Soybean allergy presents a health threat to humans and animals. The mechanism by which food/feed allergen β-conglycinin injures the intestinal barrier has not been well understood. In this study, the changes of epithelial permeability, integrity, metabolic activity, the tight junction (TJ) distribution and expression induced by β-conglycinin were evaluated using IPEC-J2 model. The results showed a significant decrease of trans-epithelial electrical resistance (TEER) (p < 0.001) and metabolic activity (p < 0.001) and a remarkable increase of alkaline phosphatase (AP) activity (p < 0.001) in a dose-dependent manner. The expression levels of tight junction occludin and ZO-1 were decreased (p < 0.05). The reduced fluorescence of targets and change of cellular morphology were recorded. The tight junction occludin and ZO-1 mRNA expression linearly declined with increasing β-conglycinin (p < 0.001). 相似文献

Phytate-removed and deamidated soybean β-conglycinin (PrDS) prepared by ion-exchange resins was supplemented to be 4% in the diet administered to ovariectomized rats to investigate its preventive effect on osteoporosis. The apparent calcium absorption rate decreased following ovariectomy and was not replenished by oral administration of phytate-removed soybean β-conglycinin (PrS) or casein. On the other hand, administration of PrDS restored the calcium absorption rate to the same level as the sham group. Markers of bone resorption, such as serum parathyroid hormone (PTH) and urinary deoxypyridinoline (DPD), increased, and the bone mineral density and breaking stress decreased following ovariectomy. However, PrDS supplementation suppressed the changes caused by the decrease in calcium absorption from the small intestine. Therefore, PrDS supplementation shows promise for the prevention of postmenopausal osteoporosis. 相似文献

Antiserum toward soybean meal was prepared by feeding a soybean meal diet containing large amounts of antigens to calves.
The antiserum was used to develop a sensitive detection method for soybean meal antigens by competitive inhibition enzyme-linked
immunosorbent assay. With this method, the effectiveness of twin-screw extrusion cooking in reducing antigenicity in soybean
meal was examined. Antigenicity was reduced to 0.1% of the original activity by extrusion cooking with screws containing kneading-disc
elements and die-end temperatures exceeding 66°C. Electrophoretic analysis of the cooked meal indicated that the reduction

Steady shear and dynamic oscillatory measurements were used to investigate the effect of concentration, ionic strength and thermal treatment on rheological properties of soybean β-conglycinin in aqueous dispersions. SDS-PAGE and Differential Scanning Calorimetry (DSC) showed that β-conglycinin exhibited partial denaturation and formation of aggregates during isolation. Under steady shear flow, strong shear-thinning behavior was observed with increasing shear rate from 0.001 to 1200 s?1. A dispersion of β-conglycinin (≥5% w/v, without applying thermal treatment) exhibited gel-like dynamic mechanical spectra at 20°C. This suggested that β-conglycinin in aqueous dispersions showed rheological properties of typical weak gel-like (entanglement) or semi diluted polymeric solution. Weak gel network of β-conglycinin was susceptible to ionic strength, suggesting that electrostatic forces play an important role in the formation of weak gel network. These properties of β-conglycinin have practical significance for the food processors in the formulation of new products. 相似文献

The hypocholesterolemic and antiatherogenic effects of rice α-globulin remain unclear. We investigated the hypocholesterolemic effect of rice α-globulin in rats fed a hypercholesterolemic diet. The rats were divided into 4 groups and were orally administrated the following three proteins or a vehicle for 4 weeks: rice protein, rice α-globulin, or soy β-conglycinin at a dose of 100 mg/kg body weight or carboxymethylcellulose to the control rats. In the rice α-globulin group, serum cholesterol concentrations were 28% lower than the control group and fecal neutral steroid excretion was increased by 30%. The hypocholesterolemic effect of rice α-globulin was equal to soy β-conglycinin in SD rats fed the hypercholesterolemic diet. However, the serum cholesterol concentrations in the rice protein group did not change compared to the control group. To investigate the antiatherogenic effects of rice α-globulin, male apolipoprotein E-deficient mice were orally administered the same dose of rice α-globulin for 9 weeks. The en face lesion area in the aorta was 46% lower than in the control group. In conclusion, administration of rice α-globulin improves hypercholesterolemia in rats fed a hypercholesterolemic diet by increasing the fecal excretion of neutral sterols, and inhibits atherosclerosis development in apolipoprotein E-deficient mice. The anti-atherosclerotic effect exerts by mechanism(s) other than the regulation of serum MCP-1 and NO concentrations. 相似文献

In legumes, the seed storage proteins accumulate within specialized organelles called protein storage vacuoles (PSVs). In several plant species, PSVs are differentiated into subdomains that accumulate different kinds of proteins. Even though the existence of subdomains is common in cereals and legumes, it has not been reported in soybean PSVs. The two most abundant seed proteins of soybean, 7S and 11S globulins, have different temporal accumulation patterns and exhibit considerable solubility differences that could result in differential accretion of these proteins within the PSVs. Here, we employed confocal fluorescent microscopy to examine the presence or absence of subdomains within the soybean PSVs. Eosin-stained sections of FAA-fixed paraffin embedded soybean seeds, when viewed by confocal fluorescence microscopy, revealed the presence of intricate subdomains within the PSVs. However, fluorescence immunolabeling studies demonstrated that the 7S and 11S globulins were evenly distributed within the PSVs and failed to corroborate the existence of subdomains within the PSVs. Similarly, confocal scanning microscopy examination of free-hand, vibratome and cryostat sections also failed to demonstrate the existence of subdomains within PSVs. The subdomains, which were prominently seen in PSVs of FAA-fixed soybean seeds, were not observed when the seeds were fixed either in glutaraldehyde/paraformaldehyde or glutaraldehyde. Our studies demonstrate that the apparent subdomains observed in FAA-fixed seeds may be a fixation artifact. 相似文献
