Adsorption isotherms of sesame oil in a concentrated miscella system   总被引:2,自引:0,他引:2  
The adsorption of peroxides, unsaturated carbonyls, free fatty acids and carotenoids from unrefined sesame oil on vegetable carbon (5%, w/w) in a miscella system was studied. Three different solvent conditions (hexane/ethanol, 100:0, 95:5 and 75:25, vol/vol), combined in a factorial design with five levels of solvent (0, 10, 20, 30 and 40%, w/w), were used to develop the miscella. Equilibrium adsorption was not achieved during the 100 min of adsorption, mainly because the oil components were involved in oxidation reactions during the adsorption process. However, for a given solvent concentration, adsorption of the oil components showed a significant linear regression on their respective initial concentration in the miscella (Ci). Peroxides and carbonyls showed, at all solvent levels investigated, an affinity for the carbon more independent of their Ci than free fatty acids and carotenoids. In general, at the same Ci, a higher adsorption was achieved as solvent concentration increased. The results indicated that free fatty acid adsorption may depend on competitive adsorption based on molecule hydrophobicity. However, in spite of the higher hydrophobicity of carotenoids, compared with free fatty acids, they might not be competing for the same adsorbing sites. Ethanol showed a prooxidant effect that increased peroxide production during adsorption but did not affect the reaction involved in carbonyl production.  相似文献

Thraustochytrids, marine protists whose dominant genera are Thraustochytrium and Schizochytrium, belong to the kingdom Chromista and are known as an industrial source of DHA. We describe here that thraustochytrid strain KH105, isolated as a DHA producer, also accumulates significant levels of β-carotene and xanthophylls including canthaxanthin and astaxanthin. A4-d cultivation using a medium composed of 10% glucose and less than 0.3% of nitrogen sources in a half-concentration of seawater gave an astaxanthin production up to 6.1 mg/L, and canthaxanthin content reached more than 10 mg/L under conditions where a higher concentration of nitrogen sources (6%) was employed. It might be advantageous in mass production systems for these carotenoids to be extracted readily by simply suspending the cells with organic solvents such as acetone and chloroform. Analyses on the morphological and life history features of the KH105 strain revealed that it belongs to the genus Schizochytrium. This particular species of thraustochytrids is thus considered to be a promising source of xanthophylls as well as DHA for use in the food industry.  相似文献

以棉花、玉米、大豆、甘薯四种作物为材料,各采集叶片30张(处于不同部位、不同功能期),分别测定其反射光谱和叶绿素、类胡萝卜素含量。目的在于探讨利用高光谱技术提取类胡萝卜素信息的可行性方法。结果表明,由于叶绿素与类胡萝卜素间存在显的相关性,在叶片水平,利用高光谱反射率估测叶片类胡萝卜素绝对量是可行的。与类胡萝卜素/叶绿素比值或类胡萝卜素含量相比,类胡萝卜素密度(单位叶片面积类胡萝卜素总量,Cardens)与光谱反射率间的相关性更为稳定。类胡萝卜素光谱吸收峰(470nm)附近的反射光谱与Cardens间的相关性较差,基于类胡萝卜素吸收峰附近反射光谱的光谱指数(如PSSRc、PSNDc)与Cardens间也表现出较弱的相关性。叶绿素光谱指数(如SR705、ND705)与Cardens间存在良好的相关性,红边光谱区的微分光谱、包络线归一化吸收深度等高光谱指数与Cardens间也表现出了良好的相关性。  相似文献

