Human physiology normally contains pathogenic and non-pathogenic microorganisms in the gastrointestinal flora. Disturbance of these microorganism balance results in the formation of infection. Extensive use of antibiotics for cure of these disturbances like Helicobacter pylori (H. pylori) infection leads to patient discomfort and associated side-effects. There is a need to adopt adjunct or alternative approach in order to minimize such conditions. Probiotics is one of the potential therapies to cure gastrointestinal discomforts especially associated with H. pylori. It competes through non-immune and immune systems. This review article concludes that probiotics are used to eradicate the infection at increased rate, and decreased associated side-effects are caused by triple therapy. A proper evaluation of these probiotics is demanded before their use in future as a commercial product. Furthermore, their effect on immune system requires more research work so that their usage for other chronic disorders can also be considered. 相似文献

The potential effect of FOS and inulin upon probiotic bacterium performance in curdled milk matrices
Inulin and fructooligosaccharides were studied for their prebiotic effect upon growth/survival of probiotic bacteria and technological potential in probiotic food processing, via characterization of glycolysis, proteolysis and lipolysis in curdled milk matrices; the ultimate goal is the manufacture of synbiotic cheeses. Prebiotic compounds did not significantly affect growth/viability of all strains studied, except Lactobacillus acidophilus La-5. Proteolysis indices revealed considerable casein degradation in probiotic and synbiotic matrices inoculated with Bifidobacterium lactis B94 and Lactobacillus casei-01; lower values were achieved in those inoculated with L. acidophilus La-5, yet a synbiotic effect was apparent in NPN values. Lipolysis was not extensive over storage, irrespective of matrix type; however, interesting differences in terms of the qualitative free fatty acids profile were observed. CLA isomers, and α-linolenic and γ−linolenic acids were detected upon 15 d of ripening of all inoculated matrices. Principal component analysis was able to discriminate the various matrices according to degree of maturation, throughout the ripening period. Microbiological and biochemical parameters unfolded a very good technological potential, especially of B. lactis B94 and L. casei-01, to produce novel types of functional dairy matrices - although extrapolation to actual cheeses should still be done with care, because e.g. syneresis was not considered. 相似文献

Over the years, the attempts to elucidate the role of beneficial microorganisms in shaping human health are becoming fairly apparent. The functional impact conferred by such microbes is not only transmitted by viable cells or their metabolites but also through non-viable cells. Extensive research to unveil the protective action of such wonder bugs has resulted in categorizing the beneficial microflora and their bioactive metabolites into a variety of functional biotic concepts based on their intended applications in various forms. In the modern era, these are often termed as probiotics, prebiotics, synbiotics, postbiotics, next-generation probiotics, psychobiotics, oncobiotics, pharmabiotics, and metabiotics. Currently, the concept of traditional probiotics is being widened to include microbes beyond lactic acid bacteria. Indeed, this diversification has broadened the functional food portfolio from food to pharmaceuticals. Xylanase from Aspergillus foetidus MTCC 4898 was purified using ammonium sulphate fractionation followed by ion exchange and gel permeation chromatography with 12.26-fold purity and 29.9% recovery. Purified xylanase was found to be 29.9 kDa. Optimum temperature and pH for xylanase activity of purified xylanase were found to be 50 °C and 5.3 respectively. Presence of additives like polyethylene glycol, sodium azide, Tween 80, KCl and NaCl increased the stability of purified xylanase by 35, 29, 28, 32 and 43% respectively at 50 °C after 180 min. Kinetic parameters like Km and Vmax were found to be 4 mg/ml and 7288 μmol/mg/min respectively. The purified xylanase was found to be an endoxylanase as it produced only xylooligosaccharides (XOS) from birchwood xylan. Production of XOS was carried out from xylan extracted from agro-residues using β-xylosidase free xylanase. Maximum yield of XOS was 7.28 ± 0.14 mg/ml and 4.52 ± 0.21 mg/ml from wheat straw xylan and rice straw xylan respectively. XOS mixture was suitable for food industry looking at its high thermal stability at low pH. Prebiotic effect of XOS was evaluated by in vitro fermentation of XOS using known probiotic strains of Bifidobacterium spp. 相似文献

