The ability of pulsed laser irradiations to cause damage on the biofouling organisms is recently being investigated. If this technique is employed in industries such as power generation wherein a large quantity of water is being used for the cooling purpose, many organisms other than the targeted would get affected. In this study, we have investigated the damage caused by the pulsed laser irradiations from an Nd:YAG laser (fluence 0.1J/cm(2)) for varying durations such as 2, 5, 10, 30, 60 and 300 s on two marine diatom species namely Skeletonema costatum and Chaetoceros gracilis. Upon exposure to low power laser irradiations, these diatom species showed mortalities between 52.6+/-9.3% to 97.7+/-3.1% in the case of S. costatum and 57.8+/-2.5% to 98.9+/-0.6% in the case of C. gracilis for 2 and 300 s of irradiations, respectively. The mortality increased with the increase in the duration of laser irradiation. The estimation of the chlorophyll a concentration in the irradiated samples showed a considerable reduction varying between 9.8% and 57% in C. gracilis and 3% and 70.3% in S. costatum for 2 and 300 s of irradiations, respectively. The laser-survived cells grew as the non-irradiated (control) samples. C. gracilis frustules were broken by the laser whilst, the cell materials were drained out of the frustules in the case of S. costatum. The study therefore showed that the low power pulsed laser irradiations could cause significant damage on the two species of planktonic diatoms.  相似文献

A method for automatic identification of diatoms (single-celled algae with silica shells) based on extraction of features on the contour of the cells by multi-scale mathematical morphology is presented. After extracting the contour of the cell, it is smoothed adaptively, encoded using Freeman chain code, and converted into a curvature representation which is invariant under translation and scale change. A curvature scale space is built from these data, and the most important features are extracted from it by unsupervised cluster analysis. The resulting pattern vectors, which are also rotation-invariant, provide the input for automatic identification of diatoms by decision trees and k-nearest neighbor classifiers. The method is tested on two large sets of diatom images. The techniques used are applicable to other shapes besides diatoms. Andrei C. Jalba received his B.Sc. (1998) and M.Sc. (1999) in Applied Electronics and Information Engineering from “Politehnica” University of Bucharest, Romania. He recently obtained a Ph.D. degree at the Institute for Mathematics and Computing Science of the University of Groningen, where he now is a postdoctoral researcher. His research interests include computer vision, pattern recognition, image processing, and parallel computing. Michael Wilkinson obtained an M.Sc. in astronomy from the Kapteyn Laboratory, University of Groningen (RuG) in 1993, after which he worked on image analysis of intestinal bacteria at the Department of Medical Microbiology, RuG. This work formed the basis of his Ph.D. at the Institute of Mathematics and Computing Science (IWI), RuG, in 1995. He was appointed as researcher at the Centre for High Performance Computing (also RuG) working on simulating the intestinal microbial ecosystem on parallel computers. During that time he edited the book “Digital Image Analysis of Microbes” (John Wiley, UK, 1998) together with Frits Schut. After this he worked as a researcher at the IWI on image analysis of diatoms. He is currently assistant professor at the IWI. Jos B.T.M. Roerdink received his M.Sc. (1979) in theoretical physics from the University of Nijmegen, the Netherlands. Following his Ph.D. (1983) from the University of Utrecht and a 2-year position (1983--1985) as a Postdoctoral Fellow at the University of California, San Diego, both in the area of stochastic processes, he joined the Centre for Mathematics and Computer Science in Amsterdam. There he worked from 1986-1992 on image processing and tomographic reconstruction. He was appointed associate professor (1992) and full professor (2003), respectively, at the Institute for Mathematics and Computing Science of the University of Groningen, where he currently holds a chair in Scientific Visualization and Computer Graphics. His current research interests include biomedical visualization, neuroimaging and bioinformatics. Micha Bayer graduated from St. Andrews University, Scotland, with an M.Sc. in Marine Biology in 1994. He obtained his Ph.D. in Marine Biology from there in 1998, and then followed this up with two postdoctoral positions at the Royal Botanic Garden Edinburgh, Scotland, first on the ADIAC and then on the DIADIST project. In both of these projects he was responsible for establishing the collections of diatom training data to be used for the pattern recognition systems. From 2002–2003 he was enrolled for an M.Sc. in information technology at the University of Glasgow, Scotland, and is now working as a grid developer at the National e-Science Centre at Glasgow University. Stephen Juggins is a senior lecturer at the School of Geography, Politics and Sociology, University of Newcastle. His research focuses on the use of diatoms for monitoring environmental change and on the analysis of ecological and palaeoecological data. He has worked in Europe, North America and Central Asia on problems of river water quality, historical lake acidification, coastal eutrophication and Quaternary climate change.  相似文献

In this paper we present a multi-scale method based on mathematical morphology which can successfully be used in pattern classification tasks. A connected operator similar to the morphological hat-transform is defined, and two scale-space representations are built. The most important features are extracted from the scale spaces by unsupervised cluster analysis, and the resulting pattern vectors provide the input of a decision tree classifier. We report classification results obtained using contour features, texture features, and a combination of these. The method has been tested on two large sets, a database of diatom images and a set of images from the Brodatz texture database. For the diatom images, the method is applied twice, once on the curvature of the outline (contour), and once on the grey-scale image itself.  相似文献

