超声波辅助喷雾加湿法富集发芽黑糙米生物活性物质工艺的响应面优化

摘    要:本研究选用黑糙米作为试验原料,结合超声波和喷雾加湿法对黑糙米进行预处理,以黑糙米发芽后制得的发芽糙米γ-氨基丁酸(γ-aminobutyric acid,GABA)含量和多酚含量为测定指标,在单因素实验的基础上设计三因素三水平的响应面优化试验,并对数据进行拟合和相关性分析,确定超声波辅助喷雾加湿法富集发芽黑糙米生物活性物质的最佳工艺参数。结果表明,发芽黑糙米生物活性物质的最佳工艺参数为:超声功率144 W、超声温度40℃、超声时间45 min、单次循环喷雾加湿量10 mL、间隔时间5 min。在此条件下,发芽黑糙米GABA含量为83.71 mg/100 g,发芽黑糙米多酚含量为419.55 mg/100 g。综上,说明该响应面模型准确度较高,所得到的优化工艺条件具有一定的可行性,可为发芽黑糙米生物活性物质富集的研究提供参考,具有广阔的应用前景。

Abstract:In this study,the ultrasound and spray humidification treatment were applied to pretreat the black brown rice,γ-aminobutyric acid( GABA) contents and polyphenols contents in germinated brown rice from black brown rice were used as evaluation index,an response surface methodology involving three independent variables at three different levels based on single factor experiments were used to determine optimal the processing technological parameter of ultrasonic-assisted spray humidification for enriching bioactive substances in germinated black brown rice. Results showed that,the optimum conditions were determined as follows: Ultrasonic power 144 W,ultrasonic temperature 40 ℃,ultrasonic time 45 min,the amount of single-pass cycle spray humidification 10 m L,interval time 5 min. Under these conditions,γ-aminobutyric acid( GABA) contents from germinated black brown rice was 83.71 mg/100 g,the yield of polyphenols from germinated black brown rice was419.55 mg/100 g. In conclusion,the response surface model in this study had higher accuracy.The optimized process conditions obtained have certain feasibility,which an provide reference for enriching bioactive substances in germinated black brown rice,and have broad application prospects.
