多柱串联离子交换分离L-苯丙氨酸的研究

摘    要:利用离子交换法结合多柱串联吸附、洗脱工艺从酶转化液中高效分离提取L-苯丙氨酸,研究了多柱串联吸附及洗脱过程的控制与洗脱尾流的利用。该法无吸附损失,树脂对L-苯丙氨酸的吸附量达135mg/g树脂。通过洗脱出口pH变化反映L-苯丙氨酸洗脱程度,收集pH3·3~10·7范围内的L-苯丙氨酸洗脱液。采用洗脱尾流串联洗脱除去杂质,并回收尾流中的L-苯丙氨酸,洗脱收集液的L-苯丙氨酸浓度达32g/L,收率97%。

Abstract:High efficiency separation of L-phenylalanine by ion exchange with tandem columns was studied, such as contol of adsorption and desorption and utilization of elution wake flow. There is no loss in tandem adsorption, and adsorbance of L-phenylalanine is 135 mg/g resin. Absorption control is ascertained by pH of extract, the range of collection for L-phe in extract is from pH 3.3 to pH 10.7. Impurities are eluted by elution wake flow from tandem columns and L-phenylalanine in wake flow is recovered. The concentration of elution is above 32g/L, 97% of L-Phenylalanine is obtained by desorption.
