 % non-decay-corrected RCY), excellent radiochemical purity (>99 %), and high molar activity (>37 GBq μmol−1). Excellent in vitro binding specificity of [11C]GNE-1023 toward LRRK2 was demonstrated in cross-species studies, including rat and nonhuman primate brain tissues by autoradiography experiments. Subsequent whole-body biodistribution studies indicated limited brain uptake and urinary and hepatobiliary elimination of this radioligand. 采用Thermecmaster-Z型热加工模拟实验机对Ti-1023合金进行了等温恒应变速率压缩试验,变形温度范围为:740-820℃;变形速率为:0.001~1s^-1。在对试验数据进行系统分析的基础上,全面揭示了Ti-1023合金在等温锻造条件下的热态变形行为。分析了锻造热力参数对流动应力的影响,提出了以Zener-Hollomn参数表征的Ti-1023合金本构方程。并初步分析了Ti-1023合金等温锻造过程中显微组织演化规律。根据分析结果,Ti-1023合金等温锻造宜在略低于相变点温度下以相对高的变形速率进行。 相似文献

研究了工业化制备的超大规格Ti-1023合金铸锭在不同磁场强度下的纵向凝固组织和Fe元素成分分布,并对其影响进行了探讨。结果表明:当磁场强度为10Gs和30Gs时,铸锭组织主要由粗大柱状晶,竹节状的小柱状晶和等轴晶组成;当磁场搅拌强度为20Gs时,凝固组织转变为由底部粗大柱状晶和细小等轴晶组成。Ti-1023合金铸锭中Fe元素在不同磁场强度下遵循正偏析规律,沿铸锭长度方向Fe元素含量从底部到顶部逐渐增加,沿径向从边缘到中心逐渐增加。随着磁场强度从10Gs增加到30Gs,合金元素Fe的宏观偏析率先减小后增大。 相似文献
