以转基因水稻中最常用的CaMV35S启动子、NOS终止子、Cry1Ab/Ac基因、HPT基因及SPS水稻内标基因为研究对象,利用5 种不同的荧光信号(FAM、HEX、Taxas Red、Cy5、Cy5.5)进行多重实时聚合酶链式反应(real-time polymerase chain reaction,real-time PCR)检测方法的研究。通过引物组合筛选、反应体系优化、特异性测试、灵敏度测试、适用性测试等一系列实验,建立了5 重real-time PCR方法,灵敏度可达0.032%。此方法具有灵敏度高、结果准确、通量大等优点,可实现水稻中转基因成分的快速、高效检测。 相似文献

In this paper, low-cost and two-cycle hardware structures of the PRINCE lightweight block cipher are presented. In the first structure, we proposed an area-constrained structure, and in the second structure, a high-speed implementation of the PRINCE cipher is presented. The substitution box (S-box) and the inverse of S-box (S-box−1) blocks are the most complex blocks in the PRINCE cipher. These blocks are designed by an efficient structure with low critical path delay. In the low-cost structure, the S-boxes and S-boxes−1 are shared between the round computations and the intermediate step of PRINCE cipher. Therefore, the proposed architecture is implemented based on the lowest number of computation resources. The two-cycle implementation of PRINCE cipher is designed by a processing element (PE), which is a general and reconfigurable element. This structure has a regular form with the minimum number of the control signal. Implementation results of the proposed structures in 180-nm CMOS technology and Virtex-4 and Virtex-6 FPGA families are achieved. 本文采用高通测序方法对大凌河流域五刺金鱼藻、格菱、狐尾藻、水蓼及芦苇5种水生植物不同季节的微生物群落进行试验分析,分析表明:藻类密度在沉水植物表层要高于浮叶植物表层,微生物群落在浮叶植物根部的结构与其表面存在明显差异。各水生植物的优势生物群落依次为变形菌、梭菌、柔膜菌、鞭脂杆菌、杆菌、疣微菌门,且各微生物群落在季节丰度上存在明显差异性。由此可见,水环境对生物群落的影响要高于植物类型。研究成果可为大凌河流域生态修复工程提供重要理论依据。 相似文献

A novel autophagy inhibitor, autophazole (Atz), which promoted cancer cell death via caspase activation, is described. This compound was identified from cell-based high-content screening of an imidazole library. The results showed that Atz was internalized into lysosomes of cells where it induced lysosomal membrane permeabilization (LMP). This process generated nonfunctional autolysosomes, thereby inhibiting autophagy. In addition, Atz was found to promote LMP-mediated apoptosis. Specifically, LMP induced by Atz caused release of cathepsins from lysosomes into the cytosol. Cathepsins in the cytosol cleaved Bid to generate tBid, which subsequently activated Bax to induce mitochondrial outer membrane permeabilization (MOMP). This event led to cancer cell death via caspase activation. Overall, the findings suggest that Atz will serve as a new chemical probe in efforts aimed at gaining a better understanding of the autophagic process. 相似文献

High-throughput synthesis and screening methods have been developed for the discovery of highly active lead compounds for the selective catalytic reduction as well as direct decomposition of NO in the temperature range 200–300 °C. The discovery libraries for primary screening consisted of 16 × 16 catalyst arrays on 4in. square quartz wafers. Catalysts were prepared by robotic liquid dispensing techniques and screened for catalytic activity in Symyx' scanning mass spectrometer. The scanning mass spectrometer is a fast serial screening tool that uses flat wafer catalyst surfaces, local laser heating, a scanning/sniffing nozzle and a quadrupolar mass spectrometer to compare relative catalytic activities. The feed consisted of NO/NH3 mixtures with optional O2 cofeed and Kr as the internal standard in Ar carrier gas. QMS detection allowed for tracking of H2O, N2, NO, O2, N2O and Kr. Screening protocols for catalytic materials encompassed metal precursors and carriers for supported vanadia systems, extensive doping of V2O5/TiO2, and broad screening of mixed redox metal oxides and supported base and noble metal systems. More than 500 samples could be screened in a single day. Active hits (high NO consumption accompanied by corresponding N2 production) identified in discovery libraries were re-synthesized as focus libraries for lead confirmation and further optimization. These libraries used shallower compositional gradients, for example 56 points (compositions) per ternary, with four 56-point ternaries per 4in. wafer. Broad screening ternaries were generally 8 or 15 points. The focus libraries more clearly reveal the trends and provide guidelines for secondary screening and scale-up. High conversions were achieved in scanning mass spectrometer so the scalability risk is small for the short contact time reactions. 相似文献

序批式生物膜反应器(sequencing biofilm batch reactor, SBBR)是应用广泛的污水处理方法。为探究不同时期排泥对SBBR污染物去除效果与微生物群落结构的影响,本研究设置了挂膜初期、中期和后期进行排泥的反应器处理生活污水,同时结合16S rDNA高通量测序技术对微生物群落结构进行分析,并采用机器学习(machine learning, ML)的方法,在传统的微生物优势种分析基础上,对测序数据进行深度挖掘,寻找造成组间差异的关键物种。水质测定结果显示,COD去除效果在不同时期排泥的SBBR间没有明显差异,出水COD均低于30mg/L。挂膜中期排泥的SBBR的NH3-N去除率先达到稳定且高于前期和后期排泥的系统。高通量测序结果显示,各SBBR中微生物优势种均以降解有机物的物种为主。挂膜中期排泥的SBBR中,ML筛选得到的NH3-N去除相关物种(Hydrogenophaga、Gemmata和Nitrospira)与差异关键物种丰度更高,微生物群落结构稳定性更强,可从微生物层面解释分析SBBR污染物去除效果的差异。 相似文献

All-d Heuslers are a category of novel compounds combining versatile functionalities such as caloric responses and spintronics with enhanced mechanical properties. Despite the promising transport properties (anomalous Hall (AHC) and anomalous Nernst (ANC) conductivities) shown in the conventional Co2XY Heuslers with p-d hybridization, the all-d Heuslers with only d-d hybridization open a new horizon to search for new candidates with outstanding transport properties. In this work, the AHC and ANC are evaluated for thermodynamically stable ferro/ferri-magnetic all-d-metal regular Heusler compounds based on high-throughput first-principles calculations. It is observed that quite a few materials exhibit giant AHCs and ANCs, such as cubic Re2TaMn with an AHC of 2011 S cm-1, and tetragonal Pt2CrRh with an AHC of 1966 S cm-1 and an ANC of 7.50 A m-1K-1. Comprehensive analysis on the electronic structure reveals that the high AHC can be attributed to the occurrence of the Weyl nodes or gapped nodal lines in the neighborhood of the Fermi level. The correlations between such transport properties and the number of valence electrons are also thoroughly investigated, which provides a practical guidance to tailor AHC and ANC via chemical doping for transverse thermoelectric applications. 相似文献
