A simple and accurate liquid chromatographic method was developed to estimate cyproterone acetate (CA) in pharmaceuticals. The drug was chromatographed on a reversed-phase C18 column. Eluents were monitored at a wavelength of 254 nm utilizing a mixture (60:40) of acetonitrile and water. Solution concentrations were measured on a weight basis to avoid the use of an internal standard. The method was statistically validated for linearity, accuracy, precision, and selectivity. Due to its simplicity and accuracy, we believe that the method can be used for routine quality control analysis. No specific sample preparation is required except for the use of a column guard and a suitable prefilter attached to the syringe. 相似文献

This study discusses the effect of formulation composition on the physical characteristics and drug release behavior of controlled-release formulations made by roller compaction. The authors used mixture experimental design to study the effect of formulation components using diclofenac sodium as the model drug substance and varying relative amounts of microcrystalline cellulose (Avicel), hydroxypropyl methylcellulose (HPMC), and glyceryl behenate (Compritol). Dissolution studies revealed very little variability in drug release. The t70 values for the 13 formulations were found to vary between 260 and 550 min. A reduced cubic model was found to best fit the t70 data and gave an adjusted r-square of 0.9406. Each of the linear terms, the interaction terms between Compritol and Avicel and between all three of the tested factors were found to be significant. The longest release times were observed for formulations having higher concentrations of HPMC or Compritol. Tablets with higher concentrations of Avicel showed reduced ability to retard the release of the drug from the tablet matrix. Crushing strength showed systematic dependence on the formulation factors and could be modeled using a reduced quadratic model. The crushing strength values were highest at high concentrations of Avicel, while tablets with a high level of Compritol showed the lowest values. A predicted optimum formulation was derived by a numerical, multiresponse optimization technique. The validity of the model for predicting physical attributes of the product was also verified by experiment. The observed responses from the calculated optimum formulation were in very close agreement with values predicted by the model. The utility of a mixture experimental design for selecting formulation components of a roller compacted product was demonstrated. These simple statistical tools can allow a formulator to rationally select levels of various components in a formulation, improve the quality of products, and develop more robust processes. 相似文献

Spray-drying was used for the preparation of paracetamol/eudragit RS or RL or ethylcellulose microspheres to verify the possibility of their use in controlled-release solid-dosage forms formulation and try to determine advantages and limits of the technique of such use. Microspheres were first characterized by scanning electron microscopy, differential scanning calorimetry, x-ray diffractometry, and in vitro dissolution studies and then used for the preparation of tablets. During this step, the compressibility of the spray-dried powders was also evaluated. In vitro dissolution studies were performed also on the tablets and their release control was accessed. Although powders were unable to slow down drug release, tablets obtained from microsphere compression showed a good capability of controlling paracetamol release when eudragit RS or ethylcellulose was used, even at low polymer amounts. 相似文献

This paper investigates inter-tablet coating variability, specifically, the variability of tablet residence times within the spray zone of a horizontal coating pan. Results from experiments, discrete element method (DEM) computer simulations, and an analytical model developed to describe the coating mass distribution are presented.The simulations indicate that the coefficient of variation of tablet residence times, and subsequently, of coating mass, decreases with time following a power law relation. The theoretical model demonstrates that the coefficient of variation of residence time for a randomly mixed tablet bed is inversely proportional to the square root of the number of coating “trials”. DEM simulations show that during each pan revolution, tablets in the spray zone remain in a quasi-segregated state from tablets located outside the spray zone for some time period termed Δtseg. Increasing the pan's Froude number, the spray zone aspect ratio, and the tablet-tablet and tablet-pan friction coefficient all act to decrease Δtseg, leading to more uniform residence times and less inter-tablet coating variability for a given operating time. The relationship between Δtseg and tablet load is more complex due changes in bed dynamics. In addition to the variability studies, a model is developed that relates coating fraction, effective mass flow rate, Δtseg, and the desired coating mass to the allowable fraction of tablets with a coating mass lying outside of a specified range of coating masses. 相似文献

Teachers play a key role in adoption of technologies for classroom use. This study surveyed teachers regarding their intention to use tablet technology to interact with students and other teachers through smart school websites technology. The Unified Theory of Acceptance and Use of Technology (UTAUT) served as the theoretical framework for this study, which was then augmented with context-specific determinants of adoption that captured teachers’ perceptions of the effects of technology on interactions with students and peers. The results showed no resistance among teachers with regard to trying different ways of working with tablets to achieve better learning, unlike other studies using the UTAUT constructs that have found negative reactions to implementing new technologies in the workplace. This is a crucial finding, since teacher appraisals affect the ways in which this technology can be employed to foster learning through technology facilitated interactions. 相似文献

Antacid tablets based on milk proteins were developed by a wet granulation method, with the aim of controlling the problem of hyperacidity in its early stages. Optimisation of the tablet formulation was carried out using milk protein ingredients, viz. rennet casein (RC) and whey protein concentrate (WPC), with mannitol used as a functional filler. An RC‐WPC ratio and a mannitol level of 1.96% and 10.75% w/w, respectively, were found to be optimum. The RC‐WPC ratio had a significant impact on buffering capacity (BC), tablet friability and density. The filler material, mannitol, also has an impact on BC and tablet density. 相似文献
