This paper summarizes studies on the presence of acid-fast and yeast organisms in wastewater and water treatment plants and in surface water. These organisms were found to satisfy three of Bonde's criteria for indicator organisms: presence whenever pathogens are likely to be present; resistance at least equal to that of pathogens; and lack of regrowth in the post-treatment environment. This, plus prior data, indicates that these organisms are at least as acceptable as indicators of disinfection efficiency than the coliform group. 相似文献

Of 100 gas-producing bacterial isolates, 65% were Gram-negative. Bacterial growth and gas production in commercial tofu was accompanied by acidification, and pH decreases as low as 5.2. Coliform bacteria constituted 15% of the total bacterial flora. Sporeformers and yeast counts remained < 103 CFU/mL. Pasteurization was effective in lowering the bacterial counts, and retarded gas production. The addition of lactic acid (reducing pH to 5.5) or lactic acid bacteria (2 × 107 CFU/g) to tofu helped reduce gas production by about 50%. Addition of both lactic acid and Lactobacillus plantarum stopped gas production in contaminated tofu. 相似文献

Escherichia coli distribution and persistence in nearshore Lake Michigan were assessed following heavy rains and sanitary sewer overflow (SSO) and combined sewer overflow (CSO) events over a 5-year period, including an 18-day period following 25.4 cm of rainfall in which intensive studies were conducted following multiple CSO and SSO events. E. coli levels in the Milwaukee estuary and harbor following SSO and CSO events ranged from 104 to nearly 105 CFU/100 mL, which were significantly higher (p ≤ 0.05) than levels following rainfall alone. Sites outside of the breakwall but within the contamination plume (e.g., within 2 km of the harbor) were an order of magnitude lower. Locations 2–5 km from the harbor ranged from below detection limits, of < 1 to 5 CFU/100 mL. E. coli levels corrected for dilution based on specific conductivity measurements were lower than what would be expected for loss due to dilution alone, suggesting a combination of die-off and dilution, were responsible for the rapid disappearance of these organisms outside of the harbor. E. coli and fecal coliforms measured concurrently demonstrated that fecal coliforms could be recovered longer than E. coli in the open waters of the lake. E. coli isolated directly from sewage treatment plant influent were found to have a marked increase in antibiotic resistance traits for ten antibiotics commonly used in the human population compared with isolates from two animal sources of fecal pollution. However, E. coli obtained from sewage impacted water (n = 2,513) and from stormwater impacted water (n = 1,465) collected the previous year when there were no sewage overflows, were found to have no significant difference (p < 0.05) in the frequency of resistance when comparing the two conditions. E. coli survival characteristics and population dynamics are most likely influenced by multiple factors in complex systems such as the watershed/estuarine/lake environments of the Great Lakes. 相似文献

A limited number of bacteria can convert oil palm (Elaeis guineensis) sap to hydrogen with satisfactory yield and productivity. In this study, a total of 18 fermentative enriched cultures and 36 newly isolated thermotolerant bacterial strains were compared for hydrogen production from oil palm (OP) sap. The new isolates were obtained from hot springs, palm oil mill effluent and oil palm sap. The test was conducted in three steps: (i) a test for hydrogen production from mixed substrates (cellulose, starch, xylose, and glucose) and OP sap; (ii) a test for substrate concentration tolerance; and (iii) a test for thermotolerance. Five enriched candidates for each of the hydrogen producers were selected according to the criteria defined for the screening test. The hydrogen production of these selected bacterial strains from hot springs were cultivated in batch fermentation of oil palm sap at room temperature (30 ± 2 °C). Five enriched cultures, namely 81RN1, OPS, 85RN5, 89SR3-2 and 112YL1 were found to give high cumulative hydrogen formation of 1085, 1009, 994, 983 and 778 mL H2/L-OP sap, respectively, with the hydrogen content of 29.8, 29.4, 28.7, 27.1 and 27.5%, respectively. PCR–DGGE profiling showed that all these five enriched cultures consisted of species closely related to the genus Clostridium sp. based on the 16S rRNA gene. For pure cultures, the top five hydrogen producers were the isolates encoded as PS-3, PS-4, PS-5, PS-7 and PS-8 exhibiting the hydrogen production of 1973, 1774, 1335, 1170 and 1070 mL H2/L-OP sap, respectively, with the hydrogen content of 33.7, 29.6, 32.5, 31.5 and 26.4%, respectively. Identification of these high hydrogen producers using 16S rRNA sequence matching showed that the isolates PS-3 and PS-8 belonged to Clostridium beijerinckii, while the isolate PS-7 belonged to Clostridium acetobutylicum and the isolates PS-4 and PS-5 belonged to Klebsiella sp. and Klebsiella pneumoniae, respectively. Therefore, the pure culture C. beijerinckii PS-3 exhibited 1.8 folds higher hydrogen production (1973 mL H2/L-OP sap) than the enriched cultures of 81RN1 (1085 mL H2/L-OP sap). 相似文献

