For determining trypsin inhibitor activity (TIA) in soy products, the American Oil Chemists' Society (AOCS) Method Ba 12-75 has been used. It measures differences in absorbance at 410 nm of bovine trypsin activity toward a synthetic substrate (-benzoyl-DL-arginine-p-nitroanilide) in the absence and presence of an inhibitor. Recently, a significantly improved method was developed (JAOCS, 2019, 96:635–645), featuring 5 mL of total assay volume, enzyme-last sequence, and single inhibitor level in duplicate. It is proposed as the AOCS Method Ba 12a-2020. As a part of the AOCS method approval process, a collaborative study involving 12 international laboratories was conducted to evaluate the performance of the proposed method. The study involved measuring TIA in 10 selected test samples plus a blind duplicate. They included soybeans, pulses, cereals, and their processed products (flours, concentrates, and isolates). After rigorous statistical treatment of the data, only three outliers were removed from the data of two samples. Repeatability relative standard deviations (RSDr) for the 11 samples ranged from 0.99% to 5.52%. Reproducibility RSD (RSDR) ranged from 7.07% to 22.92%, with seven samples having RSDR around 10% or less. The remaining four samples had very low TIA, and their RSDR values ranged from 13.34% to 22.92%. The study has demonstrated reliable performance of the proposed AOCS method. Several collaborators carried out additional experiments addressing some aspects of the method, leading to further refinements. The proposed method is undergoing evaluation by the AOCS Uniform Methods Committee for adoption as an Official Method for measuring TIA in various legume and grain products.  相似文献

该文基于输出一阈值耦合神经网络的自动波现象,提出了一种用自动波方法求解TSP问题的方法。该方法具有鲁棒性和可靠性好、大规模并行计算等特点,可用于求解对称、非对称赋权图的TSP问题。与目前其它求解TSP问题的方法相比,自动波方法执行更为简单,不需要太多人为的选择参数等问题,且不存在局部极小点的问题,求得的解全部是最优解。其所需的计算量(迭代次数)主要取决于最短回路的长度,而与图的复杂程度、所存在的通路总数关系不大。最后文中给出了TSP求解的例子。  相似文献

The purpose of this study was to investigate the T-2 toxin level of contaminated cereal and pulse products in Turkey. T-2 toxin was detected using high performance liquid chromatography (HPLC) with UV detection at 208 nm and thin layer chromatography (TLC) was used for confirmation of the T-2 toxin-contaminated samples (> or = 1 ppm). The recovery was 93 +/- 3.3% (SD 3.29, n = 5) for chickpea spiked with a known amount of T-2 toxin (1 ppm). The detection limits for T-2 toxin for HPLC and TLC were 25 ng per injection and 50 ng per spot, respectively. A total of 50 commercially available cereal and pulse product samples, collected from markets and street bazaars, were analysed. Incidences of T-2 toxin detected in cereal and pulse products were 23.5% and 31.2%, respectively and the maximum detected amount was 1.9 ppm in a sample of dried beans. The incidence of toxin-contaminated specimens is not so low relative to the volume of specimens produced.  相似文献

The effects of the herbicide Diuron (DIU) and the bactericide Triclosan (TCS) were assessed on laboratory-grown stream biofilms. Four week-old biofilms were exposed in mesocosms to 48-hours of short pulses of either DIU or TCS. The direct and indirect effects of each toxicant on the biofilms, and the subsequent recovery of the biofilms, were evaluated according to structural and functional biomarkers. These parameters were analyzed immediately before exposure, immediately after exposure, and 9 and 16 days post-exposure. DIU caused an increase in diatom mortality (+ 79%), which persisted until the end of the experiment. TCS also affected diatom mortality (+ 41%), although the effect did not appear until 1 week post-exposure. TCS caused an increase in bacterial mortality (+ 45%); however, this parameter returned to normal values 1 week post-exposure. TCS compromised the cellular integrity of the green alga Spirogyra sp., whereas DIU did not. TCS also strongly inhibited phosphate uptake (− 71%), which did not return to normal values until 2 weeks post-exposure. DIU directly affected algae, but barely affected the heterotrophs, whereas TCS seriously impaired bacteria (direct effect) as well as autotrophs (indirect effect). However, the biofilms recovered their normal structure and function within only a few days to a few weeks. These findings demonstrate the capacity of biofilms to cope with periodic inputs of toxicants, but also the risks associated to repeated exposure or multi-contamination in aquatic ecosystems.  相似文献

