Development and monitoring of an integrated bioprocess for production of a potential malaria vaccine with Pichia pastoris

An engineered variant of the yeast Pichia pastoris was used to produce the potential malaria vaccine D1M1. This paper describes the implementation of an expanded bed adsorption chromatography step into a sequential cultivation process of the yeast in order to link protein expression, cell release, and product capture of the secreted protein in a fully automated plant. The process is operated with a semi-continuous strategy for an integrated production. Process improvements have been achieved using design of experiments. Resulting purities of product achieved up to 87% with recoveries of 51% in a single downstream operation. Furthermore, a multivariate analysis of historical batch data was used to develop a golden batch model for process monitoring.     

On-line H NMR spectroscopic investigation of hydrogen bonding in supercritical and near critical CO2–methanol up to 35 MPa and 403 K

On-line NMR spectroscopy can beneficially be applied to studies of supercritical and near-critical fluids as an alternative to optical spectroscopy. Up to now high pressure NMR experiments are predominantly accomplished using custom made NMR batch reactors. The authors present a novel high pressure cell with displacement plunger for on-line NMR experiments on compressible fluids which can be used in conjunction with commercially available SCF NMR flow probes. The on-line technique offers advantages compared to stopped flow techniques such as enhanced control of mixture composition and reaction parameters as well as the facility of engagement into the reaction. The new apparatus is used for NMR studies on hydrogen bonding of methanol in near critical and supercritical carbon dioxide up to 403 K and 35 MPa for which data on the chemical shift of the hydroxyl group and methyl group are reported and interpreted.     

Microwave assisted ultrafast synthesis of graphene oxide based magnetic nano composite for environmental remediation

We have successfully synthesized multi-layer graphene oxide and graphene oxide based magnetic nanocomposite (M/GO) by microwave-assisted modified Hummers’ method for removal of toxic lead (Pb²⁺) and cadmium (Cd²⁺) ions from aqueous solution. The X-ray diffraction spectra of synthesized graphene oxide and M/GO confirm increased interlayer spacing along c-axis. Raman spectra revealed the good quality of synthesized GO and M/GO. The wrinkles were seen in the SEM images of synthesized graphene oxide. The presence of conjugated double bond (CC) and carbonyl (CO) were confirmed by using the UV–Vis spectroscopic spectra. Brunauer–Emmett–Teller (BET) analysis showed high (126 m²/g) surface area M/GO composite which accounts for large number of active binding sites for the adsorption of heavy metal ions. The adsorption studies revealed that Pb²⁺ ions were efficiently adsorbed on GO sheets. Interestingly, M/GO showed better adsorption for cadmium ions.

Development of modified stokes expression to model the behavior of expanded beds containing polydisperse resins for protein adsorption

Expanded bed behavior was modeled by using the Richardson-Zaki correlation between the superficial velocity of the feed stream and the void fraction of the bed. A polydisperse material, Chelating excellose® (70-210 Μm in diameter, 1.21 g/cm³ in density), which has Ni²⁺ ions for the selective binding of histidine-tagged proteins, was used as the resin. A method to modify the Stokes expression to express the terminal settling velocity of the resins by introducing two empirical parameters, the effective diameter of the resins and an exponent for(ρ _p -ρ)/Μ term, was developed. Combined use of the Richardson-Zaki correlation and the modified Stokes expression was successful in modeling the bed expansion by incorporating physical properties of feed streams and the resins.     

The Study of Plasma Nitriding of AISI304 Stainless Steel

This paper presents results on the plasma nitriding of AISI 304 stainless steel at different temperatures in NH3gas. The working pressure was 100～200 Pa and the discharge voltage was 700～800V. The phase of nltrided layer formed on the surface was confirmed by X-ray diffraction. The hardness of the samples was measured by using a Vickers microhardness tester with the load of 50g. After nitriding at about 400℃ for two hours a nitrided layer consisting of single γN phase with thickness of 51.tm was obtained. Microhardness measurements showed significant increase in the hardness from 240 HV (for untreated samples) up to 950 HV (for nltrided samples at temperature of 420℃). The phase composition, the thickness, the microstructure and the surface topography of the nltrided layer as well as its properties depend essentially on the process parameters.     

月桂酸/膨胀珍珠岩复合相变材料的制备与热性能研究

以月桂酸为相变材料,膨胀珍珠岩为载质,利用真空吸附法制备出月桂酸/膨胀珍珠岩复合相变材料(LA/EP-PCMs)。通过FT-IR、SEM、DSC、TGA对LA/EP-PCMs的微观结构、相变温度、相变潜热、热稳定性进行表征。结果表明:月桂酸能较好地吸附在膨胀珍珠岩孔隙内,它们之间的化学相容性良好。LA/EP-PCMs中月桂酸饱和含量为65%,此时其相变温度为41.3℃,热焓为110.1J/g。将5%的纳米石墨纤维(NGF)作为添加剂加入到LA/EP-PCMs中,其导热系数由0.09 W/(m·K)提高到0.16 W/(m·K),增长了77.7%。熔融凝固实验表明:掺入NGF将改善复合相变材料的蓄放热能力,其强化导热机理是在相变基体外表面和内部形成了导热网络。