Thirty-nine purple non-sulfur bacteria (PNSB) were isolated from 162 environmental samples. These isolates of PNSB were tested for their ability to produce biohydrogen (Bio-H2) at 40 °C and only 14 isolates were noted to possess such ability, which were considered as thermotolerant photosynthetic Bio-H2 producing bacteria (tt-PHPB). Of 14 isolates of tt-PHPB, KKU-SN1/1 was observed to have the highest Bio-H2 productivity. Central composite design was employed to optimize the operating conditions for maximal Bio-H2 production by the strain KKU-SN1/1. Under optimal conditions (7.6 g/L malic acid, 11 g/L glutamic acid, pH 6.7, at 40 °C) the Bio-H2 production was increased by 68.28%. The purity of Bio-H2 produced was 92.22%, as suggested by gas chromatography (GC-TCD). Based on morphological characteristics and 16S rDNA sequence analysis, the strain KKU-SN1/1 was identified as Rhodopseudomonas pentothenatexigens, with 99.7% similarity. This study was aimed at assessing the prevalence of pathogenic Escherichia coli strains, relative to the total count of E. coli, faecal coliforms and other heterotrophic mesophilic aerobic bacteria (HMAB) isolated in groundwater in the equatorial region of Cameroon (Central Africa). Bacteria were isolated using standard methods. Pathogenic E. coli strains were then identified using haemagglutination and antisera tests. The maximum abundance of HMAB, faecal coliforms and E. coli strains were 4.9 × 106, 5.6 × 103 and 1 × 103 colony‐forming units (CFU)/100 mL, respectively. The count of pathogenic E. coli strains reached 3 CFU/100 mL. The counts of commensal and pathogenic E. coli strains underwent temporal and spatial fluctuations. In 21% of sampling sites, the abundance of faecal coliforms was significantly correlated to that of E. coli (P < 0.05). However, the isolated bacterial count was not significantly correlated to that of the pathogenic E. coli strains (P > 0.05). Permeable pavement systems (PPS) are frequently associated with high removal efficiencies for water quality parameters. Their effluent can, therefore, be recycled, for example, for sprinkling onto gardens. Nevertheless, some stakeholders fear that potentially pathogenic organisms within the treated run‐off could be too high, and therefore they request disinfection before recycling. The aim of this paper is, therefore, to assess the efficiency of a batch flow combined titanium dioxide (TiO2) and ultraviolet (UV) light photocatalytic reactor in removing water‐borne microbial contaminants from the effluent of PPS. Combined TiO2 and UV photocatalytic reaction times between 80 and 100 min were required for the complete removal of Escherichia coli, total coliforms and faecal Streptococci, which had mean initial counts of 1.5 × 107, 4.4 × 106 and 6.9 × 105 colony‐forming units (CFU) per 100 mL, respectively. The distribution of bacterial contaminants (Escherichia coli and total coliforms) and nutrients (nitrogen and phosphorus) in a rapidly growing metropolitan area, the Las Vegas Valley, were studied to assess the contributions of point and nonpoint pollution sources under dry weather flow (pre rainfall) and wet weather flow (post rainfall) events. Nonpoint sources emanating from urban centers and septic seepage were found to be a primary source of bacterial contamination in the Las Vegas Wash, a mainstream wash channel where the Las Vegas Valley watershed drains. Contribution of point sources (wastewater treatment plants) to E. coli and total coliforms were found to be negligible. However, data in the Tropicana/Flamingo Wash, a tributary of the Las Vegas Wash, showed high bacterial activities in the wet antecedent rainfall condition as compared to the pre-rain dry antecedent condition suggesting that the rainfall increased bacterial activities. 相似文献

为了缩短大肠菌群的检测时间,满足蔬菜、水果等易腐食品的检测需要,建立了一种利用分光光度法快速、准确检测大肠菌群数的新方法。该方法建立在传统的大肠菌群检测方法的基础上,对传统多管发酵法进行改进,将制得的伊红美兰混合液加入到乳糖胆盐培养基中作指示剂,通过其产生沉淀后溶液颜色的变化来指示大肠菌群的数量,2.5h即可得到检测结果。结果表明,向1mL乳糖胆盐培养基中加入60μL伊红美兰溶液时指示效果最好;在37℃培养2.5h后显色稳定,检测结果重现性好,精密度相对标准偏差小于2.75%。 相似文献
