Sea lice threaten the welfare of farmed Atlantic salmon and the sustainability of fish farming across the world. Chemical treatments are the major method of control but drug resistance means that alternatives are urgently needed. Selective breeding can be a cheap and effective alternative. Here, we combine experimental trials and diagnostics to provide a practical protocol for quantifying resistance to sea lice. We then combined quantitative genetics with epidemiological modelling to make the first prediction of the response to selection, quantified in terms of reduced need for chemical treatments. We infected over 1400 young fish with Lepeophtheirus salmonis, the most important species in the Northern Hemisphere. Mechanisms of resistance were expressed early in infection. Consequently, the number of lice per fish and the ranking of families were very similar at 7 and 17 days post infection, providing a stable window for assessing susceptibility to infection. The heritability of lice numbers within this time window was moderately high at 0.3, confirming that selective breeding is viable. We combined an epidemiological model of sea lice infection and control on a salmon farm with genetic variation in susceptibility among individuals. We simulated 10 generations of selective breeding and examined the frequency of treatments needed to control infection. Our model predicted that substantially fewer chemical treatments are needed to control lice outbreaks in selected populations and chemical treatment could be unnecessary after 10 generations of selection. Selective breeding for sea lice resistance should reduce the impact of sea lice on fish health and thus substantially improve the sustainability of Atlantic salmon production. 相似文献

Vulnerability, scar, and reciprocal-relations models of depressive symptoms and self-esteem were compared among people with severe mental illness (SMI; N=260) participating in a partnership-based intervention study. Assessments were conducted at baseline, midway through the intervention (after 4 months), and at termination (after 9 months). Cross-lagged, structural equation modeling analyses revealed that participants' baseline depressive symptoms predicted a decrease in self-esteem in the first 4 months but not in the subsequent 5 months of participation. Exploratory regression analyses indicated that improved social functioning buffered this deleterious effect of depressive symptoms. These findings, which are consistent with the scar model, highlight the fragile nature of the self and the importance of social functioning in recovery from SMI. The role of olfaction and diffusible pheromones in mate location behavior of sea lice, Lepeophtheirus salmonis, was assessed with Y-tube behavioral bioassays. The pheromone emitting animals were located in a chamber in one arm of a Y-tube arena, with artificial seawater flowing through both arms. Adult male sea lice displayed both activation and directional responses to seawater conditioned with preadult II virgin females, but were only activated by mated adult female conditioned water. Further, when males were given the choice of preadult II virgin females or mated adult females, a significant number of males chose the arm with the preadult II virgin females. Adult males showed activation responses when presented with water conditioned with adult males but were not attracted to them. When presented with adult males, preadult II virgin females showed only directional responses, but not activation responses. Preadult II virgin female conditioned water was extracted using solid-phase extraction (SPE) protocols pioneered for semiochemical isolation. Adult male sea lice showed significant directional responses to the preadult II virgin female SPE extract. Distillation under vacuum was performed on the extract to give a distillate comprising components with a molecular weight range and physical properties comparable to those of compounds utilized as volatile semiochemicals by terrestrial organisms and a residue comprising components with higher molecular weight range comparable to those utilized as involatile semiochemicals. Adult males were found to be both significantly activated and attracted to the distillate, but not to the residue. It has already been known for many years that the use of some extreme-pressure (EP), antiwear or friction modifier (FM) additives in mineral oils can produce different kind of boundary or chemical reaction films on sliding contact surfaces of some kinds of steel in boundary lubrication conditions. Using a sliding ball-on-disc configuration lubricated with some kinds of EP or FM, the wear scars on the balls can always reach the same limit size at a specified applied load and sliding velocity. From the fact that the limit sizes of wear scars decrease as sliding speed is increased or applied load is decreased, the load carrying ability of a chemical film can be obtained by extrapolating the data to the condition of zero sliding speed and is so defined that if the contact pressure is greater than this load carrying ability, the contact surfaces will continuously be worn; if the contact pressure is smaller than it, no more wear will occur on the surfaces. Based on this load carrying ability, the hydrodynamic effect of sliding pairs can also be identified. Structural ceramics are finding application in automotive and truck engines. Poppet valves and valve seats, for example, require resistance to surface damage from repetitive impacts. Grinding and finishing processes can leave microcracks or residual stresses in the surfaces of ceramics which can accelerate surface-initiated degradation processes. A ball-on-inclined plane, repetitive impact testing system has been developed to study the effects of finishing method on surface durability. Tests were performed by repeatedly striking ground alumina and silicon nitride flat specimens with a silicon nitride sphere and measuring the impact damage as a function of the number of strikes. One type of alumina and three types of silicon nitride ceramics were tested. A quantitative impact damage parameter (IDP), based on mechanical stylus profiling data, was developed for use in measuring small amounts of wear. When damage did not extend beyond the depth of valleys in the machined surfaces, the IDP exhibited an excellent linear correlation with the number of impacts of silicon nitride on both silicon nitride and alumina materials. When more extensive damage occurred, a method involving digitizing the area of the contact damage from photomicrographs provided a reasonable correlation with number of impacts. These two methods are compared. Scanning electron microscopy revealed features which can affect the accuracy of impact damage measurement. Implications of the technique are discussed. 相似文献

