目的 探讨长期经饮水方式补充牛磺酸对高脂膳食所致肥胖大鼠降脂减重作用,对甘油三酯合成关键分子和胆固醇7α羟化酶(cholesterol 7α-hydroxylase, CYP7A1)表达调控关键分子的影响。方法 根据血清胆固醇水平和体重, SD大鼠被分为3组,为对照组(normal, N)饲喂基础维持饲料,肥胖组(high fat, HF)和牛磺酸组(high fat taurine, HFT)饲喂高脂饲料。N组和HF组自由饮水,而HFT组给予牛磺酸溶液代替水,持续12周。结果 与HF组相比,给予牛磺酸的HFT组大鼠体重、附睾周围脂肪重量、血清总胆固醇和低密度脂蛋白胆固醇、游离脂肪酸、固醇调节元件结合蛋白-1c (sterol regulatory element-binding protein-1c, SREBP-1c)和脂肪酸合成酶(fatty acid synthase, FAS)基因表达显著降低,粪便胆汁酸水平、脂蛋白脂肪酶(lipoprotein lipase, LPL)活性以及CYP7A1和Ser63p-c-Jun的蛋白表达显著增加,但肝细胞核因子4α(hepatocyte...  相似文献

The present study aims to investigate the beneficial role of taurine in tert-butyl hydroperoxide (TBHP)-induced oxidative stress, cytotoxicity and cell death using murine hepatocytes as the working model. TBHP caused a time-dependent increase in alkaline phosphatase leakage, extensive ROS formation and alteration in antioxidant machineries. Moreover, TBHP upregulated NF-κB phosphorylation, caused injury to cellular mitochondria by reciprocal regulation of Bcl2 family proteins; incapacitated cellular respiration, accelerated cytochrome c release from mitochondria and induced apoptotic death in hepatocytes. Taurine treatment prevented TBHP-induced oxidative insult and decreased the activation of NF-κB as well as apoptotic signalling pathways. Signal transduction studies with specific inhibitors PS1145 (IκB inhibitor) and LY294002 (PI3K inhibitor) showed that the mechanism of the protective action of taurine involves the upregulation of Akt and inhibition of NF-κB. Combining, our result suggest that taurine supplementation in regular diet may provide a potential therapeutic choice against TBHP-induced hepatic oxidative injury.  相似文献

The effects of 50% replacement of NaCl by KCl and addition of the amino acids lysine and taurine and the 5'-ribonucleotide disodium inosinate and disodium guanylate on some sensory and physicochemical parameters of fermented cooked sausages were evaluated. The partial replacement of NaCl by KCl did not alter the manufacturing process; however, defects in the sensory quality were detected. Lysine at a concentration of 0.313% and a mixture of taurine (750 mg/kg) with disodium inosinate (300 mg/kg) and disodium guanylate (300 mg/kg) reduced the sensory defects caused by KCl, allowing the fermented cooked sausages to be elaborated with reduced sodium content and high sensory quality.  相似文献

Taurine is a sulfur‐containing amino acid that is present in mammalian tissues in millimolar concentrations. Taurine is involved in a diverse array of biological and physiological functions, including bile salt conjugation, osmoregulation, membrane stabilization, calcium modulation, anti‐oxidation, and immunomodulation. The prevalence of obesity and being overweight continues to rise worldwide at an alarming rate. Obesity is associated with a higher risk of metabolic and cardiovascular diseases, cancer, and other clinical conditions. Ingestion of taurine has been shown to alleviate metabolic diseases such as hyperlipidemia, diabetes, hypertension, and obesity in animal models. A global epidemiological survey showed that 24‐h urinary taurine excretion, as a marker of dietary taurine intake, was inversely associated with BMI, blood pressure, and plasma cholesterol in humans. In addition, taurine chloramine, an endogenous product derived from activated neutrophils, has been reported to suppress obesity‐induced oxidative stress and inflammation in adipocytes. Synthetic activity and concentration of taurine in adipose tissues and plasma have been shown to decrease in humans and animals during the development of obesity, suggesting a relationship between taurine deficiency and obesity. In this review, I summarize the effects of taurine on the progression of obesity in animal models and humans. Furthermore, I discuss possible mechanisms underlying the antiobesity effects of taurine.  相似文献

运用天冬酰胺/葡萄糖模式反应体系研究了牛磺酸对富含淀粉的食品中丙烯酰胺生成的影响及其抑制机理。试验结果显示:牛磺酸能够显著抑制天冬酰胺/葡萄糖反应体系中丙烯酰胺的生成,且抑制率与浓度呈剂量关系。运用LC-QTOF分析得到2种主要的反应产物:丙烯酰胺-牛磺酸加合物和丙烯酰胺二聚体-牛磺酸加合物。此外,牛磺酸与葡萄糖共存于同一体系中加热时,葡萄糖的含量显著下降,说明牛磺酸可与葡萄糖发生反应。通过模式反应体系证明:牛磺酸能够显著降低富含淀粉食品中的丙烯酰胺含量;抑制机理主要是牛磺酸与丙烯酰胺直接发生反应,反应产物为丙烯酰胺-牛磺酸加合物和丙烯酰胺二聚体-牛磺酸加合物,从而除去体系中已经生成的丙烯酰胺。此外,牛磺酸能够与葡萄糖发生美拉德反应,从而与天冬酰胺竞争消耗体系中的葡萄糖,减少了食品中的丙烯酰胺含量。  相似文献