The effect of microwave treatment to reduce the cooking times of five pulses, namely red lentil, chickpea, pigeon pea, mung bean, and pinto bean, were determined in this study. Pulses from 10 to 18% moisture contents were treated using 400 to 600 W microwaves for 14 to 56 s. The cooking times of microwave-treated pulses were significantly lower than that of the control samples. The lowest cooking time was observed for 18% moisture content chickpea and pigeon pea treated with 600 W for 56 s. The Fourier transform mid-infrared spectra in both lipids and fingerprint regions showed the macronutrients differences among the five pulses. Major changes were observed in the amide I region of microwave treated pulses. This effect of microwave treatment was higher in red lentil, chickpea, and mung bean than in pigeon pea and pinto bean at 10% moisture content. At 18% moisture content, the change of β-sheets to aggregates was observed in all pulses due to microwave treatment.  相似文献

Despite their high nutritional value and potential health benefits, pulse intake has not increased in the last three decades. Several strategies have been implemented to increase pulse consumption, such as their incorporation in bakery products, breakfast cereals, and snacks. The inclusion of pulses in these products could be an alternative to satisfy the consumers’ demand for healthy foods. However, pulse-based snacks face important challenges, including reducing antinutritional factors, achieving consumer acceptance, and consolidating the pulse-based snacks as functional foods. This review summarizes and discusses methods for producing snacks where cereals or tubers were replaced with at least 50% pulses. Also, it briefly assesses their effect on nutritional composition, antinutritional factors, sensory acceptance, and different health benefits evaluations. Extruded snacks exhibited high protein and dietary fiber and low fat content, contrary to the high fat content of deep fat–fried snacks. Meanwhile, baked snacks presented moderate concentrations of protein, dietary fiber, and lipids. Pulses must be pretreated using process combinations such as soaking, dehulling, cooking, fermentation, germination, and extrusion to reduce the antinutritional factors. Pulse-based snacks show good sensory acceptance. However, sensory evaluation should be more rigorous using additional untrained judges. Several studies have evaluated the health benefits of pulse-based snacks. More research is needed to validate scientifically the health benefits associated with their consumption. Pulse-based snacks could be an alternative to improve the nutritional composition of commercially available snacks. The use of pulses as ingredients of healthier snacks represents an important alternative for the food industry due to their low cost, sensory characteristics, high nutritional profile, and environmental benefits.  相似文献

Five commonly consumed pulses, Mah (Vigna mungo), Green mung (Vigna radiata), Arhar (Cajanas cajan), Masur (Lens esculantus), and Moth (Vigna aconitifolia), were studied for their total phenolic content and antioxidant activity after germination (12 and 24 h) and pressure cooking. Arhar had the highest total phenolic content (6.71 mg ferulic acid/g flour) whereas Moth had the least (1.54 mg/g). All pulses, except Moth, showed a significant decrease in total phenolic content after germination. The antioxidant activity of the pulses varied from 10.61 to 36.38% (DPPH radical scavenging activity), which significantly decreased with germination in all pulses except Moth. The total phenolic content highly correlated with the antioxidant activity in the pulses. Cooking lowered the total phenolic content by 10–45% and antioxidant activity by 27–68% in the control and germinated pulses.  相似文献

Several cereals and pulses commonly consumed in India were screened for zinc and iron contents and their bioaccessibility in the same was determined by equilibrium dialysis employing an in vitro simulated digestion procedure. Zinc content of cereals ranged from 1.08 mg/100 g in rice to 2.24 mg/100 g in sorghum. Zinc content of pulses was between 2.03 mg/100 g (whole chickpea) and 2.68 mg/ 100 g (decorticated chickpea). Iron content of cereals ranged from 1.32 mg% in rice to 6.51 mg% in sorghum, while that of pulses ranged from 3.85 mg% in decorticated green gram to 6.46 mg% in black gram. Dialyzability of zinc from pulses (27–56%) was generally higher than that from cereals (5.5–21.4%). Dialyzabilities of iron were almost similar from both cereals and pulses examined and were 4.13–8.05% in cereals and 1.77–10.2 % in pulses. A significant negative correlation between inherent phytate content and zinc dialyzability value was inferred in the case of pulses. Phytic acid content of the cereals had a significant negative influence on iron dialyzability. Inherent calcium had a negative influence on zinc dialyzability in cereals. Tannin did not have any significant influence on zinc or iron dialyzabilities from cereals and pulses. While both insoluble and soluble fractions of the dietary fibre generally interfered with zinc dialyzability, the insoluble fraction alone had this effect on iron dialyzability. The lower collective negative influence of the inherent factors on zinc dialyzability from pulses is consistent with their higher concentrations in these grains, relative to cereals. The negative correlation of inherent phytic acid with zinc and iron dialyzabilities was supported by enhanced dialyzabilities of these minerals upon partial removal of phytate from the grains by treatment with fungal phytase.  相似文献
