The foliage and fruit of the tomato plantLycopersicon esculentum contains polyphenol oxidases (PPO) and peroxidases (POD) that are compartmentally separated from orthodihydroxyphenolic substrates in situ. However, when leaf tissue is damaged by insect feeding, the enzyme and phenolic substrates come in contact, resulting in the rapid oxidation of phenolics to orthoquinones. When the tomato fruitwormHeliothis zea or the beet army-wormSpodoptera exigua feed on tomato foliage, a substantial amount of the ingested chlorogenic acid is oxidized to chlorogenoquinone by PPO in the insect gut. Additionally, the digestive enzymes of the fruitworm have the potential to further activate foliar oxidase activity in the gut. Chlorogenoquinone is a highly reactive electrophilic molecule that readily binds cova-lently to nucleophilic groups of amino acids and proteins. In particular, the —SH and —NH2 groups of amino acids are susceptible to binding or alkylation. In experiments with tomato foliage, the relative growth rate of the fruitworm was negatively correlated with PPO activity. As the tomato plant matures, foliar PPO activity may increase nearly 10-fold while the growth rate of the fruitworm is severely depressed. In tomato fruit, the levels of PPO are highest in small immature fruit but are essentially negligible in mature fruit. The growth rate of larvae on fruit was also negatively correlated with PPO activity, with the fastest larval growth rate occurring when larvae fed on mature fruit. The reduction in larval growth is proposed to result from the alkylation of amino acids/protein byo-quinones, and the subsequent reduction in the nutritive quality of foliage. This alkylation reduces the digestibility of dietary protein and the bioavailability of amino acids. We believe that this mechanism of digestibility reduction may be extrapolatable to other plant-insect systems because of the ubiquitous cooccurrence of PPO and phenolic substrates among vascular plant species.  相似文献

Settling of the potato aphid,Macrosiphum euphorbiae, on feeding membranes was deterred by methanolic leaf rinses ofLycopersicon pennellii, or of its F1 with tomato,L. esculentum. The active compounds in theL. pennellii rinsates were identified as 2,3,4-tri-O-acylglucoses bearing short to medium chain length fatty acids. These compounds are localized in the glandular exudate of the type IV trichomes and may accumulate to levels in excess of 400 g/cm2. In choice assays, purified glucose esters fromL. pennellii reduced aphid settling at concentrations as low as 25 g/cm2; at concentrations of 150 g/cm2 or more, all aphids avoided treated areas. Glucose esters were also active in deterring aphid settling in no-choice assays. At 100 g/ cm2, these esters resulted in increased levels of mortality after 48 hr.  相似文献

Walking tracks of Colorado potato beetles,Leptinotarsa decemlineata Say, were recorded on a locomotion-compensator in response to wind, odors of host plantsSolanum tuberosum L. and nonhost plantsLycopersicon hirsutum f.glabratum C.H. Mull, and to mixtures of these plant species. Host-plant odor induced positive anemotactic responses in starved females, whereas odor of the nonhostL. hirsutum was neither repellent nor attractive. The attractiveness of host-plant odor, however, was neutralized in the odor blend of plant species. Masking the attractive host-plant odor will hinder the beetle's searching for host-plant patches, and this principle may be exploited in pest control by mixed cropping.The locomotion-compensator was constructed with financial support from the Foundation for Fundamental Biological Research (BION), which is subsidized by the Netherlands Organization for the Advancement of Pure Research (ZWO), grant 14-02-02. -Bergamotenoic acid, a compound previously shown to stimulate oviposition inH. zea, was converted into a set of bicyclic analogs and tested with a set of acyclic side chain analogs to ascertain the molecular structure that maximizes insect behavioral response. While changes in the bicyclic ring elicited no variation in response, alteration in the side chain structure of-bergamotenoic acid resulted in significant changes in moth preference. Free rotation about the C-C bond proximal to the carboxylic acid group appears to be an important structural factor, since saturation of the side chain double bond significantly increased activity. The carboxylic acid group seems to be required for strong oviposition stimulation, since analogs lacking the carboxylic acid group exhibited no significant oviposition activity. Oviposition preference ofH. zea was also influenced by the length of the hydrocarbon chain to which the carboxylic acid is attached. While hexanoic acid was found inactive, the ovipositional preference for the heptanoic and octanoic acids was greatest for the one 8-carbon tested. This and other work suggest that carboxylic acids of specific chain lengths influence the oviposition behavior of bothHelicoverpa andHeliothis species and may be associated with host-plant selection. The potential use of this information in designing integrated pest management strategies for control ofH. zea is discussed.  相似文献

Tomatoes are perishable products due to the activity of microorganisms and endogenous enzymes. The objective was to produce cut tomatoes with extended shelf life, using the combined hurdle effect of osmotic pretreatment (OD) and high pressure (HP), instead of a conventional one‐step thermal process. Samples were processed in a multicomponent osmotic solution at 35 °C, subsequently cold‐pasteurised in pack at 600 MPa and stored at 5–15 °C. Quality deterioration during isothermal and nonisothermal storage was kinetically modelled. Both OD process and OD‐HP combined process caused an increase in lycopene content that was well retained. Texture, colour and flavour of treated samples were evaluated as similar to fresh, with OD‐HP samples showing better retention during storage. Being microbiologically stable, shelf life of OD‐HP samples was limited by sensory deterioration, whereas OD samples were rejected due to eventual microbial growth. Shelf life of OD and OD‐HP samples was estimated at 77 and 181 days, respectively, at 5 °C.  相似文献

