用毛细管电泳-电化学检测的方法研究了荞麦中黄酮类物质:表儿茶素、芦丁、槲皮素的含量,研究了电极电位、缓冲液的pH值、分离电压及进样时间对电泳的影响,得到优化的测定条件。以直径为300μm的碳圆盘电极为检测电极,工作电极电位为0.95V(vs.SCE),在50mmol/L硼砂(pH8.5)运行缓冲液中,上述3组分在12min内完全分离。表儿茶素、芦丁、槲皮素线性范围分别为5×10-7 ̄1×10-4g/mL、1×10-6 ̄1×10-4g/mL、1×10-6 ̄1×10-4g/mL;检出限分别为1.83×10-7g/mL、2.9×10-8g/mL、1.00×10-7g/mL;3种标样7次平行进样的相对标准偏差(RSD)小于2.5%;回收率表儿茶素100.4%、芦丁98.1%、槲皮素100.1%(n=3)。该法灵敏可靠,结果令人满意。  相似文献

ABSTRACT: Grape juice processing by‐products, grape seed and pomace are a rich source of procyanidins, compounds that may afford protection against chronic disease. This study was undertaken to identify optimal extrusion conditions to enhance the contents of monomers and dimers at the expense of large molecular weight procyanidin oligomers and polymers in grape seed and pomace. Extrusion variables, temperature (160, 170, and 180 °C in grape seed, and 160, 170, 180, and 190 °C in pomace) and screw speed (100, 150, and 200 rpm in both) were tested using mixtures of grape seed as well as pomace with decorticated white sorghum flour at a ratio of 30 : 70 and moisture content of 45%. Samples of grape seed and pomace were analyzed for procyanidin composition before and after extrusion, and total anthocyanins were determined in pomace. Additionally, chromatograms from diol and normal phase high‐performance liquid chromatography were compared for the separation of procyanidins. Extrusion of both grape by‐products increased the biologically important monomer and dimers considerably across all temperature and screw speeds. Highest monomer content resulted when extruded at a temperature of 170 °C and screw speed of 200 rpm, which were 120% and 80% higher than the unextruded grape seed and pomace, respectively. Increases in monomer and dimer contents were apparently the result of reduced polymer contents, which declined by 27% to 54%, or enhanced extraction facilitated by disruption of the food matrix during extrusion. Extrusion processing reduced total anthocyanins in pomace by 18% to 53%. Practical Applications: Extrusion processing can be used to increase procyanidin monomer and dimer contents in grape seed and pomace. Procyanidins in grape by‐products have many health benefits, but most are present as large molecular weight compounds, which are poorly absorbed. Extrusion processing appears to be a promising technology to increase levels of the bioactive low molecular weight procyanidins.  相似文献

The (−) epicatechin (Epi) is a flavonoid with antioxidant and regenerative properties for cardiac tissue. In this work, a study of release assays and their corresponding adjustments to models was performed to determine the mechanism of (−) Epi release in the cellulose acetate (CA)–polyvinylpyrrolidone (PVP) system. The morphology of the material was considered, fibers, or spheres, as well as the possible chemical interactions between Epi, PVP, and CA. It was observed that in the systems in which the PVP is present, a high release is observed in comparison with those that only contained CA and (−) Epi, which showed very low release. This difference can be explained considering the possible hydrogen bond interaction between CA and (−) Epi, as well as the insolubility of the CA, in the release medium. The factors that have an important effect on the release of (−) Epi in the different systems are: the morphology, a faster release in morphologies with greater surface area, and the chemical interactions that can be formed among the (−) Epi and the components of the matrix. Another factor is the solubility of the components in the release medium, which allows the diffusion and drag phenomena to be observed simultaneously. © 2018 Wiley Periodicals, Inc. J. Appl. Polym. 研究从茶树的鲜叶、茎部和茶树土壤中分离出活性真菌,通过利用天然微生物的转化能力,使简单儿茶素转化为功能性较强的复杂型酯型儿茶素。实验结果表明:筛选出了具有转化能力的活性菌种,能有效地将简单儿茶素转化为酯型儿茶素,而且其转化能力良好。并对此种活性真菌进行形态学和生理生化的初步检测,判断其属于丝状真菌藻状菌纲,毛霉目,内囊霉科,内囊霉属。  相似文献

