本文直接采用Thomsen导出的横向各向同性介质中的相速度、群角与相角、群速度与相速度的精确函数关系,提出了一种新的具有任意各向异性强度的横向各向同性介质中的初至波旅行时计算方法,它不同于Faria等人给出的采用群速度近似公式(Byun)的仅适应弱各向异性的初至波旅行时计算方法。该新算法在计算具有任意各向异性强度的横向各向同性介质中的初至波旅行时时,并不显著地增大计算时间。分析对比由新算法得到的初至波旅行时的等值线图和由交错网格有限差分波场模拟得到的波场快照表明,无论是对弱各向异性还是对强弱各向异性,新算法都具有较高的精度;随着各向异性强度的增大,新算法的计算结果与采用近似公式计算结果的差值也随之增大。  相似文献

 波场的等效性、观测数据的有限性和观测误差等因素导致地球物理反演本身具有严重的多解性,降低这种多解性的一种重要途径就是对反演过程进行约束。传统的初至波走时层析成像方法,通常是在外部对模型空间施加显式的约束和平滑处理,这样就会破坏模型参数与传播算子之间的相关性,难以达到有效利用先验信息的目的, 甚至会影响反演的稳定性。针对这一问题,本文将先验信息分为三类,在此基础上阐述了通过正则化手段将各种先验信息纳入到反演方程当中,替代传统的外部约束模式。理论试验结果表明,正则化方法比外部约束方法有效地提高了反演的精度和分辨率;将该正则化方法应用于实际二维资料处理,也得到了比较好的反演结果。  相似文献

In cancer therapy, the application of (fractionated) harsh radiation treatment is state of the art for many types of tumors. However, ionizing radiation is a “double-edged sword”—it can kill the tumor but can also promote the selection of radioresistant tumor cell clones or even initiate carcinogenesis in the normal irradiated tissue. Individualized radiotherapy would reduce these risks and boost the treatment, but its development requires a deep understanding of DNA damage and repair processes and the corresponding control mechanisms. DNA double strand breaks (DSBs) and their repair play a critical role in the cellular response to radiation. In previous years, it has become apparent that, beyond genetic and epigenetic determinants, the structural aspects of damaged chromatin (i.e., not only of DSBs themselves but also of the whole damage-surrounding chromatin domains) form another layer of complex DSB regulation. In the present article, we summarize the application of super-resolution single molecule localization microscopy (SMLM) for investigations of these structural aspects with emphasis on the relationship between the nano-architecture of radiation-induced repair foci (IRIFs), represented here by γH2AX foci, and their chromatin environment. Using irradiated HeLa cell cultures as an example, we show repair-dependent rearrangements of damaged chromatin and analyze the architecture of γH2AX repair clusters according to topological similarities. Although HeLa cells are known to have highly aberrant genomes, the topological similarity of γH2AX was high, indicating a functional, presumptively genome type-independent relevance of structural aspects in DSB repair. Remarkably, nano-scaled chromatin rearrangements during repair depended both on the chromatin domain type and the treatment. Based on these results, we demonstrate how the nano-architecture and topology of IRIFs and chromatin can be determined, point to the methodological relevance of SMLM, and discuss the consequences of the observed phenomena for the DSB repair network regulation or, for instance, radiation treatment outcomes.  相似文献

Abstract. Since the seminal paper by Dickey and Fuller in 1979, unit‐root tests have conditioned the standard approaches to analysing time series with strong serial dependence in mean behaviour, the focus being placed on the detection of eventual unit roots in an autoregressive model fitted to the series. In this paper, we propose a completely different method to test for the type of long‐wave patterns observed not only in unit‐root time series but also in series following more complex data‐generating mechanisms. To this end, our testing device analyses the unit‐root persistence exhibited by the data while imposing very few constraints on the generating mechanism. We call our device the range unit‐root (RUR) test since it is constructed from the running ranges of the series from which we derive its limit distribution. These nonparametric statistics endow the test with a number of desirable properties, the invariance to monotonic transformations of the series and the robustness to the presence of important parameter shifts. Moreover, the RUR test outperforms the power of standard unit‐root tests on near‐unit‐root stationary time series; it is invariant with respect to the innovations distribution and asymptotically immune to noise. An extension of the RUR test, called the forward–backward range unit‐root (FB‐RUR) improves the check in the presence of additive outliers. Finally, we illustrate the performances of both range tests and their discrepancies with the Dickey–Fuller unit‐root test on exchange rate series.  相似文献

