电离辐射和紫杉醇诱导的KB细胞中SPATA5L1和Cyclin B2表达的变化

摘    要:电离辐射与紫杉醇协同作用,可以诱导体外培养的人口腔上皮癌KB细胞出现G2/M阻滞,形成多核细胞,并最终造成死亡。先前通过基因芯片筛选研究发现,在经过电离辐射和/或紫杉醇作用后,有10条与细胞分裂有关的基因存在差异表达。本研究旨在通过Real—time PCR和Western Blot定量分析其中两条基因SPATA5L1和CyclinB2的表达变化,来揭示G2/M阻滞及多核细胞形成的分子机制。结果表明,在受到6Gy以下不同剂量的电离辐射或电离辐射与40nmol/L紫杉醇协同作用后,SPATA5L1的表达量明显增加,但同时CyclinB2的表达量则下降。推测SPATA5L1上调表达和CyclinB2适度的下调表达,使受处理细胞在胞核分裂后阻止胞浆分裂而无法形成正常子代细胞,导致G2/M阻滞及多核细胞形成。
