摘    要:为了探索一株细菌JS17的生长特性,同时采用活菌计数法和分光光度法测定JS17的生长曲线,并对两种结果进行对比分析.活菌计数法能够真实可靠地反映体系中的活菌数量,但操作繁琐,要求高,耗时长;比浊法方便快捷,但测得的菌体浓度受死亡菌体及其它悬浮物的影响较大.从生长曲线得出JS17在松醇油液体培养基中摇床培养时,0~20 h为生长延滞期,20~48 h为指数生长期,48~72 h为生长稳定期,72 h之后为衰亡期.结合两种方法对细菌生长规律进行测定,就可以较为全面地反映细菌的生长繁殖以及悬浮物代谢变化的规律.

Abstract:The bacterium growth curve was determined via the two methods,surviving bacteria count and spectrophotometry,used to study the growth characteristics of a bacterium called JS17.The results obtained by means of the surviving bacteria count can reflect the actual number of live bacteria,but this method needs many operations and it is time-consuming.It is more convenient to use the spectrophotometry method,but the results got by means of this method were influenced by other suspended matter,which were possibly the chemical reaction materials or metabolism products of JS17.The growth curve of JS17,with shaking culture in 2# oil medium,shows that lag phase in 0~20 h,log phase in 20~48 h,stationary phase in 48~72 h and decline phase emerge after 72 h.The results suggest that the suspended matter affects the rule of growth curve.The metabolism of the suspended matter of bacterium JS17 deserves further study.