This study compared the antioxidative activity of green tea catechin (GTC) extract with that of rosemary extract in canola oil, pork lard, and chicken fat. The GTC extract was obtained from jasmine and longjing green teas and mainly consisted of four epicatechin isomers including (−)epigallocatechin gallate (EGCG), (−)epigallocatechin (EGC), (−)epicatechin (EC), and (−)epicatechin gallate (ECG). The oxidation was conducted at 100 ± 2°C by monitoring oxygen uptake. The oxygen consumption test demonstrated that GTC extract was much more effective than the rosemary extract against lipid oxidation in canola oil, pork lard, and chicken fat under the conditions of the present study. Together with our previous study which showed that GTC extract was more protective than butylated hydroxytoluene as an antioxidant, these results suggest that GTC as a mixture of EGCG, EGC, EC, and ECG may serve as an antioxidant in processed foods.  相似文献

为建立加压毛细管电色谱(pCEC)分离检测甘蔗中多酚类化合物绿原酸、阿魏酸、表儿茶素的方法,采用C18毛细管色谱柱,以NaH2PO4和甲醇为流动相,优化了流动相比例、缓冲盐pH、缓冲盐浓度、分离电压、流速等色谱条件。结果表明,在流动相为15.0 mmol/L NaH2PO4+12.5 mmol/L庚烷磺酸钠(pH5.0)/甲醇(50:50,V/V)、分离电压为1 kV、流动相总流速为0.06 mL/min、检测波长为220 nm的色谱条件下,绿原酸、阿魏酸、表儿茶素分离较好,在20~320 μg/mL线性关系良好,相关系数分别为0.9978、0.9922、0.9971,检出限分别为0.14、0.75、1.65 μg/mL,相对标准偏差分别为2.82%、2.69%、1.18%,平均回收率分别为96.79%、100.71%、99.01%。该方法快速、准确、重复性好,适用于甘蔗中多酚类化合物的分离检测。  相似文献

以志愿者体内实验方式考察绿茶胶囊对黑巧克力中表儿茶素和儿茶素在人体内生物利用率的影响.六位志愿者在实验前一天严格执行低黄酮类饮食,空腹过夜并留下尿样作为空白样本.试验第一阶段:志愿者摄入50g黑巧克力(含表儿茶素152mg,儿茶素33mg).一周之后的第二阶段,志愿者摄入50g黑巧克力和一片绿茶胶囊(绿茶胶囊含表儿茶素207mg,儿茶素39mg).服用后每间隔一定时间,收集志愿者尿样,测定表儿茶素和儿茶素的生物利用率.结果显示,虽然第二阶段的表儿茶素和儿茶素摄入总量均高于第一阶段,但两者在第一阶段人体中的生物利用率均高于第二阶段.因此和单独摄入黑巧克力相比,绿茶胶囊和黑巧克力同时服用可能会导致减少或者延迟人体对表儿茶素和儿茶素的吸收.  相似文献

应用高效逆流色谱法(HPCCC)对茶叶中的儿茶素和表儿茶素两种同分异构体的分离制备方法进行研究。结果表明:以正己烷-乙酸乙酯-水(体积比1:20:20)为两相溶剂系统,上相为固定相,下相为流动相,可以实现儿茶素和表儿茶素的完全分离。采用138mL的分离柱、1g儿茶素粗提物,在120min内一次制备即可得到纯度在95%以上的表儿茶素约35mg,纯度在99%以上的儿茶素325mg,纯度90%~99%的儿茶素100mg。  相似文献

Constituents of the fruit of Amomum tsao-ko were investigated following a preliminary screening of the antioxidant activity of several extracts of the fruit of this plant that showed that the dichloromethane extract and the ethyl acetatesoluble and water-soluble fractions of the 70% aqueous acetone extract had higher activity than α-tocopherol and butylated hydroxytoluene (BHT). Eleven compounds were isolated from the ethyl acetate-soluble fraction, and their structures were elucidated as (+)-hannokinol (1), meso-hannokinol (2), (+)-epicatechin (3), (−)-catechin (4), β-sitosterol (5), β-sitosterol 3-O-glucoside (6), 2,6-dimethoxyphenol (7), protocatechualdehyde (8), protocatechuic acid (9), vanillic acid (10), and p-hydroxybenzoic acid (11) based on mass and various nuclear magnetic resonance (NMR) spectroscopic techniques. This is the first isolation of epicatechin and catechin from the genus Amomum. The radical scavenging activity of the isolated compounds was evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) and colorimetric and electron spin resonance (ESR) analyses. The antioxidant activity of the compounds was also determined based on the oxidative stability index (OSI). The catechins and catechol derivatives showed strong activities in both the DPPH radical scavenging activity and antioxidant activity assays.  相似文献
