Flavivirus genus includes many deadly viruses such as the Japanese encephalitis virus (JEV) and Zika virus (ZIKV). The 5′ terminal regions (TR) of flaviviruses interact with human proteins and such interactions are critical for viral replication. One of the human proteins identified to interact with the 5′ TR of JEV is the DEAD-box helicase, DDX3X. In this study, we in vitro transcribed the 5′ TR of JEV and demonstrated its direct interaction with recombinant DDX3X (Kd of 1.66 ± 0.21 µM) using microscale thermophoresis (MST). Due to the proposed structural similarities of 5′ and 3′ TRs of flaviviruses, we investigated if the ZIKV 5′ TR could also interact with human DDX3X. Our MST studies suggested that DDX3X recognizes ZIKV 5′ TR with a Kd of 7.05 ± 0.75 µM. Next, we performed helicase assays that suggested that the binding of DDX3X leads to the unwinding of JEV and ZIKV 5′ TRs. Overall, our data indicate, for the first time, that DDX3X can directly bind and unwind in vitro transcribed flaviviral TRs. In summary, our work indicates that DDX3X could be further explored as a therapeutic target to inhibit Flaviviral replication  相似文献

采用实验及数值计算研究了乙醇和二甲醚微圆管射流火焰燃烧特性。通过实验观察到不同燃料流速下乙醇和二甲醚火焰都具有四种典型的火焰形态;使用平面激光诱导荧光测试系统获得了微射流火焰的OH基元分布,实验结果表明在较高流速下稳定燃烧的乙醇火焰比二甲醚火焰直径小,且略高于二甲醚火焰;采用考虑详细化学反应机理的数值计算对乙醇和二甲醚火焰进行了数值模拟,计算结果与实验现象吻合较好;利用一维非预混对冲火焰计算进一步研究了这两种燃料的化学反应路径,分析结果表明乙醇和二甲醚火焰的中间产物有显著差异,两种燃料化学反应特性的差异导致了不同的微火焰结构。  相似文献

Nano-structured arrays are engineered to meet the requirements of a variety of applications such as microfilters, sensors, and structural interface due to their unique mechanical characteristics, which cannot be achieved by conventional solid materials. However, it is hard to evaluate the elastic properties of nano-structured arrays owing to the discrete structure, sample size, and availability of suitable techniques. To facilitate this, we develop an advanced three-dimensional microscale vibration testing process. In the test, a specially designed three-dimensional microspecimen with tuned mass is excited by a piezoelectric actuator, and the resonance frequencies are detected by a laser device successfully. The anisotropic elastic moduli of nano-structured array composed of helical nano-springs are identified from a single spectrum. This array shows so strong characteristic anisotropy that the solid one hardly can attain. The microscale testing technique can be extended to other materials and microstructures.  相似文献

Five material properties commonly used to describe the fire behavior of solids were evaluated as sole explanatory variables for four small‐scale fire tests with pass/fail outcomes by using a physically based probabilistic (phlogistic) burning model. The phlogistic model describes the likelihood of passing vertical Bunsen burner tests and a regulatory heat release rate test reasonably well over a wide range of material properties, as deduced from the correlation coefficient and mean deviation of the predicted and measured values. Of the thermal, combustion, and fire properties examined, the best predictors of the likelihood of passing the fire tests of this study were the heat of combustion of the sample, the heat release capacity, and the heat release parameter. The relative merits and drawbacks of qualitative (threshold) and quantitative (probabilistic) approaches to predicting fire test results using thermal and combustion properties are discussed. Differential thermal analysis (DTA) and thermogravimetry (TG) methods have been developed to evaluate the thermal and thermooxidation stability of different lubricating oils. Measurements are carried out either by heating the oil sample at a constant rate (direct programme) or by keeping it at a fixed high temperature (isothermal programme). Oxygen at a constant flow rate is used for thermooxidation measurements. Tested samples included different viscosity grades: neutral base oils, unadditivated lubricating oil blends, and industrial oils. The mechanism of thermal decomposition for ZDDP and the estimation of its optimum concentration was also considered.  相似文献