The aim of this human study was to assess the influence of prebiotic-induced gut microbiota modulation on PUFA-derived bacterial metabolites production. Therefore, we analyzed the circulating fatty acid profile including CLA/CLnA in obese women treated during 3 months with inulin-type fructan prebiotics. In these patients, we had already determined gut microbiota composition by phylogenetic microarray and qPCR analysis of 16S rDNA. Some PUFA-derived bacterial metabolites were detected in the serum of obese patients. Despite the prebiotic-induced modulation of gut microbiota, including changes in CLA/CLnA-producing bacteria, the treatment did not impact significantly on the circulating level of these metabolites. However, some PUFA-derived bacterial metabolites were positively correlated with specific fecal bacteria (Bifidobacterium spp., Eubacterium ventriosum and Lactobacillus spp.) and inversely correlated with serum cholesterol (total, LDL, HDL). These correlations suggest a potential beneficial effect of some of these metabolites but this remains to be confirmed by further investigation. 相似文献

Since many years, the role of the colonic microbiota in maintaining the host's overall health and well-being has been recognized. Dietary modulation of the microbiota composition and activity has been achieved by the use of pre-, pro- and synbiotics. In this review, we will summarize the available evidence on the modification of bacterial metabolism by dietary intervention with pre-, pro- and synbiotics. Enhanced production of SCFA as a marker of increased saccharolytic fermentation is well documented in animal and in vitro studies. Decreased production of potentially toxic protein fermentation metabolites, such as sulfides, phenolic and indolic compounds, has been less frequently demonstrated. Besides, pre-, pro- and synbiotics also affect other metabolic pathways such as the deconjugation of secondary bile acids, bacterial enzyme activities and mineral absorption. Data from human studies are less conclusive. The emergence of new analytical techniques such as metabolite profiling has revealed new pathways affected by dietary intervention. However, an important challenge for current and future research is to relate changes in bacterial metabolism to concrete health benefits. Potential targets and expected benefits have been identified: reduced risk for the metabolic syndrome and prevention of colorectal cancer. 相似文献

为了解益生菌与益生元对人体肠道菌群的调节作用,选择符合试验要求的成年志愿者,分别食用益生菌和益生元,其中益生菌组150人,益生元组200人,男女各半。结果表明:益生菌与益生元均可增加人体肠道内的双歧杆菌和乳杆菌:均可使肠道内肠杆菌数量下降。但益生菌组双歧杆菌和乳杆菌增加的数量及肠杆菌和拟杆菌降低的数量均高于益生元组,说明在该试验条件下,益生菌组的效果要好于益生元组。 相似文献

The goal of this research was to determine the effect of different doses of galactooligosaccharide (GOS) on the fecal microbiota of healthy adults, with a focus on bifidobacteria. The study was designed as a single-blinded study, with eighteen subjects consuming GOS-containing chocolate chews at four increasing dosage levels; 0, 2.5, 5.0, and 10.0 g. Subjects consumed each dose for 3 weeks, with a two-week baseline period preceding the study and a two-week washout period at the end. Fecal samples were collected weekly and analyzed by cultural and molecular methods. Cultural methods were used for bifidobacteria, Bacteroides, enterobacteria, enterococci, lactobacilli, and total anaerobes; culture-independent methods included denaturing gradient gel electrophoresis (DGGE) and quantitative real-time PCR (qRT-PCR) using Bifidobacterium-specific primers. All three methods revealed an increase in bifidobacteria populations, as the GOS dosage increased to 5 or 10 g. Enumeration of bifidobacteria by qRT-PCR showed a high inter-subject variation in bifidogenic effect and indicated a subset of 9 GOS responders among the eighteen subjects. There were no differences, however, in the initial levels of bifidobacteria between the responding individuals and the non-responding individuals. Collectively, this study showed that a high purity GOS, administered in a confection product at doses of 5 g or higher, was bifidogenic, while a dose of 2.5 g showed no significant effect. However, the results also showed that even when GOS was administered for many weeks and at high doses, there were still some individuals for which a bifidogenic response did not occur. 相似文献
