AQP3 Increases Intercellular Cohesion in NSCLC A549 Cell Spheroids through Exploratory Cell Protrusions

Tumor cell aggregation is critical for cell survival following the loss of extracellular matrix attachment and dissemination. However, the underlying mechanotransduction of clustering solitary tumor cells is poorly understood, especially in non-small cell lung cancers (NSCLC). Here, we examined whether cell surface protrusions played an important role in facilitating the physical contact between floating cells detached from a substrate. We employed poly-2-hydroxyethyl methacrylate-based 3D culture methods to mimic in vivo tumor cell cluster formation. The suprastructural analysis of human NSCLC A549 cell spheroids showed that finger-like protrusions clung together via the actin cytoskeleton. Time-lapse holotomography demonstrated that the finger-like protrusions of free-floating cells in 3D culture displayed exploratory coalescence. Global gene expression analysis demonstrated that the genes in the organic hydroxyl transport were particularly enriched in the A549 cell spheroids. Particularly, the knockdown of the water channel aquaporin 3 gene (AQP3) impaired multicellular aggregate formation in 3D culture through the rearrangement of the actomyosin cytoskeleton. Moreover, the cells with reduced levels of AQP3 decreased their transmigration. Overall, these data indicate that cell detachment-upregulated AQP3 contributes to cell surface protrusions through actomyosin cytoskeleton remodeling, causing the aggressive aggregation of free-floating cells dependent on the property of the substratum and collective metastasis.     

Collective reflection strategy for moderating conformity tendency and promoting reflective judgment performance

Reflective judgement is crucial for medical-related practitioners in dealing with controversial issues. However, the conformity phenomenon is likely to occur and interfere with reflective judgement learning during interactive activities. Effective strategies are required to moderate the conformity behaviour tendency (CBT) and improve reflective judgement performance (RJP). This study demonstrates two significant results: (a) Compared with the guided self-reflection learning strategy, the online collective reflection (OCR) learning strategy effectively weakened the learners' general CBT while dealing with professional controversial issues; and (b) a significantly negative correlation between the RJP achieved and the change of CBT in online environment was detected in the OCR group. The implications and potential applications in higher education were discussed. Further studies are needed to confirm the long-term effects and the extending application to other professional studies.

Their article also posed a danger that mirrors a disturbing trend in the public discourse about conflicts and injustice. In response to the recent escalation of international violence and years of mixed results of engagement in debates centered around grievances of injustice (such as the Palestinian-Israeli conflict), many politicians, pundits, and scholars have advocated addressing conflicts by separating them from the underlying grievances and the ongoing injustice. Reciprocal Relationships Between Collective Efficacy and Team Performance in Women's Ice Hockey.

This study examined reciprocal relationships between collective efficacy and team performance over a season of competition in women's intercollegiate ice hockey within weekends where the opponent was constant for 2 games. Collective efficacy beliefs within 12 teams were assessed prior to both games for at least 7 weekends. Team performance indexes produced an overall measure of performance for each game. The average influence of Saturday collective efficacy on Saturday performance was moderate and positive after controlling for Friday performance. The average influence of Friday performance on Saturday collective efficacy was small and positive after removing the influence of Friday collective efficacy from Friday performance. 大港油田官109-1断块稠油油藏碱/表面活性剂吞吐采油技术

大港油田官109-1断块稠油油藏的油井由于井筒、近井地带发生原油有机沉积,储层原油流动性差,采用多种方法都不能正常生产。为此研制了一种碱性解堵液,用于油井化学吞吐。该解堵液为复配碱和抗硬剂等添加剂的10%两性木质素表面活性剂溶液,能使玻璃片表面由亲油变强亲水,在石英上的接触角为6°(稀释至1/2和1/4浓度时为11°和12°),与羊大站原油间的界面张力为1.75×10-3~3.18×10-3mN/m。该解堵液与黏度1~50 Pa.s的不同区块稠油按体积比1∶1混合形成的乳状液,60℃、175 s-1黏度为2~6 mPa.s,在配液水矿化度高达6000mg/L以上或钙镁离子浓度高达250 mg/L时,该解堵液仍能保持对稠油的乳化降黏能力。该解堵液从实验岩心中驱出的原油量比地层水多75%,驱出液为乳状液。通过一个井例介绍了化学吞吐工艺:先用一种降黏解堵液对油井进行预处理,清洗油管、炮眼及近井地带,再注入碱性解堵液(该井例中为500 m3),注入时间约15小时,然后开井生产。2000年2月至2002年6月在16口新投产井实施化学吞吐(第一轮次),15口井有效,其中2口井先后转注水,其余13口井到2005年底仍继续产油。图1表6参5。
