Flavivirus genus includes many deadly viruses such as the Japanese encephalitis virus (JEV) and Zika virus (ZIKV). The 5′ terminal regions (TR) of flaviviruses interact with human proteins and such interactions are critical for viral replication. One of the human proteins identified to interact with the 5′ TR of JEV is the DEAD-box helicase, DDX3X. In this study, we in vitro transcribed the 5′ TR of JEV and demonstrated its direct interaction with recombinant DDX3X (Kd of 1.66 ± 0.21 µM) using microscale thermophoresis (MST). Due to the proposed structural similarities of 5′ and 3′ TRs of flaviviruses, we investigated if the ZIKV 5′ TR could also interact with human DDX3X. Our MST studies suggested that DDX3X recognizes ZIKV 5′ TR with a Kd of 7.05 ± 0.75 µM. Next, we performed helicase assays that suggested that the binding of DDX3X leads to the unwinding of JEV and ZIKV 5′ TRs. Overall, our data indicate, for the first time, that DDX3X can directly bind and unwind in vitro transcribed flaviviral TRs. In summary, our work indicates that DDX3X could be further explored as a therapeutic target to inhibit Flaviviral replication  相似文献

基于气溶胶中PM2.5微细颗粒物拟流体特性,对气液交叉流阵列中PM2.5在气溶胶流体传热传质边界层内热泳和扩散泳运动进行拟传质机理分析,与跟随气体的对流传质相叠加,建立了气液交叉流阵列PM2.5热泳和扩散泳拟传质模型,并进行了实验检验。实验在固定对流条件下,考察了不同气液相温度差导致的热泳、不同气相湿度差导致的扩散泳和颗粒粒径等因素对气液交叉流阵列PM2.5拟传质系数的影响。实验数据统计值与模型表达趋势一致,在初始温差40℃、初始湿度0.118 kg/kg条件下,100排气液交叉流阵列PM2.5拟传质系数模型预测值为3.33×10-3 m/s、实验值为3.75×10-3 m/s。  相似文献

The effect of heat and mass transfer in a MHD non‐Darcian flow of a micropolar fluid over an unsteady stretching sheet with thermophoresis and non‐uniform heat source/sink is discussed. The fluid is electrically conducting in the presence of a uniform applied magnetic field. The arising nonlinear problem is solved by the Keller box method. The effects of various physical parameters on skin friction, local Nusselt number, and Sherwood number are presented graphically and in tabular form. © 2012 Wiley Periodicals, Inc. Aptamers are attractive constructs due to their high affinity/selectivity towards a target. Here 7,8-dihydro-8-oxoguanosine (8-oxoG) has been used, due in part to its unique H-bonding capabilities (Watson–Crick or Hoogsteen), to expand the “RNA alphabet”. Its impact on the theophylline RNA aptamer was explored by modifying its binding pocket at positions G11, G25, or G26. Structural probing, with RNases A and T1, showed that modification at G11 leads to a drastic structural change, whereas the G25-/G26-modified analogues exhibited cleavage patterns similar to that of the canonical construct. The recognition properties towards three xanthine derivatives were then explored through thermophoresis. Modifying the aptamer at position G11 led to binding inhibition. Modification at G25, however, changed the selectivity towards theobromine (Kd≈160 μm ), with a poor affinity for theophylline (Kd>1.5 mm ) being observed. Overall, 8-oxoG can have an impact on the structures of aptamers in a position-dependent manner, leading to altered target selectivity.  相似文献

This study addresses the fully developed magnetohydrodynamic flow of non-Newtonian fluid in a microchannel using tangent hyperbolic fluid model. The physical situation has been modeled by accessing boundary layer theory along with the physical aspects of thermophoresis and Brownian motion. The heat and mass transport phenomena are depicted through graphical interpretations. The modeled equations are nondimensionalized using dimensionless variables. The obtained corresponding equations are solved by employing Runge–Kutta–Fehlberg scheme accompanied with shooting technique. The fluctuations in distinct entities of physical connotations, like, the Nusselt number, friction factor and Sherwood number are explored in this examination. A notable reduction in the concentration field of the tangent hyperbolic fluid has been obtained for a larger chemical reaction parameter. The result shows that non-Newtonian fluids exhibit higher Nusselt number than Newtonian fluids. Furthermore, a significant enhancement in Nusselt number has been attained through a rise in the power-law index and thermophoresis aspect.  相似文献

