RGD修饰钛表面对人牙龈成纤维细胞初期黏附和铺展的影响

摘    要:用羰基二咪唑(1,1'-carbonyldiimidazole,CDI)将含RGD的短肽共价连接到纯钛表面,研究接枝后的钛表面对原代培养的人牙龈成纤维细胞(human gingival fibroblasts,HGF)初期黏附和铺展的影响.结果表明,RGD修饰的纯钛表面粘附的细胞数比未修饰钛表面多,细胞铺展面积比钛表面的大,应力纤维的形成比钛表面早.RGD接枝钛表面更有利于人牙龈成纤维细胞的粘附,改善了纯钛的生物相容性.

Abstract:RGD-containing peptide was coupled on titanium surface of dental implants. It is confirmed that this peptide promotes human gingival fibroblasts (HGFs) adhesion during the early stage after implantation was tested. RGD peptides were attached covalently to the titanium surface by 1, 1'- carbonyldiimidazole (CDI). Primary HGFs were inoculated on commercially pure titanium and RGD-grafted titanium respectively. Cell attachment and proliferation was assessed by acridine orange staining technology. Cell spreading was checked by immunofluorescence staining and confocal laser scanning microscope. The number of attached and proliferated HGFs and their average spreading area on RGD modified titanium were higher than that on unmodified titanium. The actin cytoskeleton reorganization existed earlier on RGD coated samples than on uncoated ones. This research may develop a new potential method to enhance the integration between human gingival soft tissue and titanium implant surfaces.