Light absorption by carotenoids is known to vary substantially with the shape or conformation of the pigment molecule induced by the molecular environment, but the role of interactions between carotenoid pigments and the proteins to which they are bound, and the resulting impact on organismal coloration, remain unclear. Here, we present a spectroscopic investigation of feathers from the brilliant red scarlet ibis (Eudocimus ruber, Threskiornithidae), the orange-red summer tanager (Piranga rubra, Cardinalidae) and the violet-purple feathers of the white-browed purpletuft (Iodopleura isabellae, Tityridae). Despite their striking differences in colour, all three of these feathers contain canthaxanthin (β,β-carotene-4,4′-dione) as their primary pigment. Reflectance and resonance Raman (rR) spectroscopy were used to investigate the induced molecular structural changes and carotenoid–protein interactions responsible for the different coloration in these plumage samples. The results demonstrate a significant variation between species in the peak frequency of the strong ethylenic vibration (ν1) peak in the rR spectra, the most significant of which is found in I. isabellae feathers and is correlated with a red-shift in canthaxanthin absorption that results in violet reflectance. Neither polarizability of the protein environment nor planarization of the molecule upon binding can entirely account for the full extent of the colour shift. Therefore, we suggest that head-to-tail molecular alignment (i.e. J-aggregation) of the protein-bound carotenoid molecules is an additional factor.  相似文献

Background and Aims: Carotenoids and chlorophylls perform a number of essential roles in plants making their accurate quantification important to a variety of studies. We aimed to develop an extraction protocol to accurately determine the photosynthetic pigments in grapevine leaf and berry tissue, specifically focusing on limiting the degradation of these pigments. Methods and Results: An extraction protocol for grapevine leaf and berry tissue was systematically optimised by identifying a number of critical parameters. Extracted pigments were analysed using Reversed Phase-High Performance Liquid Chromatography (RP-HPLC). Specific parameters that were optimised included avoiding freeze-drying the material; the volume of acetone and the time required to extract all the pigments from the tissue; the addition of 0.1% (v/v) N-ethyldiisopropylamine to berry extracts to minimise pigment degradation during the extraction procedure; and avoiding concentration of the extracts that otherwise resulted in differential degradation of pigments. Additionally, the method of extraction and normalisation with an internal standard was adapted and improved for accuracy. The optimised protocol was validated using authentic standards and its utility shown by analysing the pigment content of berries and leaves at different growth stages. Conclusions: A method has been developed that is able to extract and accurately quantify, by means of HPLC profiling, the levels of photosynthetic pigments from grape berries and leaves. The method avoided any degradation of the pigments during the extraction and was applicable to both berries and leaves in different stages of growth and development, indicating its general usefulness to vegetative and reproductive organs, even if their metabolic states are very different. Significance of Study: The divergence of methods used for photosynthetic pigment analysis in plants, each with specific advantages and disadvantages were considered and used to optimise a number of parameters in a single method that proved to be applicable to plant organs in different developmental stages. The method is fast, applicable to vegetative and reproductive grapevine tissues, avoids degradation of pigments and ensures maximum accuracy when quantifying these important pigments.  相似文献

A study of chlorophyll and carotenoid composition of nine single-variety virgin olive oils from the main Spanish producing regions has shown differences depending on variety and ripening degree of the fruits. Pheophytin a was the major pigment in all the oils studied (44–58% total pigments), followed by lutein (18–38%) and β-carotene (6–17%). The chlorophyll pigments group also included pheophytin b and in some cases chlorophylls a and b . The carotenoid fraction also included the xanthophylls neoxanthin, violaxanthin, luteoxanthin, antheraxanthin, mutatoxanthin and β-cryptoxanthin. The mean provitamin A activity of the oils was 260 μg kg−1 expressed as retinol equivalent. cis -α-Carotene (tentative), mono- and di-esterified xanthophylls and pheophorbide a were pigments exclusive to Arbequina variety, a fact that could be used as a chemo-taxonomic differentiator of the oils of this variety. The ratio between the chlorophyll fraction and the carotenoid fraction was maintained in most cases around 1, demonstrating that the green and yellow fractions were in balance, notwithstanding their greater or lesser total pigment content. The considerable range in the lutein/β-carotene ratio (between 1·3 and 5·1 depending on variety) makes this ratio a differentiator of single-variety oils. Despite the different sources of the oils, some general trends have been shown which suggest what chlorophyll and carotenoid pigments should be expected in an olive oil so as to include it within the denomination ‘virgin’.  相似文献