This paper studies the effect of surface modification of multiwall carbon nanotubes (MWCNTs) prior to dispersion in engine oil to improve the tribological properties. The MWCNTs are stabilised in the lubricant with two different surfactants cetrimonium bromide (CTAB) and sorbitan monooleate (SPAN 80) and the effect of surfactants on the tribological properties has been studied. Pristine and surface modified MWCNTs in weight per cent range of 0·5% are dispersed in CI4 plus diesel engine oil. The foaming tendency and other physico-chemical properties of test lubricant have been studied to investigate the effect of nano materials and surfactants. The anti-wear and anti-friction properties are tested on a four ball wear tester and the comparison is made to assess the relative performance of pristine MWCNTs over surface modified MWCNTs. A strong influence of the surface modification technique is found on lubricating and physico-chemical properties. Both CTAB and SPAN 80 could keep the MWCNTs stable in the lubricant without compromising the foaming tendency of lubricant and other physico-chemical properties. The friction and wear characteristics of lubricants have improved with the dispersion of surface modified MWCNTs while there is no improvement in the properties of lubricant dispersed with pristine MWCNTs. 相似文献

There is an urgent need to better understand the mechanisms involved in scar formation in the brain. It is well known that astrocytes are critically engaged in this process. Here, we analyze incipient scar formation one week after a discrete ischemic insult to the cerebral cortex. We show that the infarct border zone is characterized by pronounced changes in the organization and subcellular localization of the major astrocytic protein AQP4. Specifically, there is a loss of AQP4 from astrocytic endfoot membranes that anchor astrocytes to pericapillary basal laminae and a disassembly of the supramolecular AQP4 complexes that normally abound in these membranes. This disassembly may be mechanistically coupled to a downregulation of the newly discovered AQP4 isoform AQP4ex. AQP4 has adhesive properties and is assumed to facilitate astrocyte mobility by permitting rapid volume changes at the leading edges of migrating astrocytes. Corneal transparency relies on the precise arrangement and orientation of collagen fibrils, made of mostly Type I and V collagen fibrils and proteoglycans (PGs). PGs are essential for correct collagen fibrillogenesis and maintaining corneal homeostasis. We investigated the spatial and temporal distribution of glycosaminoglycans (GAGs) and PGs after a chemical injury. The chemical composition of chondroitin sulfate (CS)/dermatan sulfate (DS) and heparan sulfate (HS) were characterized in mouse corneas 5 and 14 days after alkali burn (AB), and compared to uninjured corneas. The expression profile and corneal distribution of CS/DSPGs and keratan sulfate (KS) PGs were also analyzed. We found a significant overall increase in CS after AB, with an increase in sulfated forms of CS and a decrease in lesser sulfated forms of CS. Expression of the CSPGs biglycan and versican was increased after AB, while decorin expression was decreased. We also found an increase in KS expression 14 days after AB, with an increase in lumican and mimecan expression, and a decrease in keratocan expression. No significant changes in HS composition were noted after AB. Taken together, our study reveals significant changes in the composition of the extracellular matrix following a corneal chemical injury. 相似文献
