The intracellular distribution of the anthracyclinic antibiotic adriamycin in living cultured cells has been investigated by confocal microscopy. In human melanoma cells (M14), adriamycin was localized inside the nuclei. When adriamycin-treated M14 cells were allowed to recover in drug-free medium, a complete efflux of the drug from the nucleus was revealed. In recovered cells, a weakly fluorescent signal was observed in the perinuclear region. When M14 cells were recovered in a medium containing colcemid, a microtubule depolymerizing agent, the drug transport from the nucleus to the cell periphery appeared to be inhibited, suggesting that the microtubule network is strongly involved in drug transport mechanisms. In multidrug-resistant (MDR) cells the intracellular location of adriamycin was shown to be noticeably different from that of the parental wild-type cells. In particular, in resistant human breast carcinoma cells (MCF-7), adriamycin appeared to be exclusively located within the cytoplasm whereas the nuclei were shown to be completely negative. When adriamycin treatment was performed in association with MDR revertants, such as Lonidamine (inhibitor of the energy metabolism) or verapamil (inhibitor of the P-glycoprotein efflux pump), a marked enhancement of the cytoplasmic signal was observed in resistant cells. Under these conditions, adriamycin appeared concentrated in the perinuclear region, but the nuclei were still negative. Confocal microscopy proved to be a very useful method for the study of the intracellular transport of fluorescent substances, such as anthracyclinic antibiotics, and for the investigation of the multidrug resistance phenomenon in tumour cells. 相似文献

In this paper a set of one-dimensional simulations of a-Si:H p–i–n junctions under different illumination conditions and with different intrinsic layer are presented. The simulation program ASCA permits the analysis of the internal electrical behaviour of the cell allowing a comparison among the different internal configurations determined by a change in the input set. Results about the internal electric configuration will be presented and discussed outlining their influence on the current tension characteristic curve. Considerations about the drift–diffusion and the generation–recombination balance distributions, outlined by the simulation, can be used to explain the correlation between the basic device output, the i-layer characteristics (thickness and DOS), the incident radiation intensity and photon energy. 相似文献

New electroactive and photoactive conjugated copolymers consisting of alternating 2,7‐carbazole and oligothiophene moieties linked by vinylene groups have been developed. Different oligothiophene units have been introduced to study the relationship between the polymer structure and the electronic properties. The resulting copolymers are characterized by UV‐vis spectroscopy, size‐exclusion chromatography, and thermal and electrochemical analyses. Bulk heterojunction photovoltaic cells from different copolymers and a soluble fullerene derivative, [6,6]‐phenyl‐C61 butyric acid methyl ester, have been fabricated, and promising preliminary results are obtained. For instance, non‐optimized devices using poly(N‐(4‐octyloxyphenyl)‐2,7‐carbazolenevinylene‐alt‐3″,4″‐dihexyl‐2,2′;5′,2″;5″,2″′;5″′,2″″‐quinquethiophenevinylene 1″,1″‐dioxide) as an absorbing and hole‐carrier semiconductor exhibit power conversion efficiency up to 0.8 % under air mass (AM) 1.5 illumination. This study investigates quantitatively and qualitatively the sol-gel derived bioactive glass-ceramic system (BGS)—apatite-wollastonite
(AW) type granules in the size range of 0.5–1 mm, as an effective graft material for bone augmentation and restoration. Scanning
electron micrographs (SEM) of the sintered granules revealed the rough material surface with micropores in the range 10–30
μm. X-ray diffraction (XRD) pattern of the granules revealed the presence of crystalline phases of the hydroxyapatite and
wollastonite, and the functional groups of the silicate and phosphates were identified by Fourier transform infrared spectroscopy
(FT-IR). Thein vitro cell culture studies with L929 mouse fibroblast cell line showed very few cells adhered on the BGS disc after 24 h. This
could be due to the highly reactive surface of the disc concomitant with the crystallization but not due to the cytotoxicity
of the material, since the cellular viability (MTT assay) with the material was 80‰ Cytotoxicity and cytocompatibility studies
proved that the material was non-toxic and biocompatible. After 12 weeks of implantation of the BGS granules in the tibia
bone of New Zealand white rabbits, the granules were found to be well osteointegrated, as observed in the radiographs. Angiogram
with barium sulphate and Indian ink after 12 weeks showed the presence of microcapillaries in the vicinity of the implant
site implicating high vascularity. Gross observation of the implant site did not show any inflammation or necrosis. SEM of
the implanted site after 24 weeks revealed good osteointegration of the material with the newly formed bone and host bone.
New bone was also observed within the material, which was degrading. Histological evaluation of the bone healing with the
BGS granules in the tibial defect at all time intervals was without inflammation or fibrous tissue encapsulation. After 2
weeks the new bone was observed as a trabeculae network around the granules, and by 6 weeks the defect was completely closed
with immature woven bone. By 12 weeks mature woven bone was observed, and new immature woven bone was seen within the cracks
of the granules. After 24 weeks the defect was completely healed with lamellar bone and the size of the granules decreased.
Histomorphometrically the area percentage of new bone formed was 67.77% after 12 weeks and 63.37% after 24 weeks. Less bone
formation after 24 weeks was due to an increased implant surface area contributed by the material degradation and active bone
remodeling. The osteostimulative and osteoconductive potential of the BGS granules was established by tetracycline labelling
of the mineralizing areas by 2 and 6 weeks. This sol-gel derived BGS granules proved to be bioactive and resorbable which