目的 优化海蜇低分子肽(low molecular weight peptide from Rhopilema esculentum Kishinouye, RKLP)制备工艺,评价其抗氧化活性及分析氨基酸组成。方法 海蜇利用碱性和中性蛋白酶双酶分步酶解制备海蜇肽,以1,1-二苯基-2-三硝基苯肼(1,1-diphenyl-2-picrylhydrazyl,DPPH)自由基清除率为指标,响应面实验优化酶解工艺条件。采用超滤技术,获得分子量≤3.5 kDa RKLP。通过测定DPPH自由基、2,2’-联氮-双-3-乙基苯并噻唑啉-6-磺酸[2,2’-azino-bis(3-ethylbenzothiazoline-6-sulfonicacid),ABTS]自由基及超氧阴离子(O2-)自由基的清除率,评价RKLP抗氧化活性。氨基酸分析仪分析RKLP氨基酸组成。结果 酶解制备海蜇抗氧化肽的最佳工艺条件为:酶解温度50℃、3%碱性蛋白酶pH 7.5酶解2 h、1%中性蛋白酶pH 7.0酶解2 h,所得的DPPH自由基清除率为71.52%±0.59%,接近与预测值70.57%。RKLP水解度为6...  相似文献

使用荞麦(Fagopyrum esculentum)进行不同时间的发芽试验,然后评估其麦芽质量.由于根芽的增长,除根损失不断增加,α-淀粉酶活力增加,特别是在发芽的前三天,其后活力水平基本不变,可溶性β-淀粉酶的活力在发芽5天后达到最大值.荞麦麦芽的淀粉分解能力和浸出率与大麦麦芽相比是较低的,但可以通过调整糖化工艺,使其达到工业要求的浸出率.随发芽时间的延长,过滤速度也增加,发芽4天后,麦汁黏度开始下降.HPLC分析证实麦芽糖和葡萄糖是麦汁中的主要糖类.总可溶性氮、库尔巴哈值以及游离氨基氮(FAN)随发芽时间的增加而增加,主要是C组的氨基酸不断增加.荞麦制麦的最佳发芽时间是于15℃发芽4~5天.  相似文献

Effects of 3-pentanone and eight low molecular weight aliphatic alcohols and aldehydes identifed in the mixtures of volatiles released byAmaranthus palmeri S. Wats. (AMAPA) residues were determined on germination of onion, carrot, AMAPA, and tomato seeds. Three-day exposures to these volatiles significantly inhibited germination of these assay seeds, and the inhibition was dependent upon exposure time and concentration. Based on the degree of inhibition observed in both time- and concentration-dependent assays, the following activity series was obtained: 2-heptanol > 3-methyl-1-butanol, 1-hexanol > hexanal, 1-pentanol, 3-pentanone, acetaldehyde > ethanol, 2-methyl-1-propanol. The activities of this group of compounds with that of 2-heptanone appear to be additive and related to test compound volatility and hydrophilicity. 2-Heptanol and 2-heptanone also significantly inhibited the germination of other species, including shepherdspurse, soybean, lettuce, alfalfa, common purslane, oats, and lovegrass.Name of companies or commercial products are given solely for the purpose of providing specific information; their mention does not imply recommendation or endorsement by the U.S. Department of Agriculture over others not mentioned.  相似文献

Systemic induction following damage has been found in many plant species. Despite this widespread appreciation for the importance of induction, few studies have characterized the spatial variability of induction. We used tomato, Lycopersicon esculentum, to examine how damage to a single leaf affected the spatial distribution of systemic induction of proteinase inhibition in leaves above the damaged leaf. We crushed each leaflet of the second true leaf with forceps and measured the spatial distribution of proteinase inhibition in leaves 3, 4, and 5 at 8, 16, 24, 48, 72, and 120 hr. Constitutive levels of proteinase inhibitor activity were quantified in undamaged plants. We hypothesized that, due to vascular control of signal movement, systemic induction would show both among and within leaf variability. Following damage to leaf 2, induction was most pronounced in leaf 5 and minimal in leaf 3. In general, proteinase inhibitor activity was greatest at 24 hr and then declined. As predicted by vascular architecture, the near side of leaves in adjacent orthostichies showed higher induction than the far side of leaves. There was no increase in proteinase inhibitor activity in the undamaged neighboring plants. Overall our results demonstrate that systemic induction of proteinase inhibitors is partially controlled by vascular architecture and that future studies on systemic induction should examine the vascular architecture of the plants being studied. We argue that this spatial variation may influence the performance of herbivores sensitive to induced chemical changes.  相似文献

A system specifically designed for the non-destructive analysis of volatile organic compounds in fresh tomatoes, based on a dynamic headspace technique, has been set up to quantify the volatile aroma constituents of tomato fruits. This system will reduce the high variability associated to sample selection in post-harvest studies. Volatile compounds with a major contribution to aroma have been quantitatively determined in two traditional tomato cultivars and one commercial F1 hybrid. One of the traditional cultivars, “De la Pera,” got the highest values of odor and aroma in sensory tests performed using a trained panel of 10 assessors. The same cultivar showed significantly higher contents of all eight volatile compounds studied (hexanal, trans-2-hexenal, cis-3-hexenol, 1-hexanol, 1-nitro-3-methylbutane, 6-methyl-5-hepten-2-one, 2-isobutylthiazole, and β-ionone). Significant differences among traditional and hybrid tomato cultivars can be detected for volatile compounds, thus allowing the use of volatile determination as a possible tool in tomato breeding programs.  相似文献
