银耳多糖分离提取工艺的优化研究

摘    要:银耳多糖是银耳中的主要生物活性物质之一,可由银耳子实体、孢子及发酵液中分离、纯化后获得。大量研究表明,银耳多糖具有免疫调节、抗肿瘤、抗衰老、降血脂和降血糖等多种生理活性。采用超声辅助热水浸提法来提取银耳多糖,通过单因素及正交试验优化料液比、浸提温度、浸提时间和浸提液pH值,得出银耳多糖的提取工艺条件为:料液比1∶50(g/mL),浸提温度80℃,浸提时间4.5 h,pH 10,银耳多糖得率最高,为(16.25±0.37)%。方差分析结果表明料液比对银耳多糖得率具有显著性影响(p<0.10)。

Abstract:Tremella fuciformis polysaccharides were one of the main bioactive substances which could be isolated and purified from Tremella fuciformis fruiting bodies,spores and fermentation broth. Many studies in modern medicine and pharmacology have shown that Tremella fuciformis polysaccharides have many physiological functions,such as improving immunity,anti-cancer,anti-aging,reducing blood sugar and blood lipid,etc. Ultrasonic-assisted hot water extraction method was used to extract polysaccharides from Tremella fuciformis. The ratio of material to liquid,extraction temperature,extraction time and extraction pH were optimized by single factor and orthogonal experiments. The optimum extraction conditions were as follows:ratio of material to liquid was 1 ∶ 50(g/mL),extraction temperature was 80 ℃,extraction time was 4.5 h and extraction pH was 10. Under the optimum conditions,the yield of Tremella fuciformis polysaccharides could reach(16.25±0.37)%. Moreover,the ratio of material to liquid had the greatest influence on the yield of Tremella fuciformis polysaccharides(p<0.10).