There is a significant difference in performance between today's highest efficiency of CdTe solar cells and single-crystal cells of comparable band gap. The largest contribution to this difference is the voltage, where the values for the best CdTe cells are about 230 mV below the best GaAs cells when an appropriate adjustment is made for band gap. CdTe voltage and fill-factor are currently compromised by low recombination lifetime (near 1 ns), low hole density (near 1014 cm− 3), and in some cases an excessive back-contact barrier. Numerical simulation is used to evaluate how combinations of lifetime, carrier density, back electron reflection, and interfacial properties affect voltage and cell performance. Gas-phase selective oxidation of toluene has been carried out on vanadium oxide systems (5–20 wt.% of V2O5, equivalent to 0.4–1.7 theoretical monolayers) supported on TiO2–sepiolite (with titania loading around the theoretical monolayer, 12 wt.%) and on sepiolite. A study has been made on both the influence of vanadia loading and of the support on the catalytic behaviour of the supported vanadium systems. The reducibility by H2 TPR was also studied as well as the acid and basic/redox sites from the results of conversion of the 2-propanol test reaction of the solids. Benzaldehyde, benzoic acid and several coupling products were the main ones detected, attaining over 50% selectivity towards the benzaldehyde and benzoic acid products at a total conversion around 10%. The activity and selectivity to the selective products exhibited by vanadium systems supported on mixed support were superior to those exhibited by the systems supported on sepiolite and increased notably in both series with the increase in vanadium loading. The best catalytic behaviour exhibited by the vanadium systems supported on mixed support, which also exhibited the highest density of sites capable of being reduced (as well as their reducibility) and of those responsible for propanone formation, could be attributed not only to the different balance of the vanadia species existing in the two supports (monomeric + oligomeric/polymeric), but also to such other factors as the nature of the support and, concretely, its chemical composition. 相似文献

Automated monitoring of the microbiological quality of heat-processed foods by the resazurin reduction test was applied to microtitration plate incubator-fluorimeter technology. The appearance and disappearance of the fluorescing peak of resorufin was monitored on microtitration trays. Pasteurized or ultra-high temperature-treated starch-based soup was used as the model food system. Bacillus subtilis spores (ultra-high temperature treatment) and vegetative cells of Enterococcus faecalis (pasteurization) were inoculated into the soup before the heat treatment at levels which resulted in some survival. The timing of appearance of maximum fluorescence correlated with the number of bacteria in pre-incubated samples. Automated resazurin-reduction fluorimetry was compared with conventional plating, turbidometry and microcolony count by the direct epifluorescent filter technique. The results of the resazurin test correlated well with those of all the other methods tested. Fluorimetry had the advantage that the results could be read within 1–5h and the reproducibility was superior to the other methods. 相似文献