碱性单细胞凝胶电泳预测肿瘤细胞内在放射敏感性研究   总被引:3,自引:0,他引:3  
应用克隆形成法和碱性单细胞凝胶电泳技术检测辐射诱导的人红白血病细胞株K562、人结肠腺癌细胞株LS-T-117和鼠胶质瘤细胞株C6的初始DNA单链断裂数及单链断裂后的修复与细胞内在放射敏感性之间的关系。结果表明,3种细胞系的放射敏感性依次为K562>LS-T-117>C6;3种细胞系的DNA迁移距离都随着照射剂量的增加而增大,呈良好的剂量-效应关系。在相同剂量下,辐射诱导的DNA单链的初始断裂数目也依次为K562>LS-T-117>C6;3种细胞系经10Gy X射线照射并在PBS中培养不同时间后DNA迁移距离都有较大幅度的下降,但下降幅度依次为C6>LS-T-117>K562,在相同剂量下辐射诱导的DNA单链断裂后的修复能力也依次为C6>LS-T-117> K562。结果显示,辐射诱导的DNA单链断裂及修复与细胞内在放射敏感性有很好的相关性,可用于人体肿瘤细胞内在放射敏感性的预测。  相似文献

Understanding and quantifying the learning–forgetting process helps predict the performance of an individual (or a group of individuals), estimate labor costs, bid on new and repeated orders, estimate costs of strikes, schedule production, develop training programs, set time standards, and improve work methods [IIE Trans. 29 (1997) 759]. Although there is agreement that the form of the learning curve is as presented by [J. Aeronaut. Sci. 3 (1936) 122], scientists and practitioners have not yet developed a full understanding of the behavior and factors affecting the forgetting process. The paucity of research on forgetting curves has been attributed to the practical difficulties involved in obtaining data concerning the level of forgetting as a function of time [IIE Transactions 21 (1989) 376]. The learn–forget curve model (LFCM) was shown to have many advantages over other theoretical models that capture the learning–forgetting relationship. However, the deficiency of the LFCM is in the assumption that the time for total forgetting is invariant of the experience gained prior to interruption. This paper attempts to correct this deficiency by incorporating the findings of [Int. J. Ind. Ergon. 10 (1992) 217] into the LFCM. Numerical examples are used to illustrate the behavior of the modified LFCM (MLFCM) and compare results to those of the LFCM.  相似文献

Confocal laser scanning microscopy (CLSM) offers improved depth discrimination and spatial resolution to the analysis of biologic samples. We demonstrate in this paper that such technology is valuable in examining DNA single-strand breaks in human cells. The single-cell-gel (SCG) assay is a new technique for measuring DNA strand breaks in individual cells. Cells embedded in lowmelting-point agarose are treated with varying concentrations of hydrogen peroxide to induce DNA strand breaks. Following cell lysis and alkaline electrophoresis, which enables single-stranded break detection, analysis of the resulting “comets” provides an accurate method of comparing changes in DNA migration patterns, which have been shown to reflect the DNA damage levels. A statistically significant difference (p < 0.01) in single-stranded DNA damage levels was detected in cells exposed to hydrogen peroxide concentrations as low as 10 nm for 2 min. LSM analysis of the SCG technique allows rapid, sensitive and reproducible quantitation of single-stranded breaks of cellular DNA.  相似文献
