牙鲆淋巴囊肿病的PCR诊断方法研究 总被引:14,自引:0,他引:14
以中国养殖牙鲆 (Paralichthysolivaceus)淋巴囊肿病毒 (Lymphocystisdiseasevirus,LCDVcn)主要衣壳蛋白 (majorcapsidprotein ,MCP)基因的中间保守序列为目标基因 ,设计了一对特异性引物。该引物可扩增出 172bp的病毒DNA片段 ,其最小DNA检出量为 0 0 183ng。用PCR法从人工感染淋巴囊肿病毒 3天的牙鲆血、鳃、肝、脾、肠、胃及自然发病牙鲆的肿瘤中 ,分别检测到了LCDV的存在。本实验结果证明 ,PCR法对于早期检测LCDV是十分有效的。 相似文献

During the years 1986–1989 The Finnish Work Environment Fund initiated a multicenter, intervention study (programme) to prevent musculoskeletal disorders at work. The programme consisted of four different projects comprising methodological, epidemiological, and interventive themes. The main results were as follows: (1) More than half of the occupational physiotherapists and physicians involved in the project felt that they did not have adequate training for testing the performance capacity of the musculoskeletal system. Several of the most commonly used musculoskeletal tests had either poor validity in predicting future musculoskeletal troubles or had poor reliability. (2) A controlled intervention study showed that active training of neck-shoulder muscles can prevent musculoskeletal troubles, even more than ergonomic actions. (3) It is possible to reduce sick-leave due to low-back disorders by intervention measures directed toward both the work (environment, tools) and the workers (working methods, positive attitude to work, preparedness to keep fit). (4) The identification of ergonomic hazards with the help of teamwork proved to be feasible. The practical improvements, however, were difficult to implement in the prevailing conditions. MALDI-TOF MS profiling for microorganism detection has already been demonstrated in the 1990s, but has evolved to the first-line identification method in many laboratories just during the past five years. While this application of MALDI-TOF MS has proven its broad applicability, accuracy, robustness, and cost-effectiveness it is of particular interest to expand the capabilities of the mass spectrometric platform. Resistance detection is the most desirable further application of MALDI-TOF MS in microbiology, but maybe also the most challenging. Different approaches have been published regarding diverse antibiotic drugs and distinct microorganism classes. We recently reported the successful use of the loop-mediated isothermal amplification (LAMP) reaction for hepatitis B virus (HBV) DNA amplification and its optimal primer design method. In this study, we report the development of an integrated isothermal device for both amplification and detection of targeted HBV DNA. It has two major components, a disposable polymethyl methacrylate (PMMA) micro-reactor and a temperature-regulated optical detection unit (base apparatus) for real-time monitoring of the turbidity changes due to the precipitation of DNA amplification by-product, magnesium pyrophosphate. We have established a correlation curve (R2 = 0.99) between the concentration of pyrophosphate ions and the level of turbidity by using a simulated chemical reaction to evaluate the characteristics of our device. For the applications of rapid pathogens detection, we also have established a standard curve (R2 = 0.96) by using LAMP reaction with a standard template in our device. Moreover, we also have successfully used the device on seven clinical serum specimens where HBV DNA levels have been confirmed by real-time PCR. The result indicates that different amounts of HBV DNA can be successfully detected by using this device within 1 h. 相似文献

[Cook, 1986] and [Cadigan and Farrell, 2002]), considering herein local influence diagnostics for a statistical model where estimation involves an estimating equation in which all observations are not necessarily independent of each other. Visuo-spatial neglect (VSN) is a post-stroke condition in which a patient fails to respond to stimuli on one side of the visual
field. Using an established pencil-and-paper-based method for the assessment of VSN (the Rivermead Behavioural Inattention
Test) as a reference, a battery of computer-based hand-drawing tests is developed and shown to be effective in distinguishing
between stroke subjects with and without neglect. The novel approach adopts measurements both of the outcome and the process
by which the drawing tasks are executed. This approach provides a novel diagnostic capability which results in increased test
sensitivity, a more objective assessment and a reduction in overall evaluation time. The paper describes the development of
a binary assessment system using the computer-based acquisition and analysis of task data alongside feature selection techniques
to maximise performance.

Thermal spray process is a technique of coating manufacturing implementing a wide variety of materials and processes. This technique is characterized by up to 150 processing parameters influencing the coating properties. The control of the coating quality is needed through the consideration of a robust methodology that takes into account the parameter interdependencies, the process variability and offers the ability to quantify the processing parameter-process response relationships. The aim of this work is to introduce a new approach based on artificial intelligence responding to these requirements. A detailed procedure is presented considering an artificial neural network (ANN) structure which encodes implicitly the physical phenomena governing the process. The implementation of such a structure was coupled to experimental results of an optic sensor controlling the powder particle fusion state before the coating formation. The optimization steps were discussed and the predicted results were compared to the experimental ones allowing the identification of the control factors. 相似文献
