86 guidelines for shiftworkers were assembled by a team of European experts and published as the Bulletin of Shiftwork Topics No. 3. Before publication, 24 of the personally directed guidelines were tested against the normal actions of two groups of mixed sex industrial 3-shift continuous workers (n = 120). Six of the guidelines were endorsed as their practice by a majority of both groups; six were opposed by a majority of both groups; and twelve were intermediate. The six that were opposed are closely examined. Half of them fall into specific remedies for sleep problems, that were more abruptly phrased in the questionnaire than in the guidelines. The other three are concerned with eating and drinking: breaking your sleep to join in a family meal is only a gentle suggestion in the guidelines, and not a sensible general recommendation for day-sleepers; avoiding fatty foods may only be appropriate if you have digestive problems; and avoiding coffee and tea in the last two hours before sleep appears to oppose and lose out to work and home pressures. An evaluated intervention is required to check these points.  相似文献

The cytochrome P540 (CYP) superfamily currently includes about 9000 proteins forming more than 800 families. The enzymes catalyze monooxygenation of a vast array of compounds and play essentially two roles. They provide biodefense (detoxification of xenobiotics, antibiotic production) and participate in biosynthesis of important endogenous molecules, particularly steroids. Based on these two roles, sterol 14/*alpha*/-demethylases (CYP51) belong to the second group of P450s. The CYP51 family, however, is very special as its members preserve strict functional conservation in enzyme activity in all biological kingdoms. At amino acid identity across the kingdoms as low as 25-30%, they all catalyze essentially the same three-step reaction of oxidative removal of the 14/*alpha*/-methyl group from the lanostane frame. This reaction is the required step in sterol biosynthesis of pathogenic microbes. We have shown that specific inhibition of protozoan CYP51 can potentially provide treatment for human trypanosomiases. Three sets of CYP51 inhibitors tested in vitro and in trypanosomal cells in this study include azoles [best results being 50% cell growth inhibition at <1 and at 1.3 muM for Trypanosoma cruzi (TC) and Trypanosoma brucei (TB), respectively], non-azole compounds (50% TC cell growth inhibition at 5 microM) and substrate analogs of the 14/*alpha*/-demethylase reaction. 32-Methylene cyclopropyl lanost-7-enol exhibited selectivity toward TC with 50% cell growth inhibition at 3 microM.  相似文献

Sleeping Beauty (SB) is the first DNA transposon employed for efficient transposition in vertebrate cells, opening new applications for genetic engineering and gene therapies. A transposon-based gene delivery system holds the favourable features of non-viral vectors and an attractive safety profile. Here, we employed SB to engineer HEK293 cells for optimizing the production of a chimpanzee Adenovector (chAd) belonging to the Human Mastadenovirus C species. To date, chAd vectors are employed in several clinical settings for infectious diseases, last but not least COVID-19. A robust, efficient and quick viral vector production could advance the clinical application of chAd vectors. To this aim, we firstly swapped the hAd5 E1 with chAd-C E1 gene by using the CRISPR/Cas9 system. We demonstrated that in the absence of human Ad5 E1, chimp Ad-C E1 gene did not support HEK293 survival. To improve chAd-C vector production, we engineered HEK293 cells to stably express the chAd-C precursor terminal protein (ch.pTP), which plays a crucial role in chimpanzee Adenoviral DNA replication. The results indicate that exogenous ch.pTP expression significantly ameliorate the packaging and amplification of recombinant chAd-C vectors thus, the engineered HEK293ch.pTP cells could represent a superior packaging cell line for the production of these vectors.  相似文献