The convection in a vertical channel filled with a porous medium saturated by a nanofluid is studied numerically. The effects of Brownian motion and thermophoresis are incorporated in the model used for nanofluid. Also, the flow within the porous region is governed by Brinkman's equation. The generalized eigenvalue problem for the perturbed state is obtained from a normal mode analysis and solved using the Chebyshev spectral collocation method. The Rayleigh number is expressed as an implicit function of the wavenumber with other parameters. The critical wavenumber and the critical Rayleigh number are calculated for different parameters. The preferred modes under critical conditions are detected.  相似文献

This article examines the inclined magnetic field effect on the flow of micropolar nanofluids in a vertical channel with convective boundary conditions and heat source or sink. Thermodynamics second law is employed to analyze the aspects of entropy generation. The governing differential equations are modified into dimensionless form by using suitable nondimensional variables. These transformed equations are solved by implementing the differential transform technique. The results are analyzed graphically. Skin friction and Nusselt number values are evaluated at the boundary walls of the channel. The major findings of the study are material parameter enhances the microrotation but suppresses both velocity and temperature. Magnetic parameter and angle of the implication of magnetic field decrease the velocity and microrotation. Material parameter and angle of imposed magnetic field minimize the entropy generation.  相似文献

The Buongiorno model Maxwell nanofluid flow, heat and mass transfer characteristics over a stretching sheet with a magnetic field, thermal radiation, and chemical reaction is numerically investigated in this analysis. This model incorporates the effects of Brownian motion and thermophoresis. The governing partial differential equations are transformed into a coupled nonlinear ordinary differential equation by using the similarity transformation technique. The resultant nonlinear differential equations are solved by using the Finite element method. The sketches of velocity, temperature and concentration with diverse values of magnetic field parameter (0.1 ≤ M ≤ 1.5), Deborah number (0.0 ≤ β ≤ 0.19), radiation parameter (0.1 ≤ R ≤ 0.7), Prandtl number (0.5 ≤ Pr ≤ 0.8), Brownian motion parameter (0.1 ≤ Nb ≤ 0.7), thermophoretic parameter (0.2 ≤ Nt ≤ 0.8), Chemical reaction parameter (1.0 ≤ Cr ≤ 2.5) and Lewis number (1.5 ≤ Le ≤ 3.0) have investigated and are depicted through plots. Moreover, the values of the Skin-friction coefficient, Nusselt number, and Sherwood numbers are also computed and are shown in tables. The sequels of this analysis reviewed that the values of Skin-friction coefficient and Sherwood number intensified with hiked values of Deborah number (β), whereas, the values of Nusselt number decelerate as values of (β) improves.  相似文献

The principle aim of this article is to detect the effects of externally applied magnetic force in the nanofluid flow, flowing between two co-axially rotating and expanding disks where the upper disk is continuously moving vertically upward and downward. Also, the modified Hall Effect has been considered as an effective factor of the flow. The lower disk is vertically static. The rotation and vertical motion of the disks create a three-dimensional flow of nanofluid. Heat and mass density along with the motion of the flow has been analyzed under the variation in magnetic and Hall parameters. The findings have been compared with the results in Von Karman flow of nanofluid between two rotating and stretching disks. The velocity components have been largely influenced by magnetic and Hall parameters in case of downward movement of the upper disk. The fluid temperature is detected higher in case of upward velocity and lower in case of the downward motion of the upper disk. The heat transferability of the disks is effected differently at two different disks with the influence of magnetic force and Hall effect.  相似文献