The nanoporous structure of a membrane varies in a 3-dimensional (3-D) space and has remarkable influences on the filtration or desalination achieved, fouling potentials and therefore, the quality of yielded water. Knowledge of the 3-D nanoporous structure is thus vital to understanding and predicting its performance. A novel method by incorporating transmission electronic microtomography, image processing and 3-D reconstruction is introduced to characterize membranes with nano structures. The reconstruction algorithm allows for the visualization of 3-D nanoporous structure in a non-destructive way from any directions. This novel technique leads to in-depth understanding and accurate prediction of filtration performance.  相似文献

We demonstrate the utility of focused ion beam scanning electron microscopy combined with energy dispersive x-ray spectrometry for 3D morphological and elemental correlative analysis of subcellular features. Although recent advances in super-resolution light microscopy techniques and traditional transmission electron microscopy methods can provide cellular imaging at a wide range of length scales, simultaneous 3D morphological and elemental imaging of cellular features at nanometre scale can only be achieved with techniques such as focused ion beam scanning electron microscopy with energy dispersive x-ray spectrometry capability. We demonstrate the technique by analysing the 3D silicon cell wall structure of a marine diatom, Thalassiosira pseudonana. This study also highlights the limitations of the technique in its current state and suggests several possible improvements needed for the routine use of the technique for biological specimens.  相似文献

The establishment of dreissenid mussels in the Great Lakes has been implicated in causing a resurgence of benthic macroalgae, notably Cladophora glomerata (L.) Kützing. The seasonal proliferation and nutrient retention of epiphyton on Cladophora may be important to littoral zone processes. In this study, peak diatom epiphyte density (601 diatoms μg− 1 dry mass) co-occurred with peak Cladophora biomass. The exceptionally adhesive Cocconeis pediculus came to dominate the epiphyton to the near exclusion of all other diatoms by early summer, indicating powerful constraints on the epiphyte assemblage. There was a strong relationship between epiphytic silica content and epiphyte abundance, but the relationship was different between seasons. In the spring and fall when Cladophora was growing vigorously, silica content of the diatom epiphytes was 20.1–25.6 pmol Si diatom− 1. In the summer, during Cladophora senescence, silica content was 6.5–10.0 pmol Si diatom− 1 and valves were visibly thinner. These observations suggest that diatom epiphytes may be limited by Si during peak Cladophora biomass. Areal estimates of silica content of Cladophora epiphyton was strongly related with Cladophora biomass: at 2 m depth, silica content increased from 22.7 mmol Si m− 2 in the spring to 490 mmol Si m− 2 during peak Cladophora biomass. Silica content can be a valuable proxy for diatom epiphyte abundance if the vitality of the Cladophora substrate is considered. Future work needs to assess the seasonality of pelagic versus benthic silica demand and the interannual variability of epiphyte silica content to assess how changes in Cladophora biomass may affect nearshore Si cycling.  相似文献

In this study, centric type diatom frustules obtained from a diatomaceous earth filter material were used as filler in an epoxy resin with a weight percentage of 15% in order to assess the possible effects on the compressive behavior at quasi-static and high strain rates. The high strain rate testing of frustule-filled and neat epoxy samples was performed in a split-Hopkinson pressure bar (SHPB) set-up and modeled using the commercial explicit finite element code LS-DYNA 970. Result has shown that 15% frustule filling of epoxy increased both modulus and yield strength values at quasi-static and high strain rates without significantly reducing the failure strain. Microscopic observations revealed two main deformation modes: the debonding of the frustules from the epoxy and crushing/fracture of the frustules. The modeling results have further confirmed the attainment of stress equilibrium in the samples in SHPB testing following the initial elastic region and showed good agreement with the experimental stress–time response and deformation sequence of the samples in high strain rate testing.  相似文献

Diatom silica microparticles were chemically modified with self-assembled monolayers of 3-mercaptopropyl-trimethoxysilane (MPTMS), 3-aminopropyl-trimethoxysilane (APTES) and n-(2-aminoethyl)-3-aminopropyl-trimethoxysilane (AEAPTMS), and their application for the adsorption of mercury ions (Hg(II)) is demonstrated. Fourier transform infrared spectroscopy and x-ray photoelectron spectroscopy analyses revealed that the functional groups (–SH or –NH2) were successfully grafted onto the diatom silica surface. The kinetics and efficiency of Hg(II) adsorption were markedly improved by the chemical functionalization of diatom microparticles. The relationship among the type of functional groups, pH and adsorption efficiency of mercury ions was established. The Hg(II) adsorption reached equilibrium within 60 min with maximum adsorption capacities of 185.2, 131.7 and 169.5 mg g−1 for particles functionalized with MPTMS, APTES and AEAPTMS, respectively. The adsorption behavior followed a pseudo-second-order reaction model and Langmuirian isotherm. These results show that mercapto- or amino-functionalized diatom microparticles are promising natural, cost-effective and environmentally benign adsorbents suitable for the removal of mercury ions from aqueous solutions.  相似文献