Abstract

A questionnaire was performed in order to investigate the effect of naps during duty on recovery from fatigue. Studies were performed on 12 male computer operators in each of two chemical plants working a four-team three-shift system. In one of the plants, the shift workers were able to take a 2 h nap during the night shift (nap group). In the other plant, no nap was taken (no-nap group). Before and after the two consecutive night shifts, both the nap and no-nap groups greatly extended their night sleeps, but the daytime sleep taken by the no-nap group during this period was significantly longer than that of the nap group on both the first and second days. No significant difference was found when comparing the length of the day sleep of the no-nap group with the total sleeping time (night-time nap plus subsequent day sleep) of the nap group. Therefore a night-time nap enables part of the essential sleep to be taken in advance of the day sleep following night work. During night work, both tbe nap and no-nap groups exhibited an increase in the sleepiness scores and also in the subjective feelings of fatigue concerning the complaints related to drowsiness, dullness and difficulty in concentration. However, it was found that for the no-nap group these effects continued for a large part of the recovery period following night work. It can therefore be surmised that naps taken during night-time work can be to a certain extent aid recovery from the fatigue caused by that work.  相似文献

Ventilation is important because it affects indoor environmental conditions, including air pollutant concentrations that may modify the health of the occupants of a building, or their perceptions and comfort. This paper reports, first of all, on field studies monitoring indoor overnight CO2 levels and outdoor ventilation rates in bedrooms employing room air conditioners (RACs), so the current situation of ventilation in actual high-rise residential buildings in Hong Kong can be appreciated. This is followed by a report of the results of laboratory experiments where two typical RACs were used in an examination of outdoor air ventilation characteristics in rooms employing RACs. The results of field studies showed that the outdoor ventilation rates in the measured bedrooms equipped with RACs in high-rise residential buildings in Hong Kong could not meet the ventilation requirement specified in the ASHRAE standard 62-2001 even if there was only one occupant in a bedroom. Although the use of a window-type air conditioner (WRAC) may provide a higher outdoor ventilation rate than the use of a split-type air conditioner (SARC), this may be ascribed to the fact there is more natural infiltration when a WRAC is used. The ventilation damper currently available in a WRAC does not significantly affect the outdoor ventilation rate. Therefore, such a damper cannot be expected to provide the ventilation rate as required by a ventilation code and its intended function of controlling ventilation is limited. In addition, the air exhausted from indoors to outdoors through the ventilation outlet in a WRAC is air that has just been cooled by the cooling coil (evaporator). This is unreasonable, because exhausting just-cooled and dehumidified air is a waste of energy. Therefore, an improved design for a WRAC has been suggested. Finally, the outdoor ventilation requirement for bedrooms at nighttime, when occupants are asleep, is discussed. A new ventilation rate of 3.0 l/s per person for the sleeping environment in high-rise residential buildings is proposed.  相似文献

该文针对无线传感器网络的覆盖性和连通性问题,在假设传感器节点地理位置信息已知的条件下,设计了一种包含全连通群的建立和维护以及群内节点休眠调度的全新算法。该算法采用保证群内节点彼此一跳可达的全连通群分群方法,以及分布式节能的休眠调度策略,最大程度上减少传感器网络的能量消耗,延长了网络寿命。仿真结果表明:该算法能较好地保证无线传感器网络的覆盖性和连通性,且能耗较低。  相似文献

In the subtropics, air conditioning serves to maintain an appropriate indoor thermal environment not only in workplaces during daytime, but also at night for sleeping in bedrooms in residences or guestrooms in hotels. However, current practices in air conditioning, as well as the thermal comfort theories on which these practices are based, are primarily concerned with situations in which people are awake in workplaces at daytime. Therefore, these may not be directly applicable to air conditioning for sleeping environments. This paper, reports on a theoretical study on a thermal comfort model in sleeping environments. A comfort equation applicable to sleeping thermal environments was derived by introducing appropriate modifications to Fanger's comfort model. Comfort charts which were established by solving the comfort equation, and can be used for determining thermally neutral environmental conditions under a given bedding system have been developed. A related paper reports on an experimental study on measuring the total thermal insulation values of a wide range of bedding systems commonly used in the subtropics, which are an essential input to the comfort equation developed and reported in this paper.  相似文献