White spruce (Picea glauca) emits monoterpenes that function as defensive signals and weapons after herbivore attack. We assessed the effects of drought and methyl jasmonate (MeJA) treatment, used as a proxy for herbivory, on monoterpenes and other isoprenoids in P. glauca. The emission of monoterpenes was significantly increased after MeJA treatment compared to the control, but drought suppressed the MeJA-induced increase. The composition of the emitted blend was altered strongly by stress, with drought increasing the proportion of oxygenated compounds and MeJA increasing the proportion of induced compounds such as linalool and (E)-β-ocimene. In contrast, no treatment had any significant effect on the levels of stored monoterpenes and diterpenes. Among other MEP pathway-derived isoprenoids, MeJA treatment decreased chlorophyll levels by 40%, but had no effect on carotenoids, while drought stress had no impact on either of these pigment classes. Of the three described spruce genes encoding 1-deoxy-D-xylulose-5-phosphate synthase (DXS) catalyzing the first step of the MEP pathway, the expression of only one, DXS2B, was affected by our treatments, being increased by MeJA and decreased by drought. These findings show the sensitivity of monoterpene emission to biotic and abiotic stress regimes, and the mediation of the response by DXS genes.  相似文献

The development and promotion of biofortified foods plants are a sustainable strategy for supplying essential micronutrients for human health and nutrition. We set out to identify quantitative trait loci (QTL) associated with carotenoid content in cowpea sprouts. The contents of carotenoids, including lutein, zeaxanthin, and β-carotene in sprouts of 125 accessions were quantified via high-performance liquid chromatography. Significant variation existed in the profiles of the different carotenoids. Lutein was the most abundant (58 ± 12.8 mg/100 g), followed by zeaxanthin (14.7 ± 3.1 mg/100 g) and β-carotene (13.2 ± 2.9 mg/100 g). A strong positive correlation was observed among the carotenoid compounds (r ≥ 0.87), indicating they can be improved concurrently. The accessions were distributed into three groups, following their carotenoid profiles, with accession C044 having the highest sprout carotenoid content in a single cluster. A total of 3120 genome-wide SNPs were tested for association analysis, which revealed that carotenoid biosynthesis in cowpea sprouts is a polygenic trait controlled by genes with additive and dominance effects. Seven loci were significantly associated with the variation in carotenoid content. The evidence of variation in carotenoid content and genomic regions controlling the trait creates an avenue for breeding cowpea varieties with enhanced sprouts carotenoid content.  相似文献

首先研究不同红外辐射温度(100,110,120℃)及辐射时间(2.5,5,10 min)对胡萝卜粉微生物及品质的影响,然后根据栅栏效应原理研究红外辐射-回火、红外辐射-冷激联合杀菌对胡萝卜粉微生物、色调值、类胡萝卜素含量等品质的影响。结果表明:100℃、10 min的红外辐射处理使细菌和真菌分别降低了1.9 lg(CFU/g)和2.32 lg(CFU/g),110℃、5 min的红外辐射处使细菌和真菌分别降低了1.58 lg(CFU/g)和2.57 lg(CFU/g)。在上述两种处理条件下胡萝卜粉的水分活度从0.238分别降至0.123和0.147,胡萝卜粉中总类胡萝卜素含量从308.8μg/g降至227.8μg/g和238.8μg/g,色差值(ΔE)为9.11和7.89。与红外辐射单独作用相比,联合回火后的处理没有显著影响细菌数目,处理后保持在5.40~5.80 CFU/g,霉菌、酵母数却在处理过程中显著减少,然而减少量较低,总数仍不低于4.5 lg(CFU/g)。红外辐射-冷激联合处理相比红外辐射单独处理,100℃、10 min联合冷激7 d处理可将细菌数量降低0.25 lg(CFU/g),可将霉菌与酵母数量降低0.28 lg(CFU/g),110℃、5 min联合冷激7 d处可将细菌数量降低0.26 lg(CFU/g),可将霉菌、酵母数量降低0.40 lg(CFU/g),且这些处理下胡萝卜粉的色调值、水分活度、类胡萝卜素含量变化不显著。本试验结果表明,红外辐射-冷激处理具有协同效应,且处理过程中胡萝卜粉的色调值及总类胡萝卜素含量不受影响,这为低水分粉体食品红外辐射联合杀菌提供了参考。  相似文献
