Sialidase cleaves sialic acid residues from glycans such as glycoproteins and glycolipids. In the brain, desorption of the sialic acid by sialidase is essential for synaptic plasticity, learning and memory and synaptic transmission. BTP3-Neu5Ac has been developed for sensitive imaging of sialidase enzyme activity in mammalian tissues. Sialidase activity in the rat hippocampus detected with BTP3-Neu5Ac increases rapidly by neuronal depolarization. It is presumed that an increased sialidase activity in conjunction with neural excitation is involved in the formation of the neural circuit for memory. Since sialidase inhibits the exocytosis of the excitatory neurotransmitter glutamate, the increased sialidase activity by neural excitation might play a role in the negative feedback mechanism against the glutamate release. Mammalian tissues other than the brain have also been stained with BTP3-Neu5Ac. On the basis of information on the sialidase activity imaging in the pancreas, it was found that sialidase inhibitor can be used as an anti-diabetic drug that can avoid hypoglycemia, a serious side effect of insulin secretagogues. In this review, we discuss the role of sialidase in the brain as well as in the pancreas and skin, as revealed by using a sialidase activity imaging probe. Some reports demonstrated successful genome editing in pigs by one-step zygote microinjection of mRNA of CRISPR/Cas9-related components. Given the relatively long gestation periods and the high cost of housing, the establishment of a single blastocyst-based assay for rapid optimization of the above system is required. As a proof-of-concept, we attempted to disrupt a gene (GGTA1) encoding the α-1,3-galactosyltransferase that synthesizes the α-Gal epitope using parthenogenetically activated porcine oocytes. The lack of α-Gal epitope expression can be monitored by staining with fluorescently labeled isolectin BS-I-B4 (IB4), which binds specifically to the α-Gal epitope. When oocytes were injected with guide RNA specific to GGTA1 together with enhanced green fluorescent protein (EGFP) and human Cas9 mRNAs, 65% (24/37) of the developing blastocysts exhibited green fluorescence, although almost all (96%, 23/24) showed a mosaic fluorescent pattern. Staining with IB4 revealed that the green fluorescent area often had a reduced binding activity to IB4. Of the 16 samples tested, six (five fluorescent and one non-fluorescent blastocysts) had indel mutations, suggesting a correlation between EGFP expression and mutation induction. Lévy flights have gained prominence for analysis of animal movement. In a Lévy flight, step-lengths are drawn from a heavy-tailed distribution such as a power law (PL), and a large number of empirical demonstrations have been published. Others, however, have suggested that animal movement is ill fit by PL distributions or contend a state-switching process better explains apparent Lévy flight movement patterns. We used a mix of direct behavioural observations and GPS tracking to understand step-length patterns in females of two related butterflies. We initially found movement in one species (Euphydryas editha taylori) was best fit by a bounded PL, evidence of a Lévy flight, while the other (Euphydryas phaeton) was best fit by an exponential distribution. Subsequent analyses introduced additional candidate models and used behavioural observations to sort steps based on intraspecific interactions (interactions were rare in E. phaeton but common in E. e. taylori). These analyses showed a mixed-exponential is favoured over the bounded PL for E. e. taylori and that when step-lengths were sorted into states based on the influence of harassing conspecific males, both states were best fit by simple exponential distributions. The direct behavioural observations allowed us to infer the underlying behavioural mechanism is a state-switching process driven by intraspecific interactions rather than a Lévy flight. 相似文献

以废弃的流化催化裂化催化剂(简称SFCC)为载体、β-环糊精为金属络合剂、硝酸镍为镍源,采用湿法浸渍法制备β-环糊精修饰的Ni/SFCC催化剂(简称Ni/SFCC-CD催化剂),考察其对C9石油树脂的催化加氢性能。通过BET比表面积测试、H2程序升温还原、X射线光电子能谱等手段对催化剂的物相结构进行表征,研究β-环糊精的作用机理及其对催化剂加氢性能的影响。研究结果表明:在反应温度为260 ℃、反应压力为7 MPa、反应时间为2.0 h的最优条件下,采用Ni/SFCC-CD催化C9石油树脂加氢,可制得溴值为1.45 gBr/(100 g)、色号(加纳德)小于1的水白色氢化C9石油树脂,催化剂循环使用4次后仍保持良好活性;β-环糊精的作用机理是:β-环糊精与硝酸镍产生络合作用,抑制硝酸镍的分解、控制NiO的结晶过程和增强活性组分Ni与载体之间的相互作用力,从而提高了Ni/SFCC-CD的催化活性和稳定性。 相似文献

This paper presents a comparative study of the condensation heat transfer coefficients in a smooth tube when operating with pure refrigerant R134a and its mixture with lubricant Castrol “icematic sw”. The lubricant is synthetic polyol ester based oil commonly used in lubricating the compressors. Two concentrations of R134a-oil mixtures of 2% and 5% oil (by mass) were analysed for a range of saturation temperatures of refrigerant R134a between 35 °C and 45 °C. The mass flow rate of the refrigerant and the mixtures was carefully maintained at 1 g/s, with a vapour quality varying between 1.0 and 0. The effects of vapour quality, flow rate, saturation temperature and temperature difference between saturation and tube wall on the heat transfer coefficient are investigated by analysing the experimental data. The experimental results were then compared with predictions from earlier models [Int J Heat Mass Transfer (1979), 185; 6th Int Heat Transfer Congress 3 (1974) 309; Int J Refrig 18 (1995) 524; Trans ASME 120 (1998) 193].
